Survey and Diagnostics of Building Construction

The main goal of this diploma thesis is diagnostics and vocational training in assessment of existing structure of the center of leisure time for children. The teoretical part is focused on vocational training in assessment of existing structures by ČSN ISO 13 822 - Bases for design of structures - Assessment of existing structures and assesment of brick structures. The designing strength of masonry and mortar is defined in the end of theoretical part. The practical part continues with real construction, where diagnostic methods based on technical survey are applied and in the end describes present state of real construction with plan for required steps to secure safety. As a result of this thesis is also a demonstration static report.
Child with autistic spectrum disorder

This bachelor thesis is focused on the topic of autistic spectrum disorders. The main objective is to compare ways of educating pre-school children in a regular and special kindergarten, and a description of the benefits and deficiencies in the life of a person with ASD. The first part of the work is focused on characteristics of autism, definition of individual concepts, classification, possibilities and means of education based on sources of professional literature. The second part describes the possibilities of education in a special kindergarten, where two boys of the same age and a diagnosis of child autism were selected for qualitative research. The work was done by the method of participating observation. I became a direct actor in the educational process. Research findings are based on responses, activities, approaches of individual clients and other participants in the whole process in home and school facilities. Here are some of the specific methods that are used in those institutions, and their effects on the observed clients. By regularly attending selected places, I evaluated whether the chosen techniques were successful in their versatile development, and I compared the mutual collaboration of the family and the school. A rich description of the two observed clients and their individual manifestations was created, based on the sorted data. Finally, I discuss the results of the whole research, the benefits and possible shortcomings that take place in the life of the clients, and my own view of the given situation.
Synthesis of Neuraminidase binders suitable for theranostics

Influenza viruses cause respiratory illnesses which can vary in severity depending on the strain of the virus, as well as the age and health condition of the host. Influenza remains a major threat to public health due to its nature prone to suffer mutations. As a result, vaccines have to be reformulated annually and new strains may cause sporadic global pandemics. Furthermore, the recent emergence of resistant strains of the virus against the current standard of care (oseltamivir and zanamivir) underlines the need of novel anti-influenza therapeutics. The aim of this dissertation work is to contribute to the discovery of new anti-influenza inhibitors either by rational drug-design and optimization of oseltamivir structure, or by developing screening assays suitable for the discovery of novel inhibitors of the enzymes neuraminidase or RNA-polymerase. 2 ABSTRACT In the Alternative DSM-5 Model for Personality Disorders (AMPD), the diagnosis of specific personality disorders is obtained through an evaluation of the level of impairment in personality functioning and an assessment of dimensional personality traits associated in 25 "lower order" facets and 5 "higher order" domains. The Personality Inventory for DSM-5 (PID-5) was developed for examination of personality traits within this system. This Ph.D. thesis covers five relevant publications regarding AMPD, particularly PID-5. First, the authors introduce the theoretical background of the PID-5. Attention is paid on its ease of use, data interpretation and use of these data for treatment planning in different clinical settings. Two empirical studies test PID-5 psychometric properties (internal consistency, validity, discrimination capacity and unidimensional structure) in the sample of community volunteers (n=351) and a clinical sample of psychiatric patients (n=143). PID-5 was administered individually and in a group setting using pen-paper method and online data collection. 33 respondents completed the inventory twice to check test-retest reliability. Fasciolid flukes: from genes to diagnostic tools

Liver flukes of the family Fasciolidae are parasites of mammals including human. Fascioloides magna and Fasciola hepatica are considered as a veterinary and medically important species occurring also in the Czech Republic. Fascioloides magna and F. hepatica infect wide spectrum of wild and domestic ruminants and in case of F. hepatica human can be also infected. Both flukes are responsible for damage of liver tissue and/or bile-ducts of their definitive hosts causing weight lose, anemia, reduced productivity and in specific cases the death of the host. Effective diagnosis plays the key role in control of F. hepatica and F. magna infections. Current diagnostics is predominantly based on serodiagnostic methods using specific antigens e.g. from excretory-secretory products (ESPs). Due to heterogenity of ESPs, such diagnostic markers can lack the specificity and also the reproducibility of the method is poor. Particular proteins of ESPs are often used in diagnostics of fasciolid flukes. Such approach requires biological material and laboratory procedures associated with identification, purification and antigenicity testing of selected proteins. Bachelor thesis deals with the topic of cystic fibrosis. It is a serious autosomal recessive hereditary disease. Cystic fibrosis can not be cured, it is only possible to treat the complications of this disease. The thesis is divided into the theoretical and practical part. The theoretical part is focused on the occurrence of cystic fibrosis in the world and in the Czech Republic, history, the principle of inheritance of cystic fibrosis and there are presented the symptoms and complications associated with cystic fibrosis. There are also presented diagnostic methods and principles, therapy of the disease. In the practical part of the bachelor thesis there is a public survey on the topic of cystic fibrosis. Osgood-Schlatter Abstrakt bakalářské práce: Tato bakalářská práce se zabývá sledováním výskytu etiologických faktorů u pacientů s diagnostikovaným M. Osgood-Schlatter a průběhu terapie dle sestaveného léčebného postupu. Práce se skládá z teoretické a praktické části. Teoretická část obsahuje rešeršní zpracování české a zahraniční literatury dané problematiky. V praktické části proběhlo u 40 probandů měření antropometrických parametrů, svalové síly extensorů a flexorů kolenního kloubu pomocí izokinetického dynamometru, svalového zkrácení m. quadriceps femoris, ischiokrurálních svalů, m. iliopsoas, m. tensor fasciae latae a rozsahu pohybu v hlezenním kloubu dle standardizovaných testů. U pěti probandů se v další fázi praktické části objevila diagnóza MOS. Každý z těchto probandů měl výrazně zkrácený m. quadriceps femoris, nepoměr síly mezi flexory a extenzory kolenního kloubu a omezený rozsah pohybu v hlezenním kloubu. Title: Influence of different way of strength training on body composition, postural stability and muscle and explosive strength in Men's Physique and Street Workout Athletes Objectives: The aim of the thesis was to find out the level and the difference between the parameters of body constitution, postural stability and muscular and explosive force of upper and lower limbs with groups of Men's Physique and Street Workout athletes. Methods: The research sample was represented by two groups (Men's Physique competitors and Street Workout athletes). We assessed chosen parameters of body constitution by Tanita MC-980MA, postural stability by RS Footscan, explosive force (Kistler) and muscular force (Cybex Humac Norm and Takei A5401). Assessed parameters of body constitution were percentage of body fat and fatless matter. In the tests of postural stability, we assessed total travel way of the center of pressure (TTW) in chosen stands (open/closed eyes and one- leg left/right). When testing explosive force, overall produced maximal force and height of the leap were assessed. Muscular force of upper limbs was evaluated by produced force in kg and muscular force of lower limbs was assessed with the help of muscular force moment in concentric muscle activity with angular velocity 60ř·s-1. Personality psychopathology measured by alternative model of DSM-V

The DSM-5 Alternative Personality Disorder Model includes Personality Inventory for DSM-5 (PID-5) that assess 25 specific personality traits in 5 domains. The aim of this thesis was to evaluate the method and its clinical benefits, especially by comparing it with the IPDE structured interview method on a sample of N = 29 psychiatric patients. Quantitative analysis of the results of both methods revealed a number of statistically significant correlations between the IPDE dimensional scores and the individual PID-5 traits of corresponding disorders. For disorders equally defined by both approach, the IPDE dimensional scores and PID-5 individual traits correlated by majority. Due to the overall low prevalence of undisputed personality disorder diagnosis in the sample, a qualitative analysis of 11 selected cases was performed. This analysis revealed the overlapping of the personality traits derived from both methods in 10 of these 11 cases. The methods differ in the assignment of a specific personality disorder as a diagnosis, but this is due in particular to a different theoretical definition of the methods. 6 ABSTRACT Background The existence of risks of new work-related disorders and occupational diseases in the society is constantly increasing in relation to the fast and continuous development of working conditions, techniques used, materials and overall acceleration of the working pace. Aim of the study The aim of the study is to analyse new risks in the working environment and to evaluate early diagnostic possibilities of new occupational diseases including their prevention. The study also compares the current situation in this field in the Czech Republic and other European countries. Materials and methods The doctoral dissertation is a multiple manuscript thesis focusing on one common topic - new occupational diseases. The first publication studied the acknowledgement of low-back pain disease caused by overload as an occupational disease. Specialists from European countries were inquired via questionnaire focusing on diagnostic and evaluation criteria of this disease in their countries. Representatives from countries acknowledging low-back pain diseases caused by overload provided further evaluation criteria and number of cases.
