Novel Reactive Groups for Modification DNA by Oxoosmium Complexes

DNA is naturally electroactive moleclue producing several intrinsie voltammetric signals. Some of them found appliction in electrochemical analysis of DNA interactions and damage. In particular types of applictaions is a suitable use DNA labelling by redox active tags to increase selectivity and sensitivity of analysis.One type of long-term used tags are complexes of osmium tetroxide with nittrogen ligunds (Os,L). These complexes preferentially react with pyrimidines in single stand DNA producing electractive adducts. primary reaction site for Os,L is C-C double bond in pyrimidine nucleobases. In this contribution will be presented results acquired with DNA containing chemically modified nuclebase.
Reduction of the DNA at a Pyrolytic Graphite Electrode:Opening of the Second Half of the Potential Window

The pyrolytic graphite electrode (PGE) can be used at highly negative potentials, providing reduction signals known from mercury-based electrodes. Use of the negative part of the potential window more than doubled the number of analytically useful signals provided by unlabelled DNA on PGE. It enables selective analysis of all canonical DNA bases,5-methylcytosine and uracil It also allows for distinction of overlapping signals cathodic polarization can selectively ´switch off´ DNA anodic signals and rice versa. Newly observed signals corresponding to oxidation of reduced bases and reduction of oxidized bases are also described.
Study of Synthetic Oligonacleotides Containing Guanine by Voltammetry on a Graphite Electrode

Electrochemical oxidation of specifically designed DNA hexamers containing guanine has been studied in aqueous acetate buffet (pH 5) at a pyrolytic graphite electrode (PGE) by linear sweep voltammetry (LSV) Influence of the oligonucleotide composition and concentration was investigated It seems that short oligonucleotides containing guanine produce oxidation signals. which are sensitive to their structural properties Simple electrochemical techniques us may be useful tool for monitoring of structural changes of DNA.
Electrochemical Study of Osmium Tetroxide Comptests Reactivity to DNA Bearing Butylacrylate

Coniplexes of osmium tetroside with nitrogen ligands (Os,L e.g.. with 2,2'-bipyridine (bpy) find application in redox labelling of DNA.. probing of DNA structure, and in studies of DNA interaction with other molecules Os,L preferentially react with pyrimidines in single strand DNA producing electroactive adducts Primary reaction site for Os,L is C=C double bond in pyrimidine nucleobases. In this contribution we introduce a new two-step technique of DNA modification with Os,bpy, consisting in enzymatic construdion or DNA beating butyl acrylate (BA) moieties attached to uracil or T-deaza adenine, followed by chemical modification of a reactive C=C double bond in BA residue.
Safety inspections in region Vysocina

This thesis presents a safety inspection in selected places in the Vysocina region of the Czech Republic. Six intersections in rural and municipal areas, plus three rural road sections were selected for the inspection and an accident rate analysis. The places selected were compared in terms of the accident rate and one case of each group was then selected for a detailed safety inspection encompassing visual inspection and identification of possible hazards. By eliminating such safety hazards, it is possible to ensure a traffic fluency and to preclude traffic accidents or at least minimize the harm caused by it.
Bass string instruments used in church and folk music in Prague and central Bohemia

This thesis is aiming to summarize and compare instruments used in church and folk music in Prague and central Bohemia. At first it names various types of instruments and describes their characteristics. Then pick few makers who influenced development and expansion of double bass in the land, and following with pick of particular instruments and their description. Last part of this thesis speaks of Ondrej Hornik and his collection, which is valuable source of information and tangible documentation of musical development in Czechia. Included is an insert located at the end of this work.

Integration of the Corona renderer into the ArchiCAD software Bachelor thesis - Jan Špaček, 2018 We present a prototype of a plug-in that integrates Corona Renderer into Archi- CAD. We strive to make Corona in ArchiCAD as easy to use as in other graphics applications, so the user interface is a major concern in our plug-in. Special focus was given to interactive rendering, which has proved to be a technical challenge in other host applications. We give a brief overview of the APIs that we use and describe the architecture and implementation of the plug-in. We also present the performance of interactive rendering on various scenes. 1
Audio D/A converter with external control using microcontroller

This diploma thesis deals with the design of a laboratory measurement tool with ADC converter for audio and low frequency signals. In the introduction part a theoretical background of the ADC and DAC conversions are outlined. In the next chapters the technical solution of the laboratory converter is presented, including DAC with integrated circuit AD1852 and completed with digital audio receiver CS8416. This circuit secures compatibility with external audio devices with digital interfaces, like SPDIF or AES3. The digital receiver and audio DAC are added with serial communication interface for setup of the most audio and input parameters. This setup is operated by external microcontroller ATmega32 and original firmware. The thesis contains block diagram and schematic layout of the laboratory converter, including complete technical documentation for the construction and design.
Design of urban area using advanced technologies

This thesis focuses on the design of urban areas in regards of the air flow and the creation of Urban Heat Islands. It reviews basic aspects influencing the design of streets. Every building, tree, or obstacle obstructs the air flow and creates the wind shadow, where the air pollution increases. It also creates areas of higher wind speed, in which public comfort is reduced. By understanding the importance of this process and by implementing it into the urban planning process, using advanced technology we can create healthy urban environment.
Outreach work with drug users - foreigners

The diploma thesis is aimed to improvement work of outreach drug workers who works with drug addicted people from foregin countries in Prague. The theoretical part od diploma thesis briefly describes legislative framework linked with use of illicted drugs in the Czech Republic, Drug Strategy of the Czech Republic and definiton of therm street work in the Social Services Act. Next part of the thesis focused to information from lately situation on the open drug market in Prague a nd describes period, since when has been in touch outreach workers and drug addicted people from foregin countries. Thesis further comparing approaches to drug policy in countries of orign drug of drug addicted foreginers at Prague. As well deliberately select in each country important topics for outreach work. The methods used to obtain the necessary information included document analysis, observation, structured interviews and focus group. Information was collected to help in face to face work with drug users. To better understand the environment from which drug users - foreigners come from. Which services can they know from their home country, what services drug users - foreigners can use in the Czech Republic and how to work better with this target group.
