Irony speaks, or how to depict an elf wearing a hat that makes him invisible. Irony in the first chapter of the first part of Kierkegaard's work The Concept of Irony with Constant Reference to Socrates

The presented diploma thesis deals with the text called The conception made possible, which forms the first chapter of the first part of Soren Kierkegaard's dissertation The Concept of Irony with Constant Reference to Socrates. The diploma thesis has two parts. The first part is a contribution to the scholarly discussion on irony in Kierkegaard. It is an ironic interpretation of the text in question and it illustrates and describes the way in which Kierkegaard represents irony in this text. By means of an analysis of the discrepancy between the declared aim of The conception made possible and the actual realization of this aim, the first part of the diploma thesis comes to the conclusion that even though the representation of irony is not declared to be the aim of the text it actually is its real aim. It is just by means of this analysis that it becomes clear how Kierkegaard represents irony in the text. At the same time, the first part of the thesis shows that in The conception made possible Kierkegaard uses the indirect communication. The second part of the diploma thesis is a contribution to the field of literature: it consists of a Czech translation of The conception made possible which illustrates the explanations of the first part.
Stability of vesicular complexes against changes in their concentration

This bachelor thesis is focused on the stability of vesicular systems that consist of ion pair amphiphile. These catanionic vesicles were prepared from single-chained surfactants CTAB cetyltrimethylammonium bromide) and SDS (sodium dodecyl sulphate), which were stabilized by adding double-chained surfactant DODAC (dimethyldioctadecylammonium chloride) with 43 mol.% cholesterol. The aim of the thesis was to verify the stability of vesicular systems against changes in their concentration. For the measurement, a concentration series was prepared by diluting vesicles with deionized water. The stability was evaluated by dynamic (DLS) and electrophoretic (ELS) light scattering measurements at weekly intervals for four weeks. By DLS method were determined the average values of the size of vesicles and their changes during the measurement, the method ELS provided the information about changes of charge with different concentration in time. Samples with a higher concentration, i.e. no dilution till tenfold dilution of the vesicle solution, were marked as stable, higher values of average sizes and lower values of zeta potential were measured at solution with lower concentrations, according to which such diluted vesicles can be described as unstable. There were also visual observations made, but no visible aggregation in the solutions was observed. Furthermore, the aggregation behavior of the vesicles was observed by fluorescence spectroscopy using pyrene as fluorescent probe.
pH sensitive fluorescence probes

The goal of this thesis was to suggest the suitable method for measuring bacterial cytosolic pH in bacteria strain Cupriavidus necator. Fluorescent microscopy was chosen to obtain scan of bacteria and time-resolved fluorescence was chosen to obtain a calibration curve. BCECF-AM was used as a pH-sensitive fluorescent probe. It was suggested that the external calibration is more suitable than internal one for the prokaryotic type of cells. Bacteria H16 shows a long fluorescence lifetime only on the granules containing PHB, while in PHB-4 the longest fluorescence lifetime occurs randomly throughout the cell.
Intrinsic fluorescence of bacteria Cupriavidus necator

This thesis focuses on autofluorescence of flavins in gram-negative bacteria Cupriavidus necator H16 and its mutant strain PHB-4. The main methods used were fluorescence microscopy and flow cytometry. To confirm the presence of flavins, excitation and emission spectra of the bacterial suspension were measured, which were compared with flavin standards. In the part of testing cells without stress response, the autofluorescence of bacteria in PBS buffer and cell suspensions stained with fluorescence probe BODIPY 493/503 was measured. The ratio of short fluorescence lifetime to long autofluorescence lifetime, and its dependence on fluorescence probe was compared with previous conditions. Autofluorescence of the supernatant was measured; it was found that the relative amplitude of long lifetime was multiple times higher than in the cell. In the part devoted to the stress response, this thesis was focused on the amount of dissolved oxygen in the production medium and the effect on bacterial autofluorescence. Then differently concentrated hydrogen peroxide was used, the best results were obtained from the concentration of 100 mM in media. For comparison a combination of hydrogen peroxide with ferro-ammonium sulphate was used, but there was no big difference. Sodium azide and antimycin A were selected as substances that directly influence on bacterial respiratory chain. Both compounds affected change in the ratio of the relative amplitudes, but the distribution of these lifetimes and the autofluorescence change over time was affected only by sodium azide.

The International Classification of Functioning, Disability and Health can provide a suitable framework for functional assessment, not only for occupational therapists. The main aim of this thesis is a determination of the functional picture of selected patients with central hemiparesis. Biological invasions of non-native species are one of the most serious threats negatively impairing global biodiversity. The negative influence of these invasions is especially noticeable in freshwater ecosystems also exemplified by the populations of native crayfish species in Europe. Dominance of non-native species of crayfish over native ones is relatively well known and studied, but the interactions between individual non-native invasive species are significantly less explored. As the range expansion and numbers of non-native crayfish species constantly increases, it becomes apparent that the non-native species will dominate in European waters. However, it is not clear which of these invasive species, and under which conditions will perform better. The goal of this bachelor thesis was to create a literature review focusing on factors that negatively influence the population of the native species (mainly the issue of non-native species and crayfish plague which these species often transmit). Consideration was paid to the importance of the water temperature for the crayfish and to the focused species used in the experimental part the marbled crayfish Procambarus virginalis Lyko, 2017 and signal crayfish Pacifastacus leniusculus Dana, 1852. The experiment was focused on the evaluation of survival, growth, damage to the claws, and the maturation of these species at the water temperature of ~16 °C, which is in the optimal range for signal crayfish, but suboptimal for marbled crayfish (yet, within a range presumably allowing its successful reproduction). If these species should meet more frequently in the wild, it is important to understand their relationships.
Creation and verification of methodical material using overball exercise aid for children of younger school age

The diploma thesis verifies the influence of compensatory exercise using a mini exercise ball (overball). It is focused on the muscular system of middle childhood (8-11). The diploma thesis consists of a theoretical and practical part. A collection of compensational exercises consisting of 30 parts was build based on results from the initial examination of tested participants. The examination of the muscular system was accomplished by several standardized tests (evaluation of body composition by Lomíček, Mathias test, Adams test, an examination of the muscular stereotypes. A compensatory exercise was done after the examination once a week for eight weeks as a group exercise under a professional supervisor. When the examination was over a final evaluation was done, which was identical to the initial testing. As a conclusion, the effect of compensatory exercise was completed. Based on the application of provided and verified methodological material a certain change was achieved. The best improvement was achieved during the examination of the muscular stereotypes. Three of the tested participants noticed an improvement, none of them noticed a worsening. The least improvement was recorded by Adams test, only one tested participant improved, others without any change. In some cases, there were individual changes. In particular areas, there were changes of individual character, nevertheless, an evaluation of the group was identical to the initial testing. A benefit of this thesis is to make use of a mini exercise ball (overball) to make the exercise and positive influence of the exercise more effective on the muscular system not only for middle childhood.

Development of graphomotor skills in preschool children

- anglicky Title: Development of graphomotorics in pre-school children The present diploma thesis is focused on the topic of development of graphomotorics in pre- school children. The theoretical part deals with the description of children in pre-school age and everything related to this stage of life: the curricular documents, the concept of school preparedness and readiness as well as its diagnostics. The second half of the theoretical part describes the area of graphomotoric skills - its development and stages it undergoes, children drawings and its development and finally an overview of possible roadblocks and dysfunctions in graphomotorics. The empirical presents the result of research conducted via questionnaire for parents of pre-school children. Its aim was to uncover how well the parents are informed about and how much they are interested in the development of graphomotorics of their pre- school children. Key words: Pre-school and primary school age child, school readiness, school preparedness, pre-school education, children drawing, graphomotoric dysfunction.
Parents returning to work after parental leave in the eyes of recruiters

Work, and more particularly paid work, is an essential part of decent human existence. Current conditions on the Czech labour market cannot be considered friendly towards parenthood and parenting duties. The common pressure to fully adjust to the employer's needs and very limited opportunities of family-friendly forms of employment together with other factors have led to a situation where parents of small children (and particularly mothers) find themselves in a disadvantageous position. Family policies do not aim to improve that position and institutional child care does not have sufficient capacity. The qualitative research undertaken for the purpose of this thesis therefore focuses on personal managers as representatives of employers in the employee recruitment process, more precisely on conceptions (stereotypes) personal managers have of men and women, of parents in general and most importantly of parents returning to the labour market after their parental leave. It is very important to research these conceptions (stereotypes) because they are an integral part of the employee selection process and are reflected in the decision-making about hiring/not hiring an individual for a position.
