Influence of hydroxycinnamic acids on methylglyoxal-induced protein glycation

Author: Hana Vejvalková Publishing place: Charles University in Prague, Faculty of Pharmacy in Hradec Králové Title: Influence of hydroxycinnamic acids on methylglyoxal-induced protein glycation Supervisor: PharmDr. Iva Boušová, Ph.D. The diploma thesis was aimed at potencial antiglycation effect of antioxidants of natural origin (hydroxycinnamic acids) on the modification of bovine serum albumine and asparate aminotransferase by methylglyoxal as a glycating agent. I monitored the effect of hydroxycinnamic acids on generation of glycation products using fluorescent, absorption, and electrophoretic methods. The influence of hydroxycinnamic acids on the glycation of model proteins was ambiguous. The only derivative with proven antiglycation effect on production of non-specific glycation products and pentosidin measured by fluorescence and spectrophotometry was p-coumaric acid in 100 μM concentration. On the other hand, chlorogenic acid 100 μM displayed strong proglycation effects in all methods used for detection of glycation products. The effect of all tested substances was concentration dependent. Results show that the more substituents given acid contained in its molecule, the stronger proglycation effect this compound exerted.

Title: The popularity of the sport among the general public in relations to its presentation on TV Objectives: The aim of this thesis is to determine the popularity of the sport among the general public and to compare it with its presentation on TV. The popularity and viewership of the sport is determined by questioning the public. To conduct interviews with experts on sports broadcasters from television, explaining views of sport news provided on public and commercial televisions. Final results were compared with the results of observation presentation of the sport in sport news in the thesis of Trnková (2014). The results allowed to propose recommendations which will be for use in marketing, sports and media organisations. The results will only imply a certain degree of scientific contribution. Methods: The market research of this thesis has consisted of quantitative and qualitative methods. An electronic questionnaire published on the server Vyplňto.cz, helped gather results from the public. Interviews were carried out in-depth including electronic questioning via e-mail correspondence. Presentation of sports in television broadcasting in the Czech Republic

Title: Presentation of sports in television broadcasting in the Czech Republic Objectives: The aim of this thesis is to measure and compare the airtime devoted to individual sports and sports representations on public and commercial television in the regular, prime-time sports news. The recommendations will be proposed to be used in sports-club marketing, based on the results of observations. Methods: This thesis uses, as a method of observation, the standard sport news shown at prime-time on the TV stations ČT1 and TV Nova. This is the type of indirect observation with the technical devices - USB TV tuner and PC. In terms of content, Berelson's quantitative content analysis will be used, which examines the content of media messages. The theoretical part of the thesis is based on an analysis of documents. Results: The results of observations illustratively show the different approaches between public and commercial television for the inclusion of sport in the broadcasting of regular sports news in prime-time. For both channels, the most popular sports in the sports news are football and ice hockey. In addition, there is a trend of popularity of summer sports, even in the winter time. Title: A FOUNDATION PLAN OF KARATE SPORT CLUB Aims: The aim is to explain how to establish a karate sport club in new terms of foundation of associations. Another goal is the application of the theoretical knowledges to a chosen karate club Red Dragon Team (RDT) and to design options how to improve its functioning. Methods: Based on expert literature and texts research, a study has been produced which describes basic terms and ways of analysis execution. The SWOT analysis of the current state of the inner and outer environment of the RDT sports clubwas used in the second section of this document. Results: The work provides comprehensive guide describing the particular steps for founding of a new association in practice. The work is proposing a solution based on the SWOT analysis to eliminate possible deficiencies and how to better use the potential of the club RDT. Nanopatterned alumina-based materials for electrochemical sensors and biosensors

The doctoral thesis is focused on basic research and development of nanostructured surfaces prepared using anodic alumina material. Various types of gold nanostructured surfaces and nanoporous aluminum membranes for electrochemical sensors and biosensors were prepared using the anodic oxidation method. Nanostructured surfaces were prepared by electrochemical anodization of aluminum material to form hexagonally arranged nanopores. Gold was then deposited into the nanoporous masks by electrochemical reduction from potassium dicyanoaurate solution using a pulse deposition method. The prepared nanostructured gold surfaces were electrochemically characterized by electrochemical impedance spectroscopy and voltammetry. Temperature stability and the effect of annealing on their electrochemical behavior at atmospheric pressure as well as in the vacuum were investigated. Then, gold nanostructures of various dimensions were prepared and the influence of their shape and dimensions on the electrochemical behavior was studied. Nanostructured surfaces were also modified with 11–mercaptoundecanoic acid, and the effect of this modification on the electrochemical results was studied. In the last part of the work, nanoporous aluminum membranes were prepared, and their permeability was studied.

Title of diploma thesis: Modification of Glutathione S-transferase by selected glycating agents Glycation is a non-enzymatic reaction, which leads to the binding of carbonyl compounds to free amino groups of amino acids, peptides, proteins or other biomolecules. This process is closely related to impaired protein function in diabetes mellitus, during aging, and in neurodegenerative diseases. The main purpose of this thesis was to monitor the impact of several glycating agents on the properties of the enzyme glutathione S-transeferase (GST, EC 2.5.1.18) which plays a key role in the detoxification of many exogenous as well as endogenous compounds. Methylglyoxal (0.5-2 mM), glyceraldehyde (0.5-10 mM), glucose (50 mM), and fructose (50 mM) were selected as glycating agents. MacPherson Suspension Kinematics Analysis

This bachelor thesis deals with the influence of the position of significant points of MacPherson strut on kinematic characteristics. The first part contains a description of selected suspension parameters, their impact on the driving properties of the car, and a description of MacPherson strut. Furthermore, based on provided data, is prepared a detailed analysis of the influence of kinematics points to individual characteristics. In conclusion, all findings are synoptically summarized so that they can serve as a manual for change of the kinematic characteristics.
Development of Novel Electrochemical Methods Using Various Membrane Materials for Monitoring of Selected Anticancer Drugs and Phytochelatins

Present Ph.D. Thesis is focused on the development of electrochemical methods for determination of anticancer drugs using various types of membranes for their preliminary separation. Furthermore, this Thesis reports the study of transport mechanisms of heavy metals in the presence of phytochelatins across biological membranes. Sodium anthraquinone-2-sulphonate (AQS) was used as a model compound for its similar structure with anthraquinone-based (AQ-based) anticancer drugs (doxo/daunorubicin) and also due to its better availability. All these compounds can be easily electrochemically oxidized and/or reduced. Redox behaviour of AQS was investigated by cyclic voltammetry (CV) and differential pulse voltammetry (DPV) in a cathodic region on mercury meniscus modified (m-AgSAE) and polished silver solid amalgam (p-AgSAE) electrodes, Obtained results were used for the development of a micro-volume voltammetric cell (MVVC). Its applicability for voltammetric determination of anticancer drugs was verified by using doxorubicin (DX) as a model substance. Use of Monovalent Copper for Sensitive Detection of Methyl Derivatives of Xanthine

This work is concerned with the electroanalysis of xanthine (Xan) and its methylated derivatives (1-, 3-, 7-, and 9-mXan) on a pencil graphite electrode in the presence of copper ions. The main idea of using copper consists in the in situ formation of the complex Cu(I)-mXan on the electrode surface and improvement of oxidative signals of the corresponding mXan during the detection. Linear sweep voltammetry (LSV) in connection with adsorption stripping techniques was used for the determination of mXan. To enhance the oxidation signals the elimination voltammetric procedure (EVP) was applied.
