Použití specifických aktinomycetových PCR primerů k charakterizaci bakteriálních společenstev exkrementů žížal

Tropical geophageous earthworms stimulate soil bacterial activity in soil that they ingest; it is uncertain if they affect the whole bacterial population or only subgroups. We sampled .i.Martiodrillus heterostichon, Polypheretima elongata./i. and bulk soil from a Bamboo plantation near Cali, Colombia. Pots of 1500 g of an adjacent tobacco field soil were brought to field capacity and two worms of each species placed in each pot. .i.Martiodrillus./i. gut contents and wormcasts from the soil surface and within tunnels were sampled after three months. The soil was maintained at field capacity for a further five months during which the worms continued to produce casts. Worm cast Amplified ribosomal DNA restriction analysis ARDRA (using Tag I) with actinomycete specific primers gave two strong bands at approximately 750 and 370 bp. ARDRA patterns from the three bamboo grove and four field soils had several additional bands notably at 140 and 126 bp.

J2EE design patterns

Práce si klade za cíl poskytnout ucelený pohled na problematiku J2EE návrhových vzorů zejména z pohledu praktického využití a jejich přínosu pro návrh a vývoj softwaru v prostředí Javy Enterprise Edition. V první části se věnuji seznámení se základními pojmy, na nichž celý dokument staví, což umožní čtenáři lépe nahlédnout do praktické části. Druhá část Vzory a J2EE zmiňuje celou řadu doporučení, které vykrystalizovali v průběhu několika let vývoje pod J2EE a dále zmiňuje nejrůznější strategie pro řešení celé řady problémů, s nimiž se softwaroví vývojáři denně setkávají. Dále následuje katalog 21 J2EE návrhových vzorů s jejich detailním popisem a možnostmi praktického využití. Klíčovou a nedílnou součástí této práce je ukázkový příklad, který ilustruje využití návrhových vzorů v reálném prostředí na reálném projektu. Toto propojení teorie s praxí ukazuje jak lze aplikovat návrhové vzory na skutečný problém. Zhodnocení přínosu vzorů pak uzavírá tuto práci a dává si za cíl objektivně zhodnotit světlé i stinné stránky dané problematiky.

Potential of brassinosteroids for forest seed management with regard to species utilizable as Christmas trees

The aim of this work is to assess the effect of brassinosteroids on germination of selected forest tree species with a potential to be used for the production of Christmas trees.The synthetically prepared analog 24-epibrassinolide (was tested on germination of seeds of Scots pine (Pinus sylvestris L.). Prior to launch of germination, the seeds were being soaked for 48 hours at three different concentrations of the tested brassinosteroid (BR-treatments) and in distilled water only (control), respectively. Seeds of each above-mentioned treatment were exposed to two alternative regimes: the stressed and unstressed one, respectively. In the stressed regime, seeds were exposed to a stress episode with temperatures up to 42 °C. In the unstressed regime the temperatures followed a pattern prescribed by Czech seed testing rules. Fully germinated seeds (the length of root radicle at least 4 times longer than the size of seed) were calculated. The root radicle length of each germinated seed was measured as well.The best results were recorded in the BR-treatment with the highest concentration of the tested brassinosteroid (0.4 mg.dm-3) in both germination regimes.
As contrasted to the control, the high concentration treatment increased germination capacity in the unstressed and stressed regime by 16.8% and 31%, respectively.

Design of log-periodic antenna

This diploma thesis deal with the design of log-periodic antenna. The log-periodic antenna is self-complementary and broadband antenna. Antenna self-complementary structures are described only by angles. Input impedance of log-periodic antenna is equal to theoretical impedance ohms. The input impedance and radiation patterns of broadband antenna structures are independent of frequency over theoretically unlimited bandwidths. The log-periodic antennas provide linearly polarized radiation pattern. In this thesis is designed antenna for bandwidths 1 to 5GHz. In this diploma thesis is introduced design balanced unit, this is compound broadband balun and broadband impedance transformer. The broadband balun is based on conversion of microstrip line to balanced stripline. The Klopfenstein impedance taper is used for design. For simulation was used software from firma Zeland software IE3D.

Design of Upper Limb Assistive Device

This work deals with a new approach of designing ortho-prosthetic devices in terms of mechanical solution and innovative design. The work was addressed to a specific patient with congenital malformation of upper limbs called phocomelia. A custom-made compensatory aid was made on basis of personal consultations and meetings on specialized workplace; however the aid was built in a way and from parts for other kind of disability. As it turned out, the use of this equipment is associated with many complications, and therefore as a result, this solution proved as unsatisfactory. Nevertheless, thanks to this compensation aid it was possible to specify the problems, to reveal important patterns and highlight the needs for development of entirely new and unique aid designed for patients with phocomelia disability. The work is based on these findings and knowledge acquired from bibliographic search in the field of orthotics, prosthetics, assistive devices and prosthetic equipment. A complete redesign was done with the aim to eliminate all detected problems. The presented result is a unique tool for people with specific congenital disability of upper limbs, with working label 4TE.

Real-time web applications

In my bachelor work I focus on two servers, which allow us to reach the real-time communication between the client and the server. My main goal is to familiarise PHP application programmers with the real-time communication problems on the internet, to point out differences from typical PHP applications programming and chiefly to focus on practical aspects of this issue. To achieve that, I have structured the dissertation work so that the minimum of the required theory would be explained at first and then I will focus on practical demonstrations, which will be properly commented. My main contribution to this issue is primarily a simple pattern for web pages running in Node.js environment that could be used as an initial point of any kind of web presentation.

Science & Technology Park in Zlín

The subject of this thesis is the project of scientific and technological park in the Tomas Bata University in Zlín. This park is divided into three workplaces: the educational and training center, a workplace for applied research, and business center. This project is situated at the location of a former factory area of Baťa Company; specifically in its eastern part. This dissertation is focused on the analysis of the given location, evaluates it in terms of its potential, and finally sets out the main priorities that should be applied in the draft. The accompanying report then describes and specifies the project, as drawn graphically, in the form of situational patterns, plans, sections, perspectives, and visualizations. Specifically, the project deals with the reconstruction of the former manufacture building No. 24, proposes a construction of a new building no. 25, and provides a solution for space in between of these two buildings. The project primarily seeks to make the entire site more attractive, and bring it to the function which would implement the functions in the assignment of the technology park. The function of the technology park is divided with regard to the investors requirements. From the urban point of view, the project seeks to approach the problem stemming out of the essence of the place. The use of objects remaining on the site preserved, is designed not to disturb the architectural expression and static essence. Architectural expression of new objects is selected not to oppose the overall architectural expression of the site. The project also includes and evaluates the usage of solar energy.

Facebook and Students of Erasmus Program: Change of Social Environment ant Its Impact on Facebook Usage Strategies, Overlap of Communities and Accumulation of Social Capital

The main objective of this thesis is to analyze, if change of social environment caused by participation of students on Erasmus program lead to the change of Facebook usage strategies. On the basis of these information, overlap of online and offline communities and accumulation of social capital is analyzed. Combination of qualitative content analysis and analysis of information extracted from graphical tasks is used in the paper. An innovative approach based on deductive derivation of categorization matrixes from analytical, quantitative tools is adopted in order to analyze qualitative data. Concerning the analysis, stress is put on particular patterns of change of Facebook usage strategies and subsequently, general model is created. On the basis of so created model, conclusions referring to direction of overlap of online and offline communities and nature of bridging and bonding social capital accumulation are drawn.
