The regulation of gene expression is achieved at many levels. Chromatin-based gene regulation has been the central focus of many decades of research; however, posttranscriptional control mechanisms are emerging as a fundamental complement to direct protein synthesis. This thesis is focused on a specific mechanism of posttranscriptional control - the translational regulation of mRNAs in the cell cytoplasm. This control is a consequence of the balance between translational repression and activation and hinges on the selective recognition of regulated mRNAs by RNA-binding proteins and their ability to recruit RNA modifying proteins. In this thesis, Caenorhabditis elegans germline was used to study translational control of the germ cell-enriched gene, gld-2. Mutants of known RNA-binding proteins of the PUF and CPB protein families were analyzed by performing Western blots, using anti-GLD-2 antibodies. Yeast 3-Hybrid system was used to identify the cis-regulatory sites in the gld-2 mRNA conferring translational regulation by members of PUF and CPB protein families. Potential autoregulatory loop of gld-2 gene expression was also investigated. The Quaternary Paleoecological Analysis of the Soos National Nature Reserve The aim of this proposed diploma thesis is to describe the structure of diatom communities in the diatomite deposits of the Soos National Nature Reserve and compare the finding of this thesis with those of older publications. Another aim was to reconstruct the history of the basin trough with the outputs of diatom analysis accomplished in the 3,4 m long sediment profile. Care of The Soul and Problem of Education

Care of the Soul and Problem of Education The Czech philosopher Jan Patočka shows two different modalities of soul concepts while thinking about Jan Amos Komenský's heritage for contemporary pedagogy. The close-set soul conceptus supposes simply recognizeable world, modern ideas of its controllability and scientific structure scheme. Education, which means care of the soul in three ways - care of the soul in the sense of cosmos order, in the sense of public and in the personal way, means (following close-set conceptus) formation, preparation and setting up the undone young being for life in the modern society. Reflexions of our contemporary situation find out, that modern situation appears as a crisis. This crisis demands a new approach to education, which could be found in the open-set soul concept. The open-set soul could be described as something incomplete, which is searching its fundamentals all the time. Another characteristic shows affinity of the soul for something superior or sense of own finality. Education in this way unveils itself like repeatedly reached humanity. This kind of education does not mean formation or setting up, this kind of education means revealing context of the world, society and finally own personality. Open-set mind should bring responsibility through this revealing....
Visualization of the Chemical Space

Exploration of chemical space is an important part of the drug discovery pro- cess and its importance grows with computation power which permits to explore greater parts of the chemical space. One of the possibilities of the exploration is vi- sualization. Currently, several method for visualization are implemented but there exists no framework focusing primarily on support of the development of new me- thods. In this work we summarized requirements for such framework and with respect to these requirement we implemented such framework. The framework is modular and offers easy implementation of every single part of the visualization pipeline. The framework incorporates an application that provides graphical in- terface for modules' manipulation and presentation of visualization results. For simple utilization of the application without the necessity of implementation of one's own module, several visualization methods have been implemented. Spatio-temporal aspects of emerging "edge cities" in Prague metropolitan region

The phenomenon edge city has its roots in U.S. metropolitan areas. Similar urban formations currently also appear in other developed cities. Transformations in post-socialist cities starting in early 90th caused the emergence of new socio-spatial formations in suburban area of Prague metropolitan area. The case study of commercial zone Čestlice-Průhonice shows the newly created cluster of advanced services in Prague suburban area resulting in high concentration of daily population and vehicles. The main aim of this study is to uncover the time dimension of outer-spatial relations of the commercial zone by analyzing the daily rhythm of transportation. The method of direct observation of passing vehicles and its registration numbers is used. Furthermore, it has been proved that the commercial area has a regional significance. In this thesis, new socio-spacial formations in suburban fringe of Prague are ilustrated as an important subject for further geographic research. Natural forest disturbances palaeoecology

This Bc. thesis deals with the disturbances, which form an integral part of many ecosystems. The main foci are natural disturbances (in contrast to anthropogenic ones) of temperate and boreal forests. I deal with the causes of major disturbancy factors (fires, storms, disease and insect pathogens) and their impact on the ecosystem. I focused in detail on fires, because they represent an essential factor in the development of many ecosystems. Moreover, they can be studied in the past by using several tools of palaeoecological analysis: dendroecology, anthracology and pollen analysis combined with micro-charcoal particles quantification. The last part of the thesis discusses the role of disturbances in the secular succession of Holocene vegetation in Central Europe. It contains a chapter that describes a research strategy focused to the study of natural disturbances through paleoecological methods in a specific location (Rynholec, Central Bohemia). Searching relevant articles in extensive collections

Searching text in articles is usually implemented with fulltext search. Using more advanced techniques however, it is possible to achieve significantly better results. The subject of this work is to create a universal library for searching extensible collections, specialized in czech language. The library makes use of tools capable of working with morphology while considering importance of words. It also conducts an experiment with word pairs, which adds context into the search process. The success rate of this experiment is tried on an extensible collection of data. Created library is a unique tool for processing extensible collections of czech text, while at the same time it is ready for further extension by new languages and methods.
Dissecting the nuclear function of the interleukin-1alpha

Interleukin-1alpha (IL-1alpha) is a well-known proinflammatory mediator acting as a secreted molecule. However, in addition to its ability to activate its membrane-bound receptor, there is growing evidence on its noncanonical nuclear function, which classifies IL-1alpha as a "dual function cytokine". This nuclear action depends on the evolutionary conserved N-terminal domain of IL-1alpha. After proteolytic processing, the N-terminal domain of IL-1alpha translocates into nucleus. Histone acetyltransferase (HAT) complexes were previously identified as nuclear targets of IL-1alpha precursor. However, the specific protein which is responsible for the interaction between IL-1alpha and HAT complexes has not been identified yet. To dissect this interaction, the N-terminal domain of IL-1alpha was produced in yeast. Suitability of this experimental setup for testing the interaction between IL-1alpha and eukaryotic HAT complexes was evaluated in this study. IL-1alpha has been analyzed in this study using bioinformatics approaches as well. Putative amphipatic acidic helixes of IL-1alpha have been characterized. Further Determinants of Private Equity

This thesis examines determinants of two variables important in evaluating private equity investments. First of them is the price of companies acquired by private equity funds expressed as a multiple of a profit indicator and the impact of primary and secondary buyouts. The second variable researched is the internal rate of return, a popular tool to measure profitability of private equity funds. The internal rate of return is studied for different groups of private equity funds and is reflected in a post-crisis perspective. The first conclusion stemming from the research is that the profit multiples in secondary buyouts are estimated to be higher compared to primary buyouts. The second conclusion proves the underperformance of real estate funds and the outperformance of distressed debt, turnaround and secondaries funds.
