Matoušková, Petra ; Skálová, Martina (referee) ; Milichovský, František (advisor)
The subject and scope of my thesis is proposal of a communication mix for the shop and e-shop, which mainly operates is the current state analysis of a company in order to discover its core problems, which could be remedied by otimizing the current or drawing up a new marketing communication mix. The theoretical part of the thesis deals with basic marketing terminology.
Re-interpretation of Learning Enviroment
Hamrník, Ondřej ; Matoušková, Petra (referee) ; Mléčka, Jan (advisor)
Transformation of educational space running in these days all over the world. Every one is conscious that our educational system is obsolete and we have to change it. Instead of designing of new school building o choose building which is running for education for almost one hundred and fifty years. One of these building is gymnasium in Hradec Kralove

Újlak - Psychiatric Hospital
Valeková, Ivana ; Matoušková, Petra (referee) ; Mléčka, Jan (advisor)
The aim was to design a new conception for both new building of psychiatric hospital and psychiatric cure on the other hand. Site is based in Velke Zaluzie, villacee in the southwestern Slovakia. Placed in site of historically protected park with a dominant building of the Old Esterhazy Mansion uses principles from moder psychiatry and space modularity to ideal inter-building connections. With high sustainability aspect a new organisation of the site was created on the whole area of the park with new functions for existing buildinge and less dominant based structure od psychiatric hospital.
Renewal of Old Brno District
Sopoušková, Petra ; Matoušková, Petra (referee) ; Mléčka, Jan (advisor)
The work deals with the development of the Old Brno District . Old Brno District is currently marked by waiting for the implementation of the land use plan, which should solve the problem of traffic. The implementation, however, will probably take a few decades. I propose to use the period of time before the change of traffic routes and begin a gradual development that will prefigure target form of the district. Well-designed architecture and urbanism is not the only condition for well-functioning city. Therefore, I propose a process which should lead to the target form, but to create social, economic, ecological and aesthetic values independently of it. For this purpose vacant spaces and temporary structures are used.
The efflux transport activity of P-glycoprotein - 9.2 fromHaemonchus contortus
Kohoutová, Kateřina ; Matoušková, Petra (advisor) ; Boušová, Iva (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Kateřina Kohoutová Supervisor: doc. Ing. Petra Matoušková, Ph.D. Title of diploma thesis: The efflux transport activity of P-glycoprotein-9.2 from Haemonchus contortus Haemonchosis is a globally widespread disease affecting small ruminants. The causative agent of the disease is the gastrointestinal nematode Haemonchus contortus, which feeds of the blood of its host. The treatment of haemonchosis with anthelmintics is complicated by the worldwide spread of resistance. One possible cause of the development of anthelmintic resistance is a decrease in anthelmintic concentration in the worm due to increased expression of P-glycoproteins from ABC transporter superfamily. The study of the transport activity of these transporters could help to understand the mechanism of resistance development and make the treatment of haemonchosis more effective. The aim of this thesis was to characterize the transport activity of P-glycoprotein-9.2 and its interaction with a selected inhibitor using a heterologous expression system. The adherent cell line HEK293 was transfected with a plasmid carrying the Pgp-9.2 gene of the Haemonchus contortus. Successful expression was verified by dot blot. Unfortunately,...
Constuction of plasmid and probes for detection of P-glycoprotein in Haemonchus contortus
Kaduchová, Valentýna ; Matoušková, Petra (advisor) ; Novotná, Eva (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Bc. Valentýna Funioková Supervisor: Doc. Ing. Petra Matoušková, Ph.D. Title of diploma thesis: Construction of plasmid and probes for detection of P-glycoprotein in Haemonchus contortus Anthelmintic resistence in parasitic nematodes has become an alarming issue worlwide. Haemonchus contortus, a pathogenic nematode of small ruminants, has become resistant to all types of anthelmintic drugs. Important players of the anthelmintic resistence are ABC transporters, most notably P-glycoproteins. In Haemonchus contortus, 10 homologues of Pgp have been identified. Research of individual P-glycoproteins could reveal particular transporters involved in anthelmintic resistence mechanisms. The diploma thesis focuses on P-glycoprotein-9.2, whose constitutive expression is higher in resistant strains of H. contortus than in susceptible strain. The opening chapters of the diploma thesis deals with the topic of parasitic nematode H. contortus, anthelmintic resistence and description of ABC transporters. The main part of this thesis is dedicated to construction of plasmids for in vitro preparation of digoxigenin-labelled RNA probes, and detection of P-glycoprotein-9.2 mRNA by chromogenic RNA in situ...
The effect of fenbendazole on drug-metabolizing enzymes in Haemonchus contortus
Šulcová, Kateřina ; Matoušková, Petra (advisor) ; Skálová, Lenka (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Bc. Kateřina Šulcová Supervisor: doc. Ing. Petra Matoušková, Ph.D. Title of diploma thesis: Effect of fenbendazole on biotransformation enzymes of Barber's pole Barber's pole worm (Haemonchus contortus) is a parasitic nematode in the gastrointestinal tract of small ruminants. In infected animals it causes a disease called haemonchoses. Its signs are anaemia, weight loss and animals are in general not thriving. In numerous cases it can cause death. For treatment of this disease substances called anthelmintics are used. Parasite resistance to all groups of these drugs has been a worldwide problem for several years. It is probably due to frequent and inaccurate use of anthelmintics when the parasites are exposed to sub-lethal doses of the drug. Mechanisms of resistence vary depending on anthelmintic group and its mechanism of action. This thesis focuses on studying mechanism of resistence of H. contortus to fenbendazole, a benzimidazole anthelmintic. Changes in expression of some biotransformation enzymes and efflux transporters genes could contribute to resistence. In this study expression of selected genes of biotransformation enzymes and efflux transporters from CYP (cytochromes P450), UGT...
New fungal fucosidases and their synthetic potential
Glozlová, Michaela ; Beránek, Martin (advisor) ; Matoušková, Petra (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Bc. Michaela Glozlová Supervisor: prof. PharmDr. Martin Beránek, Ph.D. Consultants: doc. RNDr. Pavla Bojarová, Ph.D., Mgr. Pavlína Nekvasilová Title of diploma thesis: New fungal fucosidases and their synthetic potential The literary research focuses on α-L-fucosidases, their occurrence, functions, and use in the synthesis of fucosylated compounds, which were also a part of the research, particularly their presence in the human body and involvement in pathological conditions. Furthermore, it explores the oligosaccharides found in human milk, their impact on infant health, and their synthesis, which can be facilitated through the use of α-L-fucosidases. The experimental section involves the analysis of a library of filamentous fungi with the aim of identifying a representative strain with α-L-fucosidase activity towards pNP-α-L-fucoside. Recombinant α-L-fucosidases were selected based on searches in genetic databases and prepared. The feasibility of their production in Pichia pastoris and Escherichia coli was tested, and the produced enzymes were purified by ion exchange chromatography, characterized and their regioselectivity and synthetic potential for the synthesis of fucosylated...
