Roles of cytoskeleton in mouse polyomavirus trafficking
Klímová, Lucie ; Forstová, Jitka (advisor) ; Šmahel, Michal (referee)
6 Roles of cytoskeleton in mouse polyomavirus trafficking ABSTRACT: Mouse polyomavirus (mPyV) is small non-enveloped DNA virus. Its endocytic pathway is studied for a potential utilisation of polyomaviral virus-like particles in gene therapy and/or immunotherapy. mPyV enter cells by internalisation into smooth monopinocytic vesicles. During it's journey through the cell, it pass through early endosomes, and at the time 3 hours post infection, it is localised in endoplasmic reticulum and recycling endosomes. Many aspects of mPyV trafficking and nuclear entry are not clear yet. Time-lapse live imaging fluorescence confocal microscopy was used to describe the mouse polyomavirus intracellular movements. For these studies, we utilised mPyV fluorophore-labeled virions and cells expressing GFP-tagged g-actin or alpha-tubulin. Some virion-loaded vesicles were seen to move with actin organised into dynamic structures. Some of these structures resembled actin comets created by Listeria or vaccinia virus. At the same time post infection (40-60 min post infection), movement of the virion loaded vesicles along mirotubules was observed suggesting the simultaneous involvement of actin and tubulin during mPyV trafficking. Dynamitin, a dominant negative inhibitor of dynein-dynactin function reduced mPyV infection. Taken...
Sports Associations and New Media: Comparison of the Vision and Reality of Communication on the Example of the Czech Athletic Federation (case study)
Klímová, Lucie ; Turková, Kateřina (advisor) ; Macková, Veronika (referee)
Nowadays, new media play a big role in the world and among other things, they also change the role of classical media. Their use also significantly affects daily life in society. The possibility of communication through these channels is currently used not only by publicly known personalities and commercial entities, but also, for example, by non-profit organizations or sports associations. Diploma thesis Sports associations and new media: comparison of the vision and the reality of communication on the example of the Czech Athletic Federation (ČAS) is a case study, which examines the using of new media for the effective communication of the association with fans and journalists. The aim of this thesis is to describe the functioning of ČAS on social networks, namely on Facebook, Instagram and Twitter, and then to compare the actual state of communication with the ideas and visions of the employees of the association. First, the most important terms used in the thesis are described. The theoretical part defines new media and describes in detail the representatives of new media, which is further researched, namely Facebook, Instagram and Twitter. Then the Czech Athletic Association and social networks are introduced. The second chapter defines the research methods that were used. It describes how the...
Nursing severity in communication with patient´s family.
KLÍMOVÁ, Lucie
The aim of this bachelor thesis is to map the difficulty of communication between a nurse and a patient 's family on a standard and Acute Medical Unit. To this end, two research questions were set up: 1. "What difficult situations do nurses encounter in communicating with their family in a standard inpatient ward?" 2. "What difficult situations do nurses encounter in communicating with their family to acute medical unit? " The practical part is processed in the form of a qualitative research using the technique of semi-structured interview. The practical part is processed in the form of a qualitative research investigation using the technique of semi-structured interview. The interviews were based on pre-prepared questions with two groups of nurses. The research sample consisted of six nurses working in the standard inpatient department and six nurses working in the acute care beds. Interviews were recorded via audio recording and subsequently rewritten. Furthermore, the results of the research were processed by the method of coding using the technique of pencil-paper. The results of the interviews are presented through five categories, divided into subcategories. The trust of the patient's family in hospital staff is key to quality cooperation, as evidenced by research. This concerns both the acute care department and the standard care department, and the results of the research show that the involvement of the hospitalized patient's family is more demanding in the acute care department because the patient's family members are more likely to interfere with the established ward regime. Conflict situations arise in both departments for analogous reasons. Disputes arise mainly from lack of information, poor communication or fear for the safety of a close person. Confrontations are also associated with unpleasant emotions such as helplessness and stress, as confirmed by nursing staff from both of these inpatient departments. The bachelor thesis was written in such a way that it would be possible to look at the issue of the difficulty of communication between the nurse and the patient's family. The results of the bachelor thesis can serve nurses for whom they can guide the effective communication.
