Lepidoptera Community of the Woodland Complex Loučany near Klatovy

This thesis is focused on faunistic survey of Macrolepidoptera in Loučany (foothills of the Šumava Mts, Czech Republic). The research, involving different capture methods, proceeded from April to November 2015 on given transect and several other habitats. Based on the results (including partial data from 2013 and the last two decades of the 20th century), an inventory of identified species of Macrolepidoptera was made. Altogether 205 species from 16 families were found in Loučany. The most important species are for example the Black-veined White (Aporia crataegi), High Brown Fritillary (Argynnis adippe), Pearl-bordered Fritillary (Boloria euphrosyne), Purple Emperor (Apatura iris), Spruce Carpet (Thera britannica) and Sandy Carpet (Perizoma affinitata). The environmental parameters of experimental sites were noticed during research and the habitat preferences of species were analysed using specialized literature. Altogether 186 species (90,7 % of all identified species) are linked to some kind of woodland habitat. To support present biodiversity of the community of Macrolepidoptera, it is necessary to practice a suitable woodland management, conserving mosaicity and open canopy. Trypanosomes from the T. theileri group in ungulates in the Czech Republic

The Trypanosoma theileri group are blood parasites of ungulates. Confirmed vectors of these protists are horseflies or sheep keds for T. melophagium, but these trypanosomes were also detected in deer keds, mosquitoes and phlebotomus. We targeted on the investigation of possibility mosquitoes acting as vectors and additionally we targeted on the prevalence of T. theileri in mosquitoes at selected locations in the Czech Republic due to previous positive detection there. The measured prevalence has reached 6,4 % in mosquitoes, 4 % in deer keds and 16,7 % in sheep keds. The aim of experimental infections was to discover a potential of mosquitoes and phlebotomus acting as vectors of the T. theileri. Phlebotomus perniciosus and three species of mosquitoes Culex pipiens molestus, Culex pipiens quinquefasciatus and Aedes vexans were used for those experiments. Furthermore, isolates of T. theileri and T. melophagium from different sources were used. The divergence among these isolates and among used species of insect was found. The best results were achieved with Ae. aegypti and isolates from mosquitoes, where prevalence had reached 90,8 %. Effect of breeding fidelity on reproductive sukccess of diving ducks

Breeding site fidelity and its effect on reproductive success was investigated in two diving ducks species, i.e. Common Pochard (Aythya ferina) and Tufted Duck (Aythya fuligula) in the Trebon region and the surroundings using caught and individually marked females, searching for nests, and monitoring of marked individuals and its broods. In both studied species, the fidelity ratio (apparent survival, MARK software) was around 65 %. Evaluating the influencing factors, impact of previous reproductive success in the phase of rearing ducklings was found to be the only one statistically significant factor. Furthermore, we compared reproductive parameters (i.e clutch size, laying date, reproductive success) in the first and subsequent year of study. The statistically significant relationship was found between the timing of nesting in the first and in any subsequent year in the Tufted Duck . When comparing the reproductive success in relation to hatched or unhatched clutches and reared or not reared ducklings, we do not confirm any significant shift. Sex Determination in Long-eared Owl (Asio otus) by means of PCR DNA amplification

This thesis is focused on determining the sex of the Long-eared Owl (Asio otus). Bird feathers were used during determination that were taken from owl`s back while capturing and ringing. We have attempted to differentiate the sex of captured Long-eared Owl by DNA isolation from blood platelets in the bird quail and subsequent PCR reaction. This thesis also describes the PCR method, which is one of the most used methods in the todays laboratories of many different field of science. I also mention the individual components that are needed for the process of this reaction, its history and its individual steps. Part of my diploma thesis is also the characteristic of the tested order and possible gender division based on morphological features. On this basis, there is also mention of the sexual dimorphism of birds, their plumage and its subsequent coloring. At birds and also at other animals are used different techniques of molecular determination of sex, which I also refer to in my work. Carabid Beetle Breeding at School

The main focus of this work is the collection and breeding of ground beetles (Carabidae) for use at schools. In the theoretical part, attention is paid to the morphology of images and larval developmental stages; occurrence, food specialization, reproduction and importance of carabids. The following is an overview of selected representatives that can be found in Europe. Furthermore, the collection of insects is generally described, including methods of trapping ground beetles. In addition to the individual collection, various types of traps can be used as well. Part of the work is not only the theoretical part, but also a practical description of the implementation of own collection and breeding, which were performed on the base of the theoretical part. A total of 27 individuals were collected at four sites in the Hankovec Forest, which is located in the Karlštejn National Nature Reserve. The method of individual collection and collection using traps was used for collection. Of the three types of traps, the jar proved to be the most effective. 15 representatives were used to carry out the research, and were placed in three types of insectariums. The methodology describes the procedure of bat capturing, their basic determination, the anesthesia before sperm collection, the sperm sampling by electroejaculation and the procedure of subsequent analysis using the CASA equipment (Computer Assisted Sperm Analysis).

Effect of urban gradient on composition of bat communities in Central European landscapes.

Bats (Chiroptera) are usually considered as a group greatly endangered by destructing their habitats through the human activity. However, a significant part of bat species at the central Europe use human buildings as shelters, moreover the natural shelters are marginal or unknown in several species. Considering the linkage of the great part of our bat species to the fragmented area and in connection with a long term human influence on the central european landscape we ask whether at least part of our species see human activity through providing proper shelters as positive. Based on the analysis of landscape structure and ruggedness effect on density, diversity and population structure of bat assamblages acquired by standardized mist net captures located in lowlands (elevations below 500 m) of Czech Republic (204 localities, 3585 bats, 22 bat species), we found that even though most of reactions were species specific and few species seemed to be positively associated with urban environments, most of the bat species rely on the broad-leaved forest cover and other natural habitats such as water bodies. My bachelor thesis deals with an ash dieback which is caused by a pathogen called Hymenoscyphus fraxineus. This thesis deals with the evaluation of climate conditions in comparison with the occurrence of spores of the pathogen. The practical part of the thesis is based on air sampling at the research area in the Hackerova školka. Data were collected with air sampler of burkard´s type. Processing of collected samples followed in laboratory. Data were then analyzed.
