Testing of biological effect of glucans on cell lines in vitro

The aim of this bachelor thesis is isolation and testing of -glucans from selected strains of yeast. In the theoretical part, characterisation of -glucans, microbial production of -glucans by yeast, determination of polysaccharides, exopolysaccharides and in vitro testing are described. Yeast have a cell wall made up of -glucans and -glucans are known for their healing properties. In the experimental part of this work deals with characterization of selected biomass yeasts in different forms and comparison of their effects and the amount of -glucans in each form. Stains of Metschnikowia pulcherrima, Cystofilobasidium infirmominiatum and Rhodotorula kratochvilovae have been selected for this thesis. Subsequently, the thesis deals with the isolation and testing of isolated -glucans from a selected lyophilised strain of yeast Rhodotorula kratochvilovae. The selected strain was isolated in 2 ways. The first method of isolation that was more friendly to conformation of -glucans due to the effect of ultrasonic homogenization of cells. The second method of isolation was less friendly to -glucans, using acid and base to homogenise them. Analyses of both extraction methods confirmed the presence of isolated -glucans, however it was found that the recovery of both methods was relatively low and requires further optimisation of the procedure. Overall, the better extraction procedure was the second method. A sample of this extract was further tested on the HaCaT cell line. The results of the pilot tests confirmed the cytotoxicity of the extract obtained by us.
Optimization of methods for the isolation and analysis of beta-glucans produced by carotenogenic yeasts

The aim of this thesis was to optimize the methods of isolation and analysis of -glucan polysaccharides produced by carotenogenic yeasts. As part of the experiment, the yeast Rhodotorula toruloides, which was cultivated in laboratory bioreactors, was selected as the production strain. Theoretical part describes the subject of carotenogenic yeasts and the metabolites they produce. The main metabolite that this work deals with is the polysaccharide -glucan. -glucans are polysaccharides that form the cell wall in the yeast cell. They are known for their immunostimulating effects, antioxidant effects, promote wound healing and help lower cholesterol and blood glucose levels. The experimental part of the work deals with the cultivation of yeast biomass and its characterization. Furthermore, optimization of the enzymatic determination of -glucans using a commercial enzymatic kit and isolation of -glucans from yeast biomass. As part of the experiment, yeast biomass extraction methods were also further developed, enabling the separation and analysis of -glucans, lipids, lipidic substances and proteins from a small sample of yeast biomass in one extraction procedure.

Role of rags and other community events on life of czech woman in 19th century

The socio-historical and literary-historical context is delineated in the first part of the thesis. But the essence of the thesis is analysing of three novels and one book of advices written by four female writers of the second half of 19th centrury. The analysis is focused on construction of model female readers for which the books were written. The main theme is analysis of detail of ball or dance event in their influence on the life of teenage girls, the definition and description of how the marriage market works and its changing relationship with the woman's free choice of profession. Based on readings of other texts of the four female authors we followed the extent of projection of ideological orientations and attitudes of the writers into these their analyzed texts.
An analysis of tourism development in Pardubický Region and its prospects

The bachelor thesis deals with the analysis of the development of tourism in the Pardubice Region from 2004 to 2014 using statistical methods. Analyzes traffic in collective accommodation establishments, development of capacities in collective accommodation establishments, number of nights spent in collective accommodation establishments. Further statistical methods evaluated traffic analysis of selected tourist destinations: Litomyšl Chateau, State Kunětická Castle Mountain, State chateau Slatiňany, Litice castle). In this work are formulated proposals and recommendations for the promotion of tourism in the Pardubice region. The data were drawn from the Czech Statistical Office and the National Information and Consulting Centre for Culture.
Evaluation of Potential and Prerequisites of Tourism for Blansky les Netolicko Region

Working analytically evaluates the potential of tourism and assumptions for the region's tourism Blanský les - Netolicko. Based on theoretical knowledge of the potential, assumptions, their own evaluation and investigation, I tried to design appropriate activities based on the assumptions and the potential for tourism. Conclusions arising from work can function as a basis for management decisions MAS Blanský les - Netolicko.
Analysis of Tourism Development in Klasterec nad Ohri

This bachelor thesis deals with tourism and its further development in Klasterec nad Ohri. In the theoretical part is described tourism and the most important knowledge concerning tourism. There are also introduced analytical tools which were used in the practical part of the thesis. This part analyzes conditions and assumptions for tourism allocation and development in the city and also, with the help of conducted survey, evaluates a satisfactory rate of visitors and inhabitants of the city with the fulfillment of these conditions and assumptions. The mentioned analyses are the fundamentals of the SWOT analysis of tourism in Klasterec nad Ohri which is the source for the final strategy draft. The suggested strategy should lead to the development of tourism in the city.
