I. SUMMARY Background: Iron is an essential element necessary for many physiological processes involving oxygen transport, DNA-synthesis and ATP-formation. The fate of iron in the organism is tightly regulated especially at the absorption and distribution level probably mainly due to lack of specific active iron excretion mechanism. Any derangement of iron homeostatis may lead to appearance of free (unbound or loosely bound) iron, which can catalyse reactive oxygen species (ROS) production by Haber-Weiss chemistry. Cardiovascular diseases, particularly coronary heart disease (CHD), remain notwithstanding recent scientific advances important therapeutic problem. The most serious form of CHD represents acute myocardial infarction (AMI). The pathophysiology of AMI involves in most cases initial ischaemic period caused by coronary blood flow derangement due to a thrombus formation. Ischaemia alters substantially tissue homeostasis with subsequent cytosolic free iron appearance. Reconstitution of coronary blood flow (reperfusion) represents the only way for myocardial tissue recovery although on the other hand, it is linked with a release of free redox-active iron in the circulation and formation of ROS both intracellularly as well extracellularly. 1. SUMMARY IN CZECH (ABSTRACT) Reaktivní formy kyslíku a dusíku (RONS) a volné radikály se staly v posledním desetiletí významným předmětem studia v mnoha oborech včetně medicínských. Stav, při němž je rovnováha mezi vznikem a odbouráváním volných radikálů v organismu posunuta ve prospěch jejich vzniku, se nazývá oxidační stres. Při nadbytku volných radikálů v organismu dochází k poškození biomolekul a tkání, což může vést až ke vzniku onemocnění. Oxidační stres se podílí na vzniku celé řady onemocnění, např. diabetu mellitu, aterosklerózy, revmatoidní artritidy, Alzheimerovy choroby a dalších, ale také na fyziologickém procesu stárnutí. Ve snaze pozastavit průběh stárnutí a odvrátit vznik onemocnění se začala používat různá potravní aditiva s obsahem vitamínů a antioxidantů přírodního původu. Diabetes mellitus je onemocnění charakterizované hyperglykémií a je vždy provázeno oxidačním stresem. Glukóza a různé intracelulární cukry (fruktóza, ribóza, glyceraldehyd a další) se kovalentně váží na volné aminoskupiny proteinů a způsobují poškození jejich struktury. Preparation and evaluation of potential drugs inhibiting mitochondrial enzymes Summary in English Alzheimer's disease (AD) is the most common cause of senile dementia worldwide. Despite being subject to intensive research, the pathogenic mechanisms of AD are still not fully understood and consequently an effective treatment is yet to be developed. Although the aetiology of AD is still unknown, a build-up of amyloid-beta peptide (Aβ) is considered to play an important role in disease progression. The original amyloid cascade hypothesis proposed that insoluble extracellular plaques were responsible for the majority of Aβ toxicity. This hypothesis has since been refined, as recent data indicates that soluble intracellular oligomers are now responsible for the majority of Aβ induced toxic effects. The mitochondrial dysfunction also plays an important role in the pathophysiology of AD. Aβ was detected inside mitochondria and several mitochondrial proteins were found to interact directly with Aβ. Such interactions can affect a protein's function and cause damage to the mitochondria, which finally results in progression of AD. Tématem následujících stránek je analýza kultury romských osad. Přestože je sousloví "romské osady" poměrně hojně frekventované, v otázce jeho užívání panuje značný zmatek a je tedy již téměř nemožné zjistit a říci, co si pod ním ten který autor vlastně představuje. V této stati budu toto označení užívat jako termín a obsahem mé práce bude do značné míry právě snaha o jeho definici. Jako každý termín, je i pojem "romská osada" svou povahou arbitrární, jedná se o konvenční, nehodnotící nálepku. Důvodem, proč jsem jako jeden z centrálních pojmů zvolil zaužívané a významově přetížené sousloví je ten, že za prvé laický výraz "romská osada" má konotace blízké významu tohoto pojmu v přítomné stati (v tomto ohledu se tedy jedná jen o jakousi kultivaci slova běžné mluvy) a za druhé dosavadní pokusy o zavedení jiného označení pomocí neologizmu dosud vždy selhaly (ať se již jednalo o termín slovenské státní správy "obytná zoskupenia na nízkom sociokultúrnom stupni" či termín "kumulovaná romská sídliště" 1 z autorské dílny S. This thesis begins with mapping the existence of the European structural and investment funds and then goes on to examining the way these funds are to be implemented. In order to establish the Specific Objectives from the thematic ones given by the EU. Furthermore, it creates the definition of a Cultural Project which is an essential tool for achieving the Specific Objectives of the operational programs. It identifies a certain number of Specific Objectives of the programming period 2014 to 2020, which have a strong linkage to the field of culture and to the cultural projects. Criteria are defined in order to stratify the database of Specific Objectives. These criteria lead to the creation of a matrix that helps in grasping the data in a practical manner. In the conclusion a method for designing an application which works with the data is specified as is the user-interface of the said application. Finally, an appropriate positioning of the application on the internet is proposed so that it can be accessible to potential users. The thesis thus proves the fact that cultural projects have many opportunities to contribute to the programming period 2014 to 2020.
Actual trends in production and distribution of beer in the light of longterm trends in the Czech Republic's beer culture (on an example of Plzensky Prazdroj)

The purpose of this master thesis is to analyze actual and long-term trends in production and distribution of beer in the Czech area. The master thesis is divided into several parts. The first part presents basic information about the influence of brewing industry on the European and Czech economies; in the following part are described trends of distribution and production of beer in the past which influenced current state of Czech brewing industry. Last part of this thesis is concentrating on a ON premise research which concentrates on one of a current trends which is typical for the Czech beer market and therefore has a strong influence on a actual state of Czech brewing industry.
The concept of the system state tourism policy Czech Republic with a focus on cultural tourism

The focus of the thesis is an issue of system approach to formation and implementation policy of tourism in the Czech Republic with an emphasis on intersectoral cooperation in the field of cultural tourism. The work is divided into eight chapters, the first two are theoretical. The first chapter is devoted to the formulation and definition problem of the scope of the work. The second chapter deals with theoretical foundations and approaches to tourism policy issues both in general and in terms of the status of tourism in the national economy and in relation to the EU tourism policy. Attention is given to the objectives of cohesion policy, culture and tourism in the area of cultural tourism. The following two chapters deal with basic features, analysis and evaluation of policy concepts of tourism in the Czech Republic in the period after 1989. The core of the thesis is concentrated into chapter six, seven and eight. The assumption for the design of complex, dynamic tourism policy for the formation of functional institutional structure of the tourism sector management is to assess the fulfillment of the objectives of the different concepts of state policy. To identification the most important and recurrent problems of tourism policy are used the results of comparison of objectives and measures and practical experience of the author from the creation and evaluation of the conception documents. There is discussed in particular an evaluation framework and in this context there are suggested recommendations for its determination. In the field of cultural tourism is proposed a model of interdepartmental cooperation state-funded organizations CzechTourism and the National Heritage Institute. The proposal model is based on its own research, which is focused on the analysis and evaluation of assumptions monuments administered by the National Heritage Institute to participate in the creation of cultural tourism products. There are utilized data from the author primary survey and as well as secondary data from the National Heritage Institute. The model of cooperation is based on exemplification of the causal relationships between strategic objectives and processes within the strategies of both organizations by the model BSC (Balanced Scorecard) and from a proposal for their implementation using model 7 "S".
Art culture as a segment of the cultural life of nations

The principal aim of this thesis is to undertake a historical analysis of cinematography in Czech Republic and France, respectively. In more detail, this analysis will concentrate on financing that flows towards film-making in said countries, while at the same time focusing on the artistic aspect of movies that are created thanks to this financing. This paper tests two hypotheses, which will be either confirmed or rejected after the analysis. First concerns the impact of political regimes on funding available to national film-making, and what effect does this have on the quality of film. The second one assesses the film quality from the perspective of special art tendencies.
