The first section of this thesis, which is entitled "The issue of ensurance publicly available childcare for children under the age of three - the Czech Republic in the context of European Union", starts by outlining the common themes of the European Union's family policy, followed by the specific strategies for supporting families in selected European countries. The second part then outlines the family support options in the Czech Republic, with special consideration for the issue of childcare services for children under three. The third part presents a concrete model for childcare for underthrees as it has been implemented in the capital of the Federal Republic of Germany,Berlin.
Human gut microbiome: Origin, ontogenetic development, diversity and its use in anthropology

Human gut microbiome is a broad term encompassing all microorganismal life inhabiting the human gut. The bacteria living in the human gut represents the largest group of the human microbiome, make up the significant percentage of the human cellular composition and their genomes comprise a big part of the human genome. Gut microbiome has a significant role in human health and changes throughout the human life in reaction e.g. to change of diet and medical drug usage. This work pursues the acquisition and development of gut microbiome, the factors influencing its formation and diversity, and its use in anthropology.
Depletion of Treg cells for potentiation of cancer treatment with HPMA copolymer-bound cytostatic drug conjugates"

Tumor diseases are severe problem worldwide with increasing number of patients suffering from various types of malignancies. Many of approved therapeutics cause serious side toxicities. Therefore, there are intensive efforts to improve cancer treatment protocols. The aim of this study was to deplete regulatory T (Treg) cells without affecting other immunocompetent cells playing a positive role in tumor eradication. Treg cells were reported to hamper anti-tumor immunity and promote tumor growth and survival. Thus, their selective elimination could lead to induction of anti-tumor responses and tumor rejection if combined with chemotherapy with selected N-(2- hydroxypropyl)methacrylamide (HPMA) copolymer-bound drug conjugates. Original approach was to deplete of Treg cells without the use of anti-CD25 mAb that has been widely exploited for Treg cell elimination; however, its long-term persistence in circulation together with inhibitory effect on activated effector cells (CD25+ ) are its main disadvantages. Thus, Treg cells were sensitized to cell cycle-specific cytostatic drugs via application of IL-2/anti-IL-2 JES6.1 mAb immunocomplexes that induce vigorous selective proliferation of this cell population. Sexual dimorphism in tibial loading during human locomotion

Locomotion is one of the most important qualities of man and has always been associated with survival, foraging ans subsistence. During the course of evolution, men and women developed some form of gender specialization which resulted in different level of mobility between the sexes. Throughout history, the types of subsistence have changed and the degree of sedentism has increased. However, despite all the changes in subsistence, gender specialization and bone robusticity sexual dimorphism in the robusticity of the lower limb bones remained surprisingly stable. Is it possible that women load lower limb bones differenty than men (e.i. due to different body proportions)? The aim of this work is to determine wheter there is sexual dimorphism between the sexes of the recent living population in the cross-sectional properties of the tibia and in the load on the tibia during running. In this work we used images of the tibia from magnetic resonance imaging and kinematic and kinetic data during the run of 20 probands. By using musculoskeletal modeling we estimated the bending moment acting at 50 % of the tibial length as well as the angle of action of this moment. The Influence of upper-limb key muscles strength on self-sufficiency of tetraplegics

This bachelor thesis concerns the influence of upper-limb key muscles strength on self-sufficiency of tetraplegics. The paper is divided into a theoretical and a practical part. The theoretical part contains observations on spinal cord damage as well as health consequences and complications of tetraplegia, which were subjected to closer scrutiny. Furthermore, the paper focuses on self-sufficiency of tetraplegics and on the influences which affect it. It offers a thorough description of the available research on the relationship between muscle strength and self-sufficiency. The practical part describes and compares case reports of two probands of the same motor level of tetraplegia and of a similar strength of upper-limb key muscles. Self- sufficiency of the probands is attested by SCIM and a detailed description of the strategy of transfer from a wheelchair to a bed and back. Eating disorders and the importance of nutrition

This bachelor thesis encompasses problematics of eating disorders, primarily anorexia nervosa and bulimia nervosa. The beginning of the theoretical parts approached eating disorder, their definition and risk factors. The main part of the thesis is focuses on the treatment of eating disordes with cooperation team of professionals. The chapter on therapy is mainly focused on the role dietitian in treatmeant mental anorexia and mental bulimia. The thesis describes an evaluation of the nutritional status, forms of realimentation and a suitable diet. Next chapter define health complications that are an integral part of the disease. Conclusion of the theoretical part explains the term of relapse and includes the points how to prevent it. Practical part consists of an interview with respondents who have an experience with eating disorders. The aim of this research is to find out how nutritional therapist proceeded in the treatment of their diseases and how important was the cooperation with him for them. Design and analysis of the road bike made from composite material

This diploma thesis is focused on the design and analysis of road bike frame made of composite material. The thesis is divided into two parts. In the first part, there is a research focused on the usage of composite materials in relation to making bike frames. The main emphasis is placed on fiber composites, which are most common in cycling. Manufacturing process and production technology are discussed. The aim of the second part is design and the numerical simulation of frame in relation to stand-ard, which is used for the testing of frames. Rigidity and strength are evaluated. The final frame is obtained from the initial design due to several iterations. In conclusion, results are evaluated and recommendations for future work are given.
Parents of Children with Developmental Language Disorder from the perspective of speech therapy intervention

The diploma thesis is from the area of special pedagogy and speech therapy. It deals with the issue of parenthood of children with developmental dysphasia. The thesis is devided into two parts, the theoretical part and the empirical part. The thesis starts with characterisation of the family with disabilities and continues with further developmental dysphasia, its terminology, classification, etiology, symptomatology, diagnostics and therapy are characterized. The thesis defines the possibilities of speech therapy intervention in children with special impaired speech developmental dyspshasia. This main goal is achieved by research. This diploma thesis can be an inspiration for speech therapists and other professionals who work with families and their children with developmental dysphasia. It can also inspire students and other parents even whose child has been diagnosed with this disease. The thesis may be the starting point for further studies of a scientific character that focus on same subject.
The effect of melatonin on rhythmic ATP release from organotypic cultures of the rat SCN

The rhythm of ATP accumulation is an one of examples of circadian rhythmicity which is demonstrated across the animal kingdom. It is located in the suprachiasmatic nucleus of hypothalamus, which is a centre of circadian clock and imposes rhytmicity on a whole organism. The question concering the ATP rhythm in an organotypic culture and the impact of melatonin on it has been discussing in this thesis. The synthesis of melatonin is regulated by the activity of the SCN and the hormon itself is known for its feedback on the SCN and a capability of regulating it. It has been shown that ATP rhythm is significantly inhibited under the constant control of melatonin and this inhibition is dose-dependent on a scale of 0,1-10 nM. In case of one-time applied melatonin in 4 p.m. there is no reduction in the accumulation of extracellular ATP but there is a phase shift in ATP rhythm. It has also been shown that ATP rhythm is inhibited and desynchronized under the control of tetrodotoxin which blocks an electric activity of neurons. These results show that melatonin is capable of inducing phase delay of ATP rhythm in SCN and reducing an amount of extracellular ATP, that the effect of melatonin is probably mediated by specific receptors and lastly that ATP rhythm is a result of cell-cell interaction between neurons...
