Isotope hydrogeology and geothermal applications to clarify the origin, the sustainability and the character of groundwater flow: Bohemian and Aquitaine sedimentary basins

Isotopic investigations combined with geothermal applications represent powerful tools for the exploration of groundwater potential as a drinking or geothermal resource. This Ph.D. Thesis combines both approaches, environmental and radioactive isotopes together with temperature data in deep aquifers, in order to enrich and update the knowledge concerning the aquifer recharge processes in the Aquitaine Basin (France) and the aquifer recharge processes and geothermal potential in the Bohemian Cretaceous Basin (Czech Republic). Stable isotopes (18 O, 2 H, 13 C) combined with radioisotope data (14 C, 3 H) are used to estimate the recharge timing and climatic conditions prevailing during the infiltration from the Late Pleistocene up to modern time. The character of groundwater recharge and regime are necessary to generate relevant source data for the accurate modelling of complex groundwater systems. Three groups of groundwater recharge types can be distinguished throughout Europe - (i) continuous recharge and (ii) interrupted recharge during Last Glacial Maximum and (iii) a group corresponding to particular recharge conditions. The brand perception of Fairtrade at the Faculty of Economics, University of South Bohemia

The main objective of this bachelor thesis is to analyse how people at the Faculty of Economics perceive the Fairtrade brand. It means, how deep is their knowledge of this brand, what products consumers buy most, why they buy them and how they are satisfied with the quality and price of these products. The work is divided into two patrs: theoretical and practical. The theoretical part is mainly focused on basic information for understanding the essence of the bachelor thesis. The practical part is based on a questionnaire survey between people at the Faculty of Economics. The work concludes with suggestions which improve the current situation of the Fairtrade brand perception at the Faculty of Economics at the University of South Bohemia.

The role of nurses in the strabological clinic

The eyesight is one of the most important senses. The most common sight impairments among children are strabismus and amblyopia. It affects up to 7 % of children population. The children with eyesight impairment are treated in strabological outpatient departments. The Orthoptics deals with prevention and diagnosis of visual impairments and subsequent treatment of amblyopia and squinting.There are two goals in this work: to determine specific demands concerning the work of nurse in strabological outpatient department and to find out about their role from the parents point of view.The practical part was conducted using qualitative research survey. The individual semi-structural interviews were used to gather the data. The interviews were conducted with the orthoptist nurses working in strabological outpatient departments in the South Bohemia Region and with the parents of the treated clients.The research survey showed that the work of orthoptist nurse is highly specific. It requires deep knowledge of the field. There are high demands on their practical skills. Orthoptist nurse organizes functioning of the department and the training room, assists the doctor, prepares clients for the examination and conducts many examinations on their own. Handles ortoptic and pleoptic devices and manages their maintenance. Creates their own aids. Communicates with the patients of all age groups. They are supposed to have enough empathy, patience and creativity. They provide intensive educational activity, because the contribution of training at home is significant in treating amblyopia and squinting. The parents of the patients expect improvement of the impairment, kind and patient approach, and last but not least providing information, advices and solving all kind of problems.

Deep vein thrombosis and role of selected mutations for the coagulation factors genes

PharmDr. Tomáš Šimůnek, Ph.D. Title of diploma thesis: Deep vein thrombosis and role of selected mutations for the coagulation factors genes. This thesis is focused on mutations for the coagulation factors genes that can lead to deep vein thrombosis. In the first part, we describe the role of hereditary thrombophilia in the development of disease. Following is the description of each thrombophilic mutations and their meaning which is not always clear. The following mutations were detected: Factor V Leiden (G1691A), factor V R2 (H1299R), factor II prothrombin (G20210A), MTHFR (C677T, A1298C), factor XIII (V34L), PAI-1 (4G/5G) a EPCR (A4600G - haplotype A3, G4678C - haplotype A1). Most of them have been previously identified as risk factors for deep vein thrombosis, but specifically the factor XIII V34L polymorphism is considered to a protective factor. Other mutations with protective effect could be one of the genotypes of the PAI-1 and EPCR A1 haplotype. In the practical part, the samples and the results of 783 subjects were used. Crustal Structure of the Bohemian Massif Based on Seismic Refraction Data

The deep structure of the Bohemian Massif, the largest stable outcrop of the Variscan rocks in central Europe, was studied using the data of the international seismic refraction and wide- angle reflection experiments CELEBRATION 2000, ALP 2002 and SUDETES 2003. The data were interpreted by seismic tomographic inversion and by 2-D trial-and-error forward modelling of the P and S waves. Above, additional constraints on the crustal structure were imposed by reflectivity or gravity modelling, and by receiver function interpretation. Knowledge of the crustal velocity structure in the Bohemian Massif was complemented by its azimuthal variation. Though consolidated, the Bohemian Massif can be subdivided into several tectonic units separated by faults, shear zones, or thrusts reflecting varying influence of the crust forming processes. The resultant velocity models determined different types of the crust-mantle transition reflecting variable crustal thickness and delimiting contacts of these tectonic units at depth.

Závěry řešení: 1. Byla vyhodnocena úroveň modelové pražské populace (městských strážníků) karcinogenním polycyklickým aromatickým uhlovodíkům, která je závislá na sezónních vlivech se zvýšenou expozicí v průběhu zimních měsíců. 2. Expozice karcinogenním PAU je nutno považovat za zvýšené genotoxické riziko, jak bylo prokázáno zvýšenou úrovní DNA aduktů, chromosomových aberací konvenční cytrogenetickou metodou, metodou FISH a oxidací proteinů. 3. Na základě provedené studie pro potřeby systematického biomonitoringu genotoxických účinků komplexních směsí znečišťujících atmosféru považujeme jako vhodné biomarkery stanovení DNA aduktů metodu 32P-postlabeling, využití cytogenetické analýzy konvenční metodou, fluorescence in situ hybridizace s pomocí barvicích sond (FISH), hodnocení oxidačního poškození proteinů a hodnocení rozdílné vnímavosti jedinců s rozdílnou distribucí genotypů (genetický polymorfismus) i hladin vitaminů a lipidů. 4. Studium geneticky podmíněné vnímavosti k účinkům PAU naznačuje význam kombinance metabolických genotypů CY1A1-Msp I s GSTM1 a CYP1A1-Ile/Val s GSTM1. 5. Studium vlivů hladin vitaminů na rozsah genotoxických účinků PAU je ve stadiu hodnocení. Zatím byl prokázán pozitivní vliv vitaminu A na DNA adukty. 6. Při hodnocení podílu oxidativního poškození na indukci genotoxických změn bylo zjištěno, že kometový test není vhodnou metodou pro podobný typ studií. Za vhodnější je považována metoda stanovení oxidativního poškození proteinů. 7. Pro hodnocení zdravotních rizik je opakovaně prokazována nutnost monitorování karcinogenních PAU a využití těchto poznatků pro hodnocení rizika jak pro těhotné ženy a děti, tak i jedince pracující v prostředí se zvýšenou expozicí (jako např. městští strážníci v Praze). 8. V okrese Teplice se pokračovalo ve sledování průběhu a výsledků těhotenství ve vztahu ke kvalitě ovzduší. Získávané poznatky jsou dnes mezinárodně využívány i v materiálech WHO. 9. Byl navržen postup hodnocení genotoxicity komplexních směsí organických látek z ovzduší in vitro.

Submicron Structures with Deep Relief — Technology of Preparation

The dissertation thesis is focused on research and development in the field of microfabrication by the technology of electron beam lithography. In the first part of this work, the extensive study is conducted in the field of technology of electron beam lithography in terms of physical principles, writing strategies and resist materials. This is followed with description of physical principles of etching for the transfer of relief structures into substrates. The thesis describes innovative techniques in modelling, simulation, data preparation and optimization of manufacturing technology. It brings new possibilities to record deep binary or multilevel microstructures using electron beam lithography, plasma and reactive ion etching technology. Experience and knowledge in the large area of microlithography, plasma and anisotropic wet-etching of silicon have been capitalized to the design process of manufacturing of nano-patterned membranes. It was followed with practical verification and optimization of the microfabrication process.

Spinoza's Ethics Karel Kukla Spinoza's Ethics is practical instruction to a change of ethical thinking. It is a true reflexive Metod. We cannot think a compassion if we don't understand it. Author challenges us to the change of ethical thinking and refers to Spinoza's Ethics, which he introduces to a contemporary reader in an intuitive fashion. Summary of this ethical interpretation as follows: Everything is a part of our body, expressions are necessarily part of the whole. Atribute of thinking has essential role. It enables our intellect to comprehend these expressions and resign in a sense of active ability to reduce our lusts and affects. The third intuitive degree of comprehension directed to God comes out of honest resignation of intellectual comprehension. It comes out of honest self-understanding. Spinoza's Ethics is such a deep and breath-taking exploration of our human heart. It contains appealing ethical reference. It calls for unity and understanding. It leads people out of slavery. Out of thought captivity which is to think or believe we are free. Out of the real slavery he sets us free from the bond of uncontrollable lusts and confused ideas. The question is whether or not we are interested in it. Spinoza's Ethics means a practical life under way of eternity.

Novel approaches to diagnosis and treatment of selected endocrine disorders

Je známou zkušeností, že hyperkortizolismus urychluje aterosklerotické děje. Ty je možné sledovat ultrazvukovým vyšetřováním karotid a nověji též vyšetřováním cévní mikrocirkulace laser-Dopplerovskou metodou. Pro urychlení aterosklerotických změn u nemocných s Cushingovým syndromem jsou nesporně důležité známé faktory: hypertenze, obezita a hyperlipidemie. К nim se počítá zvýšená inzulínová rezistence, ale nemocní s floridním hyperkortizolismem mají normální hladiny glykemie a imunoreaktivního inzulínu. К aterogenezi u floridního hyperkortizolismu může přispívat zvýšená tvorba volných kyslíkových radikálů vznikajících při oxidaci lipidů a zvýšené hladiny pozitivního proteinu akutní fáze-orosomukoidu. Imunosupresivní charakter se projevuje snížením hladin autoprotilátek a snížením hladiny intercelulární adhezivní molekuly-1 (ICAM-1). К urychlení aterogeních dějů dochází i u nemocných, kteří jsou již dlouhou dobu po úspěšné normalizaci hyperkortizolemie. I když čerstvě operovaní a normalizovaní nemocní ztrácí do jednoho roku svou Cushingovskou obezitu, dochází později u operovaných nemocných opět к nadváze. Ta je spojena s poruchami lipidového spektra a u čtvrtiny nemocných i s hypertenzí. Nemocné po operaci pro Cushingův syndrom je nutné pravidelně sledovat, i když jsou dlouhodobě normalizováni. In this thesis, has been used less known technology of deep foundations. The author points to the diversity of utilization, such as sealing wall, anchored and unanchored sheeting, columns designed to influence tension and pressure. For the static calculation have been used programs Scia Engineer and Geo5 v19. Part of this thesis the technological process of technology of the soil improvement by DSM.
