The effect of zanubrutinib on tumor cell resistance to daunorubicin due to carbonyl reducing enzymes.
Havlíčková, Lucie ; Wsól, Vladimír (advisor) ; Matoušková, Petra (referee)
5 Abstract Charles University Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Lucie Havlíčková Supervisor: prof. Ing. Vladimír Wsól, Ph.D. Title of diploma thesis: The effect of zanubrutinib on tumor cell resistance to daunorubicin due to carbonyl reducing enzymes Anthracycline antibiotics (ANT) are considered the first-line medication for oncologic diseases, including acute myeloid leukemia. Daunorubicin (DAUN) with a carbonyl group in position 13 is often used in chemotherapeutic treatments. Nicotinamidadenindinucleotid-phosphate dependent carbonyl reducing enzymes (CRE) create its hydroxyl metabolite daunorubicinol (DAUN-ol), which demonstrates higher toxicity on myocardial tissue and lower cytotoxicity on cancer cells, which acquire tolerance towards its effects. Overexpression of enzymes catalyzing the conversion of effective antineoplastic DAUN causes the gradual development of resistance to administered ANT. For this reason, the new pharmacotherapeutic options for preventing this process are being searched for to maintain the drug in the target cell as long as possible. One option is to use inhibitors, which lower enzymatic activity CRE and prevent unfavorable DAUN metabolism on cardiotoxic and ineffective DAUN-ol. This diploma thesis aimed to study the...
Physiotherapy of functional disorders of the orofacial area in wind instrument players
Havlíčková, Lucie ; Maršáková, Kateřina (advisor) ; Malá, Jitka (referee)
Title: Physiotherapy of functional disorders of the orofacial area in wind instrument players Objective: The areas of interest of this diploma thesis are temporomandibular joint (TMJ), wind instrument playing and health problems affecting the orofacial area related to playing of the wind instruments. The aim of this diploma thesis was to determine the current functional state of the orofacial area in wind instrument players and to evaluate the effectiveness of the chosen type of physical therapy. Methods: This experiment-centered pilot study included a total of 21 wind instrument players aged 15-30. After taking anamnestic data, the subjects underwent kinesiological examination focused on the orofacial area. To evaluate the results, a visual analog pain scale (VAS) and examination of the active range of motion of TMJ using a millimeter ruler were used. If the proband experienced any TMJ pain, had a reduced active range of TMJ, or both of the previous, the patient was prescribed an one month autotherapy targeted on the orofacial area. Once the therapy was finished, the probands underwent the second orofacial examination. Horn's procedure, Wilcoxon's test and Microsoft Excel 2021, were used to analyze the collected data. Results: During the initial kinesiological examination, 38 % of the woodwind...

Neighbours' rights
Havlíčková, Lucie ; Švestka, Jiří (advisor) ; Salač, Josef (referee)
Sousedská práva 80 Resumé Legal relations between neighbours and their protection The legal relations between neighbours and outgoing rights and obligations, are the important part of human relations, which are existing between men from the beginning of evolution of the human society, as well as legal relations between neighbours are the relevant part of the legal orders of each individual state. In the Czech Republic nowadays, the legal relations between neighbours are stated in the act No. 40/1964 Coll., the Civil Code and it has the unfungible place in our legal order. The legal relations between neighbours belong into the area of the jura in re and they are connected with the problems of the property right. They represent one of the areas of the legal restrictions of propriety, which are necessary to respect in the interest of the same rights of the neighbouring proprietor. If the proprietor wants to exercise his powers, he has to suffer some of the restrictions of his powers, for the benefit of the same powers of the neighbouring proprietor, in the legal frame. The property right represents one of the most basic rights in the full scope of the legal systems, which is declared by the rules with the supreme legal force. My dissertation is systematically divided into the eight main sections. Leaving aside...
Return of patients with the diagnosis of schizophrenia to society in relation to greality of life
Havlíčková, Lucie ; Vachková, Eva (advisor) ; Šlaisová, Ivana (referee)
Author: Lucie Havlíčková Institution: Charles University in Prague, Medical faculty of Hradec Králové, Institute of social medicine, Department of nursing Title of thesis: Return of patiens diagnosed with schizophrenia in the company in connection with the quality of Life Consultant: Mgr. Eva Vachková Number of pages: 105 Number of appendices: 5 Year of defense: 2012 Key words: schizophrenia, depression, quality of Life, SQUALA, BDI-II The main aim of my bachelor's project is to target on the issue of subjective perception of patients' life quality in case of schizophrenia and their participating in social life. Theoretical part deals with schizophrenia as a disease itself, its epidemiology, clinical screen, classification and form of illness, course with its typical symptoms and the cure of course. The next important point is the quality of life in general and possible ways of its measuring. The goal of practical part was to find out about subjective life experience of schizophrenic patients on different levels, its comparing with common population. I used the quantitative research with standard questionnaires SQUALA and BDI- II. The group of respondents included those with schizophrenia diagnoses hospitalized in Mental hospital in Havlíčkův Brod, out-patients and the data were compared with...
Application of analytical pyrolysis for the detection of plastics and microplastics in the environment
Havlíčková, Lucie ; Cajthaml, Tomáš (advisor) ; Innemanová, Petra (referee)
The issue of microplastics is becoming increasingly important every year and so is the need for finding solutions. Plastic fragments smaller than 5 mm have already been found all over the planet in all spheres of the environment. Among the crucial factors of resolving this issue are, for example, restrictions in the production and overuse of plastic materials, unification and improvement of the waste management system, prevention of unintended release of plastic waste into the environment, the study of possible toxic effects of microplastics on living organisms, and the development of separation and detection techniques for the precise detection and identification of microplastics and nanoplastics. In recent years, the combination of pyrolysis, gas chromatography, and mass spectrometry (Pyr-GC-MS) has proven to be a potentially suitable instrument for the qualitative and quantitative analysis of these plastic particles. This Master's thesis proves that the Pyr-GC-MS method can, above all, be sensitive and accurate enough for the detection of microplastics in real environmental samples, where the particles are present in very low concentrations. Due to the broad diversity of physical and chemical properties of polymers, it is highly difficult to develop a single analytical method for the whole range...

Detection of microplastics in the environment
Havlíčková, Lucie ; Cajthaml, Tomáš (advisor) ; Innemanová, Petra (referee)
This bachelor thesis is focused on microplastics as small fragments of plastics which represent a burden for the environment and can be probably dangerous for living organisms including mankind. Serious health consequences caused by a long term exposition of the microplastics to organisms have been demonstrated in the recent years. It has been proven that the level of harm increases with ability to adsorb toxic substances on their surface. Primarily, it is necessary to eliminate entrance of these microparticles into the ecosystem and therefore this thesis describes routes of microplastic inputs into the environment. The microplastics occur in all levels of the environment, that represent various environmental matrices with different conditions, that must be considered for successful separation and subsequent analysis of samples. This thesis provides summarization of currently used analytical methods including their advantages and comparisons. The most frequently used techniques of separation are based on the principle of density flotation. Further identification of fragments is performed using microscopy, spectroscopy or gas chromatography. This work describes principles of the individual techniques of detection including also particular cases of analyses. The subject of this thesis is also the...
