Treatment options for untreatable amoeba Naegleria fowleri

Naegleria fowleri is a free-living protist which can penetrate the body of a previously healthy person and cause meningoencephalitis. The rapid course, disproportionately strong immune response and the lack of effective drugs are the reasons why this disease almost always ends fatally. Due to the relative rarity of this disease, it is not lucrative for pharmaceutical companies to develop a completely new drug. The only way to search for effective drugs is to test those that have already been approved for another purpose. However, clinical tests are not possible due to the low morbidity and rapid course of the disease, and research in this area is thus limited to animal models of the disease and evaluation of the effect of drugs against the pathogen in vitro. Currently used medication includes drugs that are highly toxic or poorly penetrate the site of inflammation. In the future, more effective chemotherapy against N. fowleri and more gentle towards patient could be used. This work summarizes knowledge about pathogenic amoeba, currently used drugs and presents new experimentally tested therapeutics.

Practical bachelor thesis Red steady go - story about prostitute Tereza uses the form of a broadcast documentary, which aims to capture and show the story comprehensively in the different roles of the respondent - as a prostitute, daughter, partner and an addict. The theoretical part defines the fundmental terms - the definition and first theoretical approach of prostitution, street, risk sex and gender. Furthermore divides the various forms of prostitution - street, private and club. The definition of the documentary form of the practical part, as well as a summary of the life of my respondent, is covered. Methodology of the work is described. Throughout the many levels of the character the audiodocumentary (practical part) seeks the reasons and experiences, that resulted in the present story of the respondent. The general level in the story is represented by a therapist and adictologist Ilona Preslová. Individual thematic parts are separeted by musical elements.
Selective screening of antiparasitic substances

The mitochondrion is the key organelle of the cell. It is involved in energy metabolism, the Krebs cycle, the respiratory chain, and the formation of Fe-S clusters, which form the structure of many proteins. Therefore, disruption of the mitochondria is usually fatal to the cell, especially if it contains only one mitochondrion, as is the case in Trypanosoma brucei brucei. The T. b. brucei parasite has become the primary model organism for our study because its basic biochemical processes are already well described, including mitochondrial metabolism, and it is also easy to cultivate in the laboratory. Other model organisms we used in this study were Leishmania mexicana, Cryptococcus neoformans and Candida albicans. All these organisms are causative agents of serious diseases, and although there are drugs against them, new drugs need to be developed due to the frequent emergence of resistances. Mitochondria-targeting drugs are promising for the pharmaceutical industry. Addictive substances and Visual Art

This diploma thesis discusses the topic of addictive substances and their influence on artistic creation. In the theoretical part, I first focus on the search for their connection in history on various artifacts that could be found. Among the most common are cave paintings, which I also list in my thesis, as well as sculptures and illustrations. Subsequently, I define the distribution of the types of individual addictive substances that are most often associated with visual artists, and which I describe later. These include alcohol, cannabis drugs, stimulants, psychotropic substances, and opiates. Furthermore, I characterize visual artists who were influenced by addictive substances in their work at that time. I present exhibitions that have a certain connection with this topic, and in the last line of my work, I focus on the preventive component of the relationship between school and family. In the didactic part, together with the pupils of the eight-year high school, we deal with the issue of these substances and realize an art series with the name that this diploma thesis also bears, i.e. "Addictive Substances and Visual Arts." Drug Prevention in Low-thershold Facilities for Children and Young People in Terms of Social Work

The submitted bachelor's thesis deals with drug prevention in low-threshold facilities for children and young people. The main goal was to find out how drug prevention is implemented and three research questions were developed to identify it. The Bachelor thesis is divided into two parts. The theoretical part deals with drug prevention and policy, the historical context of drug policy, low-threshold facilities and prevention in them. Qualitative research, polling methods and data analysis were chosen for the practical part. The semi-structured interview was chosen as a technique. The research set consists of 5 informers who are social workers in low-threshold facilities. Informants were approached through email communications. The results of the research were evaluated using the open-coding method. This method produced a total of 10 categories with codes and each category contains at least two or more codes. Research has found that specifically drug prevention takes many forms, but on a whole picture the devices agree. Prevention programs fall under the educational, educational and activation activities of low-threshold facilities. This activity is anchored in the Social Services Act 108/2006 Coll. Low-threshold devices are anchored in this Act under Section 62. Furthermore, it was found that drugs, addictive substances and addiction in general are the most common and popular topics among clients encountered by workers when carrying out prevention activities. This Bachelor thesis offers a glimpse into drug prevention in low-threshold facilities. The results can be used by these facilities or social workers, who carry out anti-drug prevention activities, to improve them or make them more transparent.
Study of drugs intake by lettuce

The aim of this thesis is to analyze the content of selected groups of drugs in cultivated lettuce. The theoretical part includes a chapter dedicated to the drugs used, their use, physicochemical properties, and their impact on the environment. Other chapters focused on analytical methods for determining drugs, hydroponic cultivation of lettuce, and some parameters of ecotoxicological tests. The experimental part began with the optimization of the QuEChERS extraction procedure. The optimized procedure was then applied to hydroponically grown samples of roots and leaves of lettuce containing selected groups of drugs, which were subsequently analyzed using liquid chromatography – mass spectrometry. The experimental part also addressed the ecotoxicological impact of different drug concentrations on hydroponically grown lettuce.
