Recognizing the historical period of interpretation based on the music signal parameterization

The aim of this semestral work is to summarize the existing knowledge from the area of comparison of musical recordings and to implement an evaluation system for determining the period of creation using the music signal parameterization. In the first part of this work are describe representations which can music take. Next, there is a cross-section of parameters that can be extracted from music recordings provides information on the dynamics, tempo, color, or time development of the music’s recording. In the second part is described evaluation system and its individual sub-blocks. The input data for this evaluation system is a database of 56 sound recordings of the first movement of Beethoven’s 5th Symphony. The last chapter is dedicated to a summary of the achieved results.
Saving area and power consumption in 65 nm digital standard cell library

This study aims to investigate multi-bit pulsed latches in comparison with multi-bit flip flops as one of the low-power solutions in 65 nm technology process. Topologies of pulse generators and multi-bit pulsed latches were investigated to find out which can be more suitable. The pulse generator was chosen because of its low power and a small area in comparison with other options. The pulse generator is made of a simple AND logical gate and a double-stacked inverter. The pulsed latch was also chosen because of its low power, small area, and reliability of the circuit. The chosen topology is modified PPCLA. Simulations of the chosen topology had shown that multi-bit flip flops could be replaced with more effective multi-bit pulsed latches.

Diplomová práce se zabývá typy výkonových ztrát v technologii CMOS a následně jejich řešením metodami nízké spotřeby, které lze použít v oblasti zákaznických integrovaných obvodů. Jednou z možných metod pro nízkou spotřebu výkonu je vícebitový pulzní klopný obvod, který lze použít jako náhradu za standardní vícebitový klopný obvod typu master-slave. Vícebitový pulzní klopný obvod se skládá ze dvou částí, pulzního generátoru a pulzního klopného obvodu. V diplomové práci jsou popsané topologie, které mohou být použité. Konečná topologie byla vybraná na základě nízké spotřeby a plochy. Z vybrané topologie je vytvořeno schéma vícebitového pulzního klopného obvodu, které je porovnáno se schématem vícebitového klopného obvodu. Následně jsou vytvořeny layouty potřebných vícebitových pulzních klopných obvodů a porovnány jsou se standardními layouty vícebitových klopných obvodů. V závěru práce jsou navrhnuté vícebitové pulzní klopné obvody odsimulovány v jednoduchém designu.
Recently excavated burial grounds of the Únětice culture near Kolín (Central Bohemia)

Theses is focused on analysis of early bronze age burial sites excavated during the rescue excavation caused by highway construction around the town of Kolín. Two burial sites in a range of 70 and 13 burials were examined. Information from both of these burial sites were analyzed on the basis of fieldwork research from years 2008-2009. The larger burial site was ascribed to late Únětice Culture by material finds, the smaller burial site was recognized as early Únětice Culture. Burial rite had a same standard characteristic as in other Únětice regions. Local differences in burial rite dwelled in a specific finds, which are not common in an eastern part of Middle Bohemia (amber, animal blade bone). Secondary burial disturbances was traced in all kind of burials of men, women and children. The reason why was secondary burial disturbance not connected to only one group of burials, but to all of them is not clear. Types of ceramic shapes support connection with Moravian region. Thanks to new knowledge about Únětice culture in the eastern part of Middle Bohemia, we can observe, that settlement concentration was in late Únětice culture same as in other Únětice regions. Agreement to make a contract (pactum de contrahendo)

The purpose of this Master's degree thesis is to deal with preparatory institute of private contractual law pactum de contrahendo. This institute passed through to contemporary legislative conception long progress, currently it is in Czech Republic simultaneously regulated in two codes and at the present time performed re-codification of the Czech private law is in submission of the new Czech Civil Code its explicit regulation also contained. The thesis focuses on preliminary agreement being concluded according to valid Czech Civil Code, on its purpose, utilization and function. The thesis characterises its subjects, subject- matter, content and the process between entering into the preliminary agreement and the future agreement. In several cases the thesis provides with alternative approaches to a particular issue, presents judicial opinions of legal experts and pursues case law. The thesis also describes the development of regulation of agreement to make a contract in the context of Czech law within the range, which was appropriate to the subject, the thesis refers to similar legal institutes and compares current valid parallel regulation of preliminary agreements among themselves and with newly suggested regulation with the aim to find advantages and disadvantages of particular regulation....
Functional analysis of hPrp8 mutations linked to retinitis pigmentosa.

hPrp8 is an essential pre-mRNA splicing factor. This highly conserved protein is a component of the U5 small ribonucleoprotein particle (U5 snRNP), which constitutes one of the building blocks of the spliceosome. hPrp8 acts as a key regulator of spliceosome activation and interacts directly with U5 snRNA and with the regions of pre-mRNA that are involved in the transesterification reactions during splicing. Mutations in hPrp8 have been shown to cause an autosomal dominant form of retinitis pigmentosa (RP), an inherited disease leading to progressive degeneration of retina. In this study, we analyzed the effects of the RP-associated mutations on the function of hPrp8. Using BAC recombineering, we created mutant variants of hPrp8-GFP construct and we generated stable cell lines expressing the recombinant proteins. The mutant proteins were expressed and localized to the nucleus. However, one of the missense mutations affected the localization and stability of hPrp8. Further experiments suggested that RP-associated mutations affect the ability of hPrp8 to interact with other components of the U5 snRNP and with pre-mRNA. The presented thesis introduces European funds and options for financing of individual projects and their legal framework. Regional and Structural Policy is one of the major policies of the European Union. Its main purpose is to mitigate the differences between countries and regions within the European Union and bring the disadvantaged regions to more progressive level. Summary of the EU structural policies, their purpose, principals and historical background is provided in the first chapter of this thesis. It also provides an overview of the major tools (structural and other funds) used when implementing the Regional and Structural Policy. This introduction is followed by a summary of the legal framework of the European funds and the legal basis of the Structural and Regional Policy. The mutually beneficial idea of supporting regions originates from the basic principles of the European Economic Community/European Union recorded already in the main documents of the EEC/EC - the Treaty of Rome and its amendments. Eukaryotic and prokaryotic nitric oxide synthase - structure-function studies

Nitric oxide (NO) is an important signaling molecule in organisms. It plays a role in wide spectrum of physiological and pathophysiological processes, including vasodilatation, neurotransmission and host defense. The gaseous molecule of NO is produced by oxidative reaction catalyzed by proteins from the family of nitric oxide synthases (NOSs). Three NOS isoforms were identified in mammals, endothelial (eNOS), neuronal (nNOS) and inducible or immunologic (iNOS). Some bacteria harbor genes coding for proteins homologous to the mammalian NOS oxygenase domain and showing NO-producing activity in vitro. NO generated by pathologic organisms such as B. anthracis and S. aureus is supposed to play a critical role in the pathophysiological processes during the infection. Comparative study of bacterial NOS-like proteins and mammalian NOSs confirmed their principal similarity, but also revealed differences in the interactions of distinct bacterial proteins and mammalian NOS isoforms with different analogs of substrate L-arginine and various ligands. On the basis of the kinetics measurement of NO-rebinding a second NO-binding site in the active center of NOS was predicted. CoNct usloN The aim of the thesis was to establish, in the national conditions, technology of preparation of antibody reconrbinant fragrnents and to verífo the cornplete procedure using several model monoclonal antibodies with a potential diagnostic and therapeutic use. For tfuee antibodies' mAb TU-20, M75 and MEM97, recombinant |Ťagments in various formats (scFv fragments monovalent and bivalent, i.e. diabody, and intrabody for intracellular expression) were constructed and for their heterologous expression, vectors allowing expression in E. coli (as cytoplasmic inclusions, periplasmic inclusions and in soluble form) and in Drosophila 32 cells (expression of glycosylated forms of scFv fragments into the medium) were used. In case of proteins expressed in irrsoluble form, especially scFv F|1.2.32, renaturation procedures to obtain active scFv fragments were developed and optimized. The efnect of the length of the linker -(GlyrSer).- (where x is I to 4) connecting the variable domains of the light and heavy chain on the formation of different multimeric forms of scFv rvas studied. For obtaining solell monomeric scFv fragment, the length of 20 amino acid residues tumsd out optimal.
