Prediction of effects of single point mutations on protein-nucleic acids interactions

Single point mutations are the most common type of mutations and many of them can have pathogenic effects, it is therefore useful to be able to correctly and effectively predict their impact on a given protein. Proteins interacting with nucleic acids are essential for most cellular processes. Progress in computational methods and machine learning enables the development of increasingly high-quality tools for predicting the effect of mutations on proteins. An important category of these predictors are tools for predicting the effect of single point mutations on protein interactions with nucleic acids. This thesis focuses on currently available tools for prediction of missense mutation effect on protein-NA interactions, which predict quantitative changes in protein and nucleic acid affinity and estimate the severity of the mutation based on this change. Design, preparation and structural studies of biologically relevant protein variants of cancer-related Carbonic Anhydrase IX

3 Abstract Carbonic anhydrase IX (CA IX) represents an attractive target for the development of anticancer drugs as it is overexpressed in various types of solid tumors. By its catalytic activity, CA IX assists the cancer cells to maintain the optimal intracellular pH and to acidify the extracellular milieu promoting tumor development. There are twelve enzymatically active carbonic anhydrases (CAs) present in human body, all sharing a high sequence identity and a typical β-sheet structural fold of the well-studied catalytic domain. Although, the activity of CAs is efficiently inhibited by sulfonamide-containing compounds, the design of inhibitor selective to the cancer-related CA IX has been hampered by the high sequence conservation. CA IX has several unique features compared to other members of the family, which investigation may help in the development of selective drug compounds. Namely, it is a type I transmembrane dimeric protein with unique N-terminal proteoglycan-like (PG) domain, extracellular catalytic domain, and short cytoplasmic C-terminal segment. Flat House Rozdrojovice

The subject of the bachelor's thesis is the development of project documentation for the new construction of an apartment building in Rozdrojovice. The apartment building is a detached building with nine residential units, four floors, basement. The main entrance to the building is from the south side of the building. There are nine apartment units on the upper floors, which are connected by a common main corridor with a staircase and an elevator. In the basement there are social rooms, cellar cubicles, a carriage house, a technical room and an air conditioning machine room. Each apartment on the second to fourth floor has a balcony or terrace. The structural system of the building is wall-built, transverse. The building is based on a reinforced concrete base plate. The vertical supporting structures are made of ceramic masonry. The insulation of the perimeter walls forming the envelope of the building is designed by a certified ETICS system. The ceilings are designed as monolithic reinforced concrete slabs woven around the perimeter. The roof is a flat single-layer vegetation. None of the housing units are designed as barrier-free for people with reduced mobility, but access to the building is barrier-free. If necessary, thanks to the spacious rooms and corridors in the ground-floor apartments, it is possible to change the disposition of one of the apartments to a barrier-free apartment. The material and color of the facade takes into account the design of the surrounding buildings. Each block is distinguished by a different facade color. Rainwater will be drained from the roof into the storage tank. Subsequently, it will be used in the internal separate rainwater system for flushing the toilet and watering the garden. Surplus water will be absorbed on the property into a seepage pit. The project also includes an energy label. The designed apartment building belongs to the classification class "A - extremely economical".

The theoretical section defines legal and analytical terms. In practice section, selection is made according to the fund's statute, and securities are assessed using analyses and methods. Stocks are verified using Altman's Z-score, and the results are utilized to propose portfolio expansion.

The subject of this bachelor thesis is to create project documentation for a new family house in the city of Brno in the municipal district of Dolní Heršpice The building is to be located in a developing area for the construction of family houses. The building is detached and should serve for living up to 6 persons. The building is designed as a two-storey building with a terrace and a gardent. Both floors are above the ground. The roofing is created by contact flat roof. The structural system of the house is a transverse wall system, made of ground ceramic bricks of HELUZ FAMILY 2IN1 type with integrated polystyrene foam. The building is based on the foundation strips of plain concrete. The ceiling structure is designed from HELUZ ceramic ceiling panels. The staircase connecting all floors is monolithic and the roofing is designed with a flat single-skin roof.

Milan Martínek, Ph.D. Title: A case study of physiotherapeutic treatment of a patient diagnosed with TEP of the knee joint Objectives: The aim of this bachelor thesis is to elaborate the theoretical background related to the diagnosis of TEP of the knee joint. The special part of the bachelor thesis is prepared in order to keep a case report of a patient after knee TEP implantation and to record the effects of selected physiotherapy care procedures. Methods: This bachelor thesis consists of theoretical and special part. The theoretical part deals with a literature search to understand the structure and function of the knee joint from the perspective of anatomy, kinesiology and biomechanics. The thesis also describes the problem of osteoarthritis and its therapeutic interventions, types of total knee arthroplasty and the surgical procedure for its implantation. The theoretical part in its conclusion focuses on the possibilities of solving the subsequent rehabilitation after knee joint TEP implantation. The special section includes the initial and outcome kinesiological examination, goals of therapeutic care and a thorough record of therapeutic units. Vertical green walls

ABSTRACT The concepts of vertical gardens, vertical walls, green walls, living walls or green faca-des can be referred to as vertical plantations for planting the walls or columns of various objects. They are defined under the name of vertically situated plant plantings, which, according to the used plant taxa, can be applied both indoors and outdoors. Contemporary architecture is increasingly looking for ways to return greenery to human settlements, and vertical gardens are one of the solutions that can respond to the lack of free space for greenery, especially when solving development within settlements and return at least part of nature to the interior of cities and municipalities. Last but not least, the system solutions of vertical gardens for the creation of vegetation vertical walls in the interior and on the facades of buildings offer the possibility of making vari-ous and architecturally interesting areas, permanently overgrown with vegetation. The dissertation deals with the topic of greening facades, especially using systems not associated with open ground - the so-called vertical gardens. Since the knowledge on this issue is still poorly documented and the implementation of green facades of the-se systems in our country is not much, based on these facts, I decided to create my own real model of the facade system of vertical vegetation walls in the climatic conditions of the Czech Republic with local taxa. plants. In the initial phase of the work, an analysis of the available green wall systems is performed and a planar panel of the vertical green wall is designed.
