Crisis intervention via electronic communication (on-line chat, on-line videochat)

This diploma disertation deals with the topic of on-line crisis intervention (KI further on in the text) in the Czech Republic (CR), specifically the service functioning of the online chat and videochat. This topic has not been paid attention to so far. The topic concerned is a rather new service area which provides service to the clients in a difficult situation on the internet and which has been working for past few years in a few KI centres (workplaces). No professional has tried to search it systematically or describe it in more detail yet. Considering the fact the internet is a widely used medium in our population nowadays, a high future potential of these services can be expected in the field of psychosociology. Therefore this diploma disertation aims to give a detailed description of the on-line chat and videochat functioning and necessary connections at KI centres (workplaces) in order to create a quidebook to the basics of these services for specialists and amateurs who are interested in this topic. Active labour market policy

From seventies of 20th century was in western Europe high and longterm growth of rate of unemployment. With this grew also importance of labour market policy. First were used especially pasive measures. But soonly it turned out that against unemployment must be fighted actively. The importance of active labour market policy grew with the growth of rate of unemployment. In Czech Republic came up the problem of unemployment in nineties. The work deals with usage of instruments of ALMP in Europe and Czech Republic. It explores the efects and contributions of ALMP. It evaluates particular used measures and gives the recommendations how to use ALMP. The end is concentrated on Moravian-Silesian region which is one of the most problematic locality. Study of opioid receptors

1 ABSTRACT In this Thesis, we studied properties of μ-, δ-, and κ-opioid receptors in lymphocytes isolated from rat spleen. This splenocytes were exposed to mitogen concanavalin A or opiate morphine and cultivated for 48 hours. Under physiological conditions, level of opioid receptors in immune cells is very low. Due to various factors such as presence of opioids, mitogens, long-term exposition to stress, expression of these receptors can be amplified. In this study we demonstrated, that concanavalin A causes up-regulation of μ-, δ- and κ-opioid receptors in lymphocytes isolated from rat spleen. In control cells no significant signal of μ- or δ-receptors was observed. In contrast, κ-opioid receptors were detected already in control cells. Concanavalin A stimulation caused a 2.4 - fold increase of these receptors. In lymphocytes treated with morphine only μ-opioid receptors were up-regulated, whereas in control cells, there was no signal for these receptor type. δ-opioid receptors were not detected in control or morphine treated cells. κ-opioid receptors were determined in control and also in morphine affected lymphocytes but the amount of these receptors wasn't changed by morphine. Drama education as a tool for exploring the historical heritage of the City of Prague

This Diploma Thesis deals with the drama education and its use as a tool for teaching of historical topics related to Prague and its monuments. The Diploma Thesis is divided into two parts. The theoretical part characterizes the drama education, enumerates its principles and methods of work. The theoretical part deals with inclusion of history and drama education in Framework educational program. It deals with positives of drama education and its potential problems. The practical part includes sample lessons on the topic of Prague and the history associated with it. These lessons are put into practice and described their process, including some relevant situations for this topic.
The role of aging and stress in autophagy

Autophagy is a nonspecific catabolic mechanism, important for maintaining the cellular homeostasis. Stress condition can increase the activity and support the cell survival. It is well known that normal and pathological aging are associated with a reduced autophagic potential and that experimental induction of autophagy leads to higher longevity of certain model organisms. This decreasing may be facilitated by other factors, such as chronic stress. Higher age and chronic stress are two factors contributing to the development of neurodegenerative diseases, in this context the mechanism of autophagy is now actively studied. We wanted to know if and how the combination of these two factors may influence the activity of autophagy. In our study, we investigated the role of aging and chronic stress, induced by social isolation in Wistar rats at the age of 6 (N= 16) and 12 (N= 16) months. The animals in experimental groups were separated in the individual cages for 4 weeks. Control groups animals were housed in the pair. We measured the markers of autophagy expression (Beclin-1, LC3-II, mTOR) in the hippocampus and striatum of socially isolated and control groups animals. The Key Determinants of Plane Ticket Price Dispersion

This thesis examines the plane tickets price development using both descriptive evidence and econometric analysis. The observed phenomenon of rising and falling fares in period close to departure is described and compared with the extensively developed theories, such as demand utilization and stochastic peak load pricing. Studying the fares observed on European routes using descriptive evidence revealed that the airlines accommodate fast to the uncertain demand. In the econometric part, the key factors influencing the price dispersion are determined. The contribution of this thesis is mainly in the econometric approach, as the price fluctuations are measured weekly using the coefficient of variation. This made it possible to compare how the different flight or market characteristics affect price dispersion in different week to departure. It was shown that number of sold seats, the load factor, is the crucial factor.
The role of biofluid biomarkers in the differential diagnosis of neurodegenerative diseases

Until recently, the diagnosis of Alzheimer's disease and other neurodegenerative diseases was dependent only on clinical expression, the definitive confirmation of which was possible only post- mortem. Currently, biofluid biomarkers represent a reliable source for the diagnosis of serious neurodegenerative diseases already during the patient's lifetime. The identification of reliable biomarkers is essential for increasing diagnostic accuracy, monitoring the clinical condition, more precise selection of individuals for clinical trials and the associated development of causal treatment. The aim of the bachelor's thesis was to conduct a thorough literature review focusing on clinically and experimentally used biological markers in the diagnosis of neurodegenerative diseases and their role in the pathophysiology of these diseases. The impact of brain-derived neurotrophic factor (BDNF) gene polymorphism and BDNF levels for prediction, diagnosis and follow-up of cognitive deficit in neurodegenerative diseases

Alzheimer's disease (AD) is a slowly progressing neurodegenerative disease characterized by an extracellular accumulation of senile plaques and intracellular neurofibrillary tangles. Apart from these hallmarks, AD is very heterogeneous, especially at the age of onset and the rate of progression. Genetic polymorphisms are key modulators influencing these factors. One of them is a polymorphism for the Brain-derived neurotrophic factor (BDNF), a neurotrophin involved in the synaptic plasticity in the hippocampus and medial temporal lobe structures, areas primarily affected in AD. The thesis aimed to investigate the effect of BDNF Va66Met polymorphism together with the main genetic risk factor for sporadic AD, Apolipoprotein E (APOE) polymorphism, on cognitive functions and structural brain changes. We have shown that the combination of risk alleles BDNF Met and APOE ɛ4 is associated with more severe impairment in episodic memory, egocentric orientation and smaller volumes of medial temporal lobe structures in individuals with amnestic mild cognitive impairment (aMCI), and allocentric orientation in cognitively unimpaired individuals compared to non-carriers.
