Role of tumour suppressor PML in nucleolar functions

The cell nucleus is a complex structure composed of different parts, the nucleolus and PML nuclear bodies are important compartments of the nucleus. In the nucleolus, transcription of ribosomal DNA and biogenesis of ribosomes take place. The nucleolus may regulate the expression of proteins and thus the subsequent cell growth through regulating the amount of ribosomes. The nucleolus is also a sensor of stress. PML nuclear bodies play an important role in many cellular process - response to stress, virus infection or DNA damage. PML nuclear bodies consist of many proteins, the major protein is PML protein (Promyelotic leukemia protein). PML protein is coded by PML gene, it is spliced postrancriptionally and it has several isoforms. PML protein is an important cellular regulator and also a tumor suppressor. The nucleolus and PML protein cooperate together and have a functional relationship, which is not entirely clear. It was shown that PML protein changes its localization after exposure to stress and it goes near the nucleolus or into the nucleolus and this happens mainly in primary cells (the reason can be that the level of PML protein downregulates in tumour cells). A quality management system (QMS) forms a part of the organization's management. There are various QMS systems. Usually, they are based on one of two approaches to quality issues - the normologic approach and the teleologic approach. Quality management is a dynamic process that constantly changes and develops in terms of forms, methods and content. Specific QMS types can be attributed to one of three basic concepts: industrial standards, TQM (Total Quality Management), and the ISO concept. Various approaches to quality management began to develop in the 1920s. After World War II, the issues of quality gain priority particularly in Japan and the USA, followed by Europe. At that time the first true quality management systems were established. Their significance and development rapidly grew mainly in the 1980s, when quality began to be considered a key prerequisite to success and future competitiveness. Presently, two systems are primarily used in Europe: quality management based on the ISO (International Organization for Standardization) 9000:2000 standards, and a quality model based on the TQM philosophy and principles, referred to as the EFQM Excellence Model, which has been designed and implemented by the European Foundations for Quality Management (EFQM). Direct democracy at the municipal level in the Czech Republic

The objective of the submitted diploma thesis is to analyze the subject of a direct democracy system in the Czech Republic focusing on the local level of governance. The possibility of being able to participate imminently in the municipal government and thus amending the current representative model became the subject of not only political discussions over the last few years. The thesis is composed of a foreword and five chapters, which are hereafter divided to subchapters for better transparency. Findings and author suggestions concerning the possible future forms of the local democracy are summarized in the conclusion. The introductory chapter presents the direct democracy in a theoretical way and shows how it differs from the representative democracy model. It also contains the definition of the five most respected forms of a direct democracy, which are: referendum, plebiscite, citizen initiative, petition and recall. Following two chapters are focused on the situation in Czech Republic, first in general, then with an emphasis on the municipal scene. Efforts to establish an individual form of a direct democracy can be observed already from the year 1918 with various results, new possibilities appeared later as Czech Republic had entered supranational communities. Environments and projects that support children's reading

The thesis deals with a support of children's reading. The work is aimed to factors and actors influencing reading education. The first part solves theoretical definition and the meaning of reading. Primarily there are explained two important terms related to the topic - reading and a reader. Further it is proceeding by attention given to different phases of reading education before and during school attendance. Primarily three environments influencing reading literacy were observed. These include family, school and public library. Furthermore, national representative projects supporting reading primarily in libraries are shortly presented at this thesis as well. The second part is focused on the Municipal Library of Prague, specifically Smichov branch and its activities. In this part are charted various programmes for children. Repeated activities for parents with children or for children alone are observed. Also, events for groups of preschool and school children are described. The important part of presented work deals with survey of cooperation between Smichov branch and local schools. The research results are to determine practical experience of teachers from these schools in activities of Smichov branch and in their cooperation with this library.

Formation of splicing machinery in the context of the cell nucleus

Most of the protein coding genes of higher eukaryotes contain introns which have to be removed from primary transcripts to make mRNA which can be used as a template for protein synthesis. This crucial step in the pre-mRNA processing is carried out by the spliceosome, a complex ribonucleoprotein machine formed from small ribonucleoprotein particles (snRNPs). snRNPs biogenesis is a complex process composed of several steps which take place in both the cytoplasm and the nucleus. Spliceosome assembly is highly dynamic and tightly regulated and pre-mRNA splicing depends not only on the sequence of the pre-mRNA itself but also on the nuclear context, such as the chromatin modifications. How do cells regulate where and when the spliceosome would be assembled? What determines which introns will be spliced? These are fundamental, yet unanswered, biological questions. In this work we analyzed the formation of splicing machinery in the context of the cell nucleus from several different points of view. First, we investigated the unexpected connection between splicing factor U1-70K and the survival of motor neurons (SMN) complex which is a major player in the snRNP biogenesis pathway.
