Evaluation of pH effect on solubility and precipitation of proteins from hemp (\kur{Cannabis sativa} L.) meal

In this thesis was examined the influence of pH on solubility and precipitation of proteins extracted from the flour of the Cannabis sativa L plant. For this experiment was chosen the pH in the range of 3 to 11. Monoecious breeds of canabis USO 31 and Fedora 17 were chosen for this purpose. Laboratory methods were applied after the protein extraction, in order to define the content of nitrate substances which was the highest at pH 11. Protein spectrums were detected by the SDS-PAGE analysis where especially in an alkaline environment, globuline fraction was present at a greater amount than was the albuminium one. The next step was the isoelectric precipitation of extracted proteins within the variances containing high values of nitrate substances. The content of the nitrate substances in the protein extract was spanning between 56,3 % to 92,9 %. Based on the results of the experiment, is is feasible to conclude that Fedora 17, which was extracted at the alkaline pH 11 and isoelectrically precipitate to pH value 5 and 6, contained the highest amount of protein.
The comparison of legal regulations of the sickness insurance in the Czech Republic and the Federal Republic of Germany

This submitted thesis deals with legal regulation of the sickness insurance. The thesis consists of nine chapters. The fourth to the seventh chapters, which are based on the legal regulation in the Czech Republic, and the ninth chapter, which is based on the legal regulation of the sickness insurance in the Federal Republic of Germany, are fundamental. The purpose of this thesis is to analyse the legal regulation of the sickness insurance in the Czech Republic and the basic institutes in the legal regulation in the Federal Republic of Germany and compare these two legal regulations. The research is supplemented by a historical survey of the social security, especially the sickness insurance, and by a chapter on the legal regulation valid in the Czech Republic till the end of 2008 and by a chapter focused on the legal regulation of the sickness insurance according to the regulations of the European Union. On the bases of the comparison of both the legal regulations I have arrived at the conclusion that the legal regulations are similar and contain similar institutes because of the close historical development and the social structures. The main sources of information were professional publications and commentaries, articles published in professional journals and web pages.
Oldtown's gradual (NK ČR, XVII.A.40), 1561-1567, from the manufactory of Jan Taborsky of Klokotska Hora

Oldtown's gradual The aim of this work is a monographic elaboration of the Oldtown's gradual (XVII.A.40), from 1561 to 1567, in the context of the work of Jan Taborsky of Klokotska Hora, with the emphasis on connotation of Žlutice gradual. The work elaborates and critically appraises the literature about the genesis and decor, deeply analyses its illuminations (through formal and iconographic analysis) and gives basic orientation in questions of analogies of its decor. Another goal of this work is a summarization of the decor of Oldtown's gradual and elaborated analysis of its illuminations. Oldtown's gradual is one of the best Renaissance manuscripts. The penman was Jan Taborsky of Klokotska Hora; chapter 2.1. deals with his life and production. Matous Ornys of Lindperk was the limner; chapter 2.2. deals with his work. Indisputably, he belongs to the best limners of the 16th century. The decor of the gradual is described in the chapter 4.2. Illuminations of gradual have conventional scheme of choral manuscripts. A lot of analogies are to be found in the medieval and also Renaissance manuscripts. The medieval architecture of the Mendicant orders in Silesia

The subject of this dissertation thesis is medieval architecture of mendicant orders in Silesia and focuses on convent churches of the Minorites and Dominicans. The chosen subject has not been hitherto treated in detail in specialized literature neither in Polish, German or Czech art history. Mendicant orders in Silesia are studied especially by historians who research into medieval history of these orders and, above all, the formation and development of order provinces. The focal points of the thesis research concern preserved buildings of mendicants orders built in the period between the 13th and 14th centuries. Every individual building is presented as a monograph in the form of an extended catalogue entry. The catalogue of medieval buildings of mendicant orders is the crucial part of this dissertation thesis. In this list, individual buildings are divided according to orders. First, we mention history of individual medieval towns and history of particular convent complexes. Further, we focus on the location of mendicant buildings within medieval urbanism of towns. Renaissance arcades typology of royal architectural works in Bohemia around 1550 A.D. Kostelec nad Černými Lesy, Poděbrady, Brandýs nad Labem. Chateaux owned by ruler Ferdinand I. and situated outside of Prague served as a hunting residences. The buildings became king's property in the 1540th . Various reasons for rebuilding led to conversion from gothic castles to comfortable renaissance chateaux. Renaissance appearance of chateaux with an emphasis on the form of courtyards and arcade's corridors are the main topic of this thesis. Types of arcades are examined on three selected buildings, chateaux in Brandýs nad Labem, Kostelec nad Černými Lesy and Poděbrady. For entire overview of morphology used in renaissance arcades passages it is important to point out noble mansions in whose courts the arcade's corridors were used as well. This thesis specifically attempts to bring the analogy to the selected buildings in the Czech environment and also in foreign countries. The examples in the treatises and the inspiration for certain types of arcades from Antique to the Renaissance period are also reminded.
Church Kindergarten Management

This bachelor thesis focuses on the management of church kindergarten (CK). It deals with the specifics of the CK and differences due to the municipal kindergarten. The aim is to compare sub-areas of management of CK and municipal kindergartens. Based on this I would like to find out whether there are differences between church kindergarten and municipal kindergartens management. The work should expand the awareness of professionals and managers and to facilitate the orientation of the church kindergarten, especially for beginners. Legislation looks at religious schools like private schools, but it is a separate category. In the practical part completion of a questionnaire and guided interview was chosen as the research method. The research focuses on the differences in the areas of local church nursery and municipal kindergarten management. Another endpoint is the view of managers on the church kindergarten. Since these differences are mainly legislative this is rather supplement to the theoretical part, where differences and specific have already been revealed.

Gene expression analysis in patients with myelodysplastic syndrome

Myelodysplastic syndrome (MDS) is a clonal disorder affecting maturation and differentiation of hematopoietic stem cells. MDS is characterized by ineffective hematopoiesis resulting in peripheral blood cytopenia in at least one lineage, and increased risk of development of acute myeloid leukemia (AML). This disorder mostly affects the elderly, with the incidence of 10-50/100000 people per year. Heterogeneity of MDS is further underlined by the presence of both idiopathic primary and secondary therapy-related forms, which arise from accumulation of mutational events induced by cytotoxic therapy (exposure to alkylating agents or after therapy with topoisomerase II inhibitors). In this thesis, we focused on determination of differential gene expression between MDS patients and control subjects using different microarray platforms. Using nylon membranes, we detected 4 genes (ERCC1, FLT1, NME4, PCNA) with increasing expression pattern in advanced MDS stages, which correlated with disease progression. Their strong up- regulation was also observed in patients with de novo AML, suggesting their involvement in the leukemic transformation of MDS.
