New markers of colon cancer evolution in ulcerative colitis

Background: Long-standing ulcerative colitis (UC) has an increased risk of evolving into colorectal cancer (CRC) and upregulated expression of cyclooxygenase 2 (COX-2), inducible nitric oxide synthase (iNOS), survivin, telomerase catalytic subunit (TERT), integrin-linked kinase (ILK) and transcription factors c- MYB and TCF-4, has been implicated in the development and progression of sporadic colorectal cancer. Nevertheless much less is known about their role in the process of UC-associated colon carcinogenesis. Methods: We analyzed the gene expression of these markers during the transition of colonic mucosa from chronic inflammation to epithelial neoplasia in biopsies of UC patients using quantitative real-time polymerase chain reaction and immunohistochemistry, and compared the expression profiles of this gene panel in samples of patients with CRC and in human tumor xenografts of SW620 malignant colonic cells. Additionally, we determined the expression of these genes in mouse models of sporadic and colitis-associated CRC in A/J and ICR mouse strains using quantitative RT-PCR and laser microdissection. Influence of chemical agents on stability of PTC ceramics based on BaTiO3

The thesis deals with the study of the influences of selected groups of chemical agents on the stability of PTC ceramics based on BaTiO3. The experimental part includes elaboration of methodology for determination of these influences, their evaluation, including the hypotheses for their description, and evaluation of groups of chemical agents having these influences.
Personal Voice Activity Detection

Cílem této práce je implementovat, otestovat a vyhodnotit řečníkem podmíněnou metodu pro detekci hlasu ( Voice Activity Detection , VAD) nazvanou Personal VAD. Pro detekci využívá tato metoda LSTM neuronových sítí a jejím účelem je vytvoření systému schopného spolehlivě detekovat řečové signály cílového řečníka při zachování vlastností typického VAD systému co se velikosti modelu, odezvy a nízkých nároků na zdroje týče. Systém je trénován pro klasifikaci řečových rámců do tří tříd: neřeč, řeč necílového a řeč cílového řečníka. Za tímto účelem využívá metoda speaker embedding vektory pro reprezentaci cílového řečníka jako součást vstupních příznaků. Některé z náročnějších variant systému využívají skórování rámců systémem pro verifikaci řečníka, což vede ke zvýšení spolehlivosti klasifikace. Vedle základní metody skórování představené v originálním článku byly navrženy dvě modifikace, jež základní metodu překonaly a zlepšily spolehlivost výsledného systému i v akusticky náročných prostředích.
The Effect of Bulkiness of Non-peripheral Substituents of Phthalocyanines on their Absorption and Photophysical properties

PharmDr. Veronika Nováková Ph.D. Title of thesis The Effect of Bulkiness of Non-peripheral Substituents of Phthalocyanines on their Absorption and Photophysical Properties Phthalocyanines are planar macrocyclic compounds which absorb the light over 670 nm due to their large system of conjugated double bounds. Peripheral substitution of phthalocyanines significantly affects the position of the main absorption band. Absorption at longer wavelengths is highly advantageous in any biological applications because such light does not interfere with endogenous chromophores. It was discovered that the desired red-shift of absorption band is much more pronounced at non-peripherally substituted phthalocyanines. However, it was recently found out that the presence of bulky non-peripheral substituent shifts significantly absorption maximum back to lower wavelengths. The legal nature of the curriculum documents of the Czech Republic's educational system ABSTRACT This thesis treats the selected set of curriculum documents within the Czech legal framework laid down by the Act No. 561/2004 Collection of Law, on Pre-school, Basic, Secondary, Tertiary Professional and Other Education (the Education Act), as amended. The focus aims towards the Framework Educational Programmes (FEP) and School Educational Programmes (SEP), for which the relevant FEPs were issued. The National Educational Programme according to the Education Act as amended effective until 30. September 2020 is also taken into consideration on a theoretical level. In the times of expecting considerable state-level curriculum documents contents changes, and consequent changes on the school level, this thesis aims to analyse substantial legal aspects of each type of curriculum documents and their mutual effects and to assess, whether the current legal framework reflects their presumed use and meaning appropriately. With that goal, the key notions are defined first though and a brief insight into the curriculum historical development is provided. Impact of increased temporal detail in long-term dynamic energy system model with an increasing share of volatile renewable energy sources

Long-term energy systems often simplify temporal detail resolution. However, this simplification is not appropriate for systems with an increasing share of intermittent renewable energy sources (IRES). The low temporal resolution causes a systemic bias in the model's outputs. The model overpredicts the share of the IRES on the total generation. This entails an underprediction of the total system costs, underprediction of the amount of greenhouse gas emissions and a bias in the energy mix. This could lead to an ill-informed energy policy and eventually cause a failure in the decarbonization of the energy sector. This thesis increases the temporal resolution of the TIMES-CZ model from 12 seasonal times slices and 3 daynite time slices to 24 daynite time slices and compares 8 scenarios with 1, 4 and 12 seasonal time slices. This thesis compares three representative days selection methods - simple heuristics, hierarchical clustering centroid selection method and historical day closest to the centroid selection method. The electricity generation of IRES and the total production in base year (2012) is the most accurately approximated by the 12 seasonal time slices selected by the hierarchical clustering centroid method.
Preparation of yttrium based chemically bounded phosphate ceramics

The thesis is focused first on the research of literature on the topic of phosphate ceramics and then on the search for a suitable synthesis of phosphate binder based on Y2O3-P2O5-H2O and characterization of the resulting products. Various types of syntheses were prepared with subsequent characterization of the phase and chemical composition of the final products. The samples were measured on an electron microscope to determine their surface morphology and also element maps were obtained. The samples were also analyzed by heat microscopy to determine the temperature behavior of the samples. After measurement on a heat microscope, the samples were measured again on an electron microscope, in order to detect changes caused by the action of high temperatures, in particular to assess the presence of binder or sintered phases.
Three-dimensional numerical analysis of Czech vowel production

Spatial air pressures generated in human vocal tract by vibrating vocal folds present sound sources of vowel production. This paper simulates phonation phenomena by using fluid-structure-acoustic scheme in a three-dimensional (3D) finite element model of a Czech vowel [o:]. The computational model was composed of four-layered M5-shaped vocal folds together with an idealized trachea and vocal tract. Spatial fluid flow in the trachea and in the vocal tract was obtained by unsteady viscous compressible Navier-Stokes equations. The oscillating vocal folds were modelled by a momentum equation. Large deformations were allowed. Transient analysis was performed based on separate structure and fluid solvers, which were exchanging loads acting on the vocal folds boundaries in each time iteration. The deformation of the fluid mesh during the vocal fold oscillation was realized by the arbitrary Lagrangian-Eulerian approach and by interpolation of fluid results on the deformed fluid mesh. Preliminary results show vibration characteristics of the vocal folds, which correspond to those obtained from human phonation at higher pitch. The vocal folds were self-oscillating at a reasonable frequency of 180 Hz. The vocal tract eigenfrequencies were in the ranges of the formant frequencies of Czech vowel [o:] measured on humans, during self-oscillations the formants shifted to lower frequencies.

This paper describes some current methods for customer's opinion mining and text processing. Application architecture is created along with database structure. Based on this architecture, new application is implemented. Emphasis is placed on automated downloading and processing of customers' reviews with opinion extraction and user-friendly presentation. Part of this paper is dedicated to optimizing the application for better efficiency and speed boost.
Recognition of Multi-Talker Overlapping Speech Using Neural Networks

Tato práce se zabývá rozpoznáváním řeči překrývajících se řečníků pomocí neuronové sítě. Zkoumá  problém rozpoznávání řečí od vícero řečníků a způsoby, jimiž se tento daný problém řeší. Jedná se konkrétně o aplikaci kromě tradičních komponentů jako konvoluční neuronové sítě, LSTM atd. také speciálních komponentů: attention mechanismus a gated konvoluce. A dále také aplikace techniky zvanou permutation invariant training. Součástí této práce je aplikování těchto přístupů na přidělená trénovací data, která jsou tvořena uměle vytvořenými směsmi dvou řečníků předčítající články z Wall Street Journal. Dalším krokem bylo natrénování příslušných architektur používající kombinující prvky zmíněné nahoře. Modely v této práci nahrazují akustický model. Jednalo se o dvě architektury užívající různé typy attention mechanismu a o jednu bez něj.  Experimenty ukázaly, že architektury užívající attention mechanismus v tomto typu úlohy něpřekonaly tradičnější architekturu s užitím gated konvolucí. Přesto ale ukázaly potenciál.
