Title: Quantification of training load in small area games and full ice game Objectives: The aim of this work is to compare the frequency of skills during small area games and full ice game in the training process at young school age. Methods: In the thesis was used a method of indirect observation. I recorded and analyzed 16 practices. I specifically focused on the number of individual game skills performed in small area games and full ice game. The study was conducted at the SC Kolín during the 2023/2024 season, involving 18 participants with an average age 11 ± 0,42. For comparison, I used quantitative analysis and percentage expression. Data are converted to the average number per minute. The results are presented by tables and box plots. Results: Research discovered that the most effective game in terms of individual game skills (shooting, passing, takeaway and pass interception) is 3-3 plus shooter. Players in this game achieved an average of 0,91 shot attempts, 1,78 passes and 0,18 intercepted passes per minute of game play. The small area game 3-3 with offside achieved the best result in takeaways, 0,31 average per minute of game play. The full ice game had the lowest average of the results in all observed parameters. Conclusion: In all observed parameters small area games achieved better...
Attitudes of the Czech Population Towards the Environment: Determinants of More Sustainable Traffic Behavior
Bémová, Alice ; Soukup, Petr (advisor) ; Urban, Jan (referee)
The thesis examines the relationship and attitudes of the citizens of the Czech Republic towards the environment and its protection. Specifically, it explores the willingness to reduce the use of automobiles and decision-making regarding the purchase of electric vehicles, as well as the factors influencing these choices. The introduction of the thesis reflects a shift in the perception of the environment from an anthropocentric to an ecocentric conception. The theoretical foundation for the analysis is the Value-Belief-Norm (VBN) theory, which examines the influence of values, beliefs, norms, and sociodemographic factors on environmental behavior, specifically in the context of reducing use of automobiles. The thesis employs a combined research design, including a quantitative survey conducted using the CAWI method followed by semi-structured interviews with selected respondents. The analysis reveals a growing trend of postmaterialism and an interest in supporting the sustainability of the environment in the Czech Republic. The regression analysis identifies key predictors of the willingness to reduce car use, including awareness of environmental issues, perception of the danger of automobiles to the air quality, willingness to pay for environmental protection, ecocentric attitudes, postmaterialist...
Physiology of death in relation to the transplant program
Schmidt, Matouš ; Pachl, Jan (advisor) ; Hejna, Petr (referee) ; Pokorná, Eva (referee)
Physiology of death in relation to the transplant program, MUDr. Matouš Schmidt Summary Introduction: The physiological processes associated with dying are well known, nevertheless there is very little scientific data describing these events in greater depth during the perimortal period. However, the data taken from this period may be important for a better understanding of post-mortem organ donation and for establishing a safe interval between circulatory arrest and initiation of organ delivery of DCD donors. The definition of death is based on its irreversibility. When death occurs, it is determined by proof of circulatory arrest. This means the exclusion of any possible spontaneous resumption of circulation (so-called autoresuscitation). This phenomenon has been reported in individual cases but has not yet been the subject any extensive scientific research. The scientific goal of this work was to describe the physiological processes during dying. It focuses on two main areas: firstly, circulatory death (including the phenomenon of autoresuscitation) and secondly, the metabolic change of ions and the acid-base balance during the perimortal period. Methodology: The research was conducted as part of an international academic prospective multicentre study (conducted in Canada, the Czech Republic, and the...

Mammalian proteins carrying zinc finger arrays
Vobruba, Adam ; Svoboda, Petr (advisor) ; Mašek, Jan (referee)
A zinc finger is a small peptide motif stabilised by a single zinc ion, best known for its capability to specifically bind a 3-nucleotide sequence of DNA, depending on the exact amino acids present in the DNA-binding positions. Zinc fingers are unique in their ability to freely link together and form longer tandem arrays, which can bind DNA targets of any length and sequence determined by the combination of individual fingers. These arrays can easily mutate and be rebuilt to change binding specificity, allowing great flexibility and helping zinc fingers to their widespread presence in numerous endogenous proteins of various functions. This property of zinc finger arrays also made them a suitable tool for the creation of custom DNA- binding domains for genetic engineering. This thesis provides an overview of the discovery, structure and function of these domains and then reviews and discusses selected naturally occurring mammalian zinc finger proteins and their properties, showcasing diverse uses zinc finger arrays have been adapted for throughout evolution. The history and future of zinc fingers in artificial proteins created for gene therapy and research are discussed as well. Keywords: zinc finger, ZnF, KRAB, KZFP, CTCF, PRDM9, ZFN, mammals
The role of the nascent polypeptide-associated complex (NAC) subunit β in Arabidopsis thaliana response to various abiotic stresses
Šesták, Petr ; Fíla, Jan (advisor) ; Vosolsobě, Stanislav (referee)
Nascent polypeptide-associated complex (NAC) represents a heterodimeric complex composed of an α- and a β-subunit. It is conserved in eukaryotic organisms and has also been found in archaea. This complex binds to newly synthesized proteins emerging from the ribosome and participates in their targeting to the endoplasmic reticulum. In plants, the NAC complex has been studied in the context of tolerance to various abiotic stresses. In our laboratory, we have previously characterized a T-DNA insertion mutant of Arabidopsis thaliana in both NACβ-encoding genes that exhibited a number of phenotypic defects. These included for instance the appearance of more floral organs, reduced pollen grain germination, and the production of shorter siliques with less seeds inside. The aim of this diploma thesis was to characterize the role of the NACβ subunit in Arabidopsis thaliana during seed germination under osmotic and salt stress. We have shown that the absence of NACβ leads to a reduced seed germination and a prolonged germination time, as does overexpression of this subunit. We further studied whether stress conditions affect the transcript levels of individual genes encoding for both NAC subunits by RT-qPCR. In order to investigate the molecular mechanisms by which the studied complex could influence the...
Insects as a model group of organisms for the research of knowledge about and attitudes to invertebrate animals
Jelínek, Filip ; Mourek, Jan (advisor) ; Janšta, Petr (referee)
This thesis is a literature review that aims to process the available literature on attitudes towards insects, knowledge about insects and the ways in which the perception of insects by humans can be influenced. In the chapters focused on attitudes towards insects, I show how the emotions of disgust and fear lead to negative views of insects and, conversely, how the perception of their beauty and importance promotes positive views of insects. In the knowledge section, I show how people confuse true information about insects with myths, inaccurately distinguishing between what is an insect and what is not, which insects are dangerous and which are not. In the last chapters, I present the literature that deals with the ways in which attitudes towards insects and knowledge about insects can be influenced. I deal with the effect of urbanization, experiential educational programs and the picture of insects in human culture, namely in language, poetry, fictional literature and fine arts

The role of the HPA axis in the development of affective disorders
Fiedlerová, Alžběta ; Telenský, Petr (advisor) ; Svoboda, Jan (referee)
This bachelor thesis focuses on the influence of the hypothalamic-pituitary-adrenal axis (HPA axis) on the development of affective disorders. Affective disorders such as Major Depressive Disorder or Posttraumatic Stress Syndrom are serious mental illnesses that have a serious impact on the life of the individual suffering from the disorder. The HPA axis plays a key role in the adaptation of individual to stressful situations. Dysfunction of the HPA axis can significantly affect the homeostasis of the organism, due to increased or decreased levels of hormones and neurotransmitters or deregulation of the stress system. This can lead to the development of affective disorders. This thesis focuses on the analysis of the connections between the activity of the HPA axis and the emergence of affective disorders and examines the possibilities of therapeutic treatment of this axis as a potential target for the treatment of these disorders. The results of this review of individual scientific articles support the understanding of the mechanism that links the HPA axis to affective disorders and offer a perspective for future researh. Key words: HPA axis, cortisol, major depressive disorder, anxiety disorder, posttraumatic stress disorder, bipolar disorder
