Separace chirálních pesticidů pomocí cyklodextrinů kapilární elektroforézou

In this diploma thesis, a preliminary study on the applicability of EKC-PF (with CD as chiral selector) to the separation of chiral pesticides was performed. In introduction, theoretical informations are presented concerning electrophoresis instrumentation and theory as well as different electrophoresis modes. Then, enantiomeric separation is introduced through cyclodextrins and possibilities of partial filling mode to finally terminate with a definition of chiral pesticides and some information about their enantioseparation. Concretely, in the experimental part, the evaluation of enantioresolution of several chiral pesticides (trichlorphon, cyproconazole, hexaconazole, imazalil, myclobutanil, penconazole, tebuconazole, propiconazole, and benalaxyl) using CM-β-CD as chiral selector was performed. First of all, a univariate optimization of variables (CM-β-CD injection time, voltage, temperature, pH, and buffer nature and concentration) was carried out for benalaxyl. Finally a screening, involving the effect of the CM-β-CD injection time, was performed for the other pesticides to check the potential of this CD as chiral selector.

Chromatography method optimization for HPLC determination of carotenoids.

This thesis deals with searching for optimal conditions for determination of carotenoids lutein, betacarotene and zeaxanthin. High performance liquid chromatography - HPLC was used for determination. During searching for optimal conditions many various columns and mobile phases were tested. Primary information about carotenoids separation was obtained from scientific literature. There were tested both types of chromatography - normal phases chromatography and reversed phases chromatography. The most frequently tested columns were C18 columns from different producers. The biggest problem for HPLC determination and separation of these three substance were position isomers of lutein and zeaxanthin. Separation of these substances is relatively difficult and it wasn't completely solved in this thesis. In spite of it new metod for determination and separation of three carotenoids was developed. This method used isocratic elution at column Zorbax SB C18 (50 x 4,6 mm, 2 μm), UV detection at 450 nm, mobile phase of methylene chloride:hexan:acetonitrile (2,5:2,5:95), injected volume 1 μl and flow rate at 1 ml/min.

Development and Validation of Novel HPLC Methods for the Determination of Specific Analytes in Biological Material.

1. SUMMARY The presented dissertation thesis shows an interesting connection of two research fields that seem totally different from the first sight: analytical chemistry and medicine. However, the second sight could show that both branches might exist very closely to each other and their cooperation can be even useful for both sides. The initial part of the thesis deals with theoretical basics of HPLC and chromatographic instrumentation bringing a short overview of important principles of liquid chromatography including a brief introduction of crucial parts of an HPLC instrument. Due to the fact that the heart of the chromatographic system is an analytical column, another part of the text deals with different types of stationary phases and analytical sorbents. A separate chapter is focused on monolithic columns and other materials representing a new and original trend of chromatographic techniques. The main advantages and disadvantages of monolithic columns are then summarized in the published papers listed in the Experimental part (Enclosures 1, 2 and 9). Finally, the technical section of my thesis is concluded by a review of new trends in the chromatographic instrumentation presented e.g. by Ultra Performance Liquid Chromatography, columns filled with sub-2-micron particles or automatization or...

Planar brainteasers and their application to increase mathematical literacy of special educational needs children

Key words: Pentominoes Tangram Affine mapping Special educational needs pupils Orientation of flat Resume: My thesis deals with different ways of use of Pentominoes and Tangram brain-teasers in teaching of mathematics to special educational needs pupils. In the theoretical part I introduced these brain-teasers, I explained affine mapping using their particular pieces, I analysed similar tasks in mathematics textbooks for 6 to 10-year-olds and pointed out the issues of special educational needs pupils. In the practical part I described a few lessons of experimental education with six pupils of practical basic school. In the first year we dealt with Tangram brain-teaser. We made the separate pieces of paper, we handled wooden cubes and used Tangram pictures for development of visual perception and approximation of affine mapping. The next year the pupils schemed out their Pentominoes cubes first and then they worked with them. They drew them in a grid, built pictures, played a game for two etc. At the end I summarised my thesis and I evaluated it.

Attitude of Smokers Towards the Problem of Smoking

This thesis talks about the problems of dependence on nicotine and the problems on quitting smoking. It gives us a brief overview about the possibilities of quitting and about the opinions of smokers on quitting smoking. The theoretical part gives us the list of contentual substances in nicotine, list of possible health complications from smoking and possible therapies on dependence on nicotine. By individual substances is mentioned the mechanism and its effects and possible side effects on our organism. Before the chapter dealing with health problems due to smoking is a brief overview about the dependence and the origin of dependence. The following chapter deals with side effects and health complications from smoking, there is a detailed list with influences of smoking during pregnancy or while taking contraception. Following separated chapters deal with the problematic of passive smoking and legislative smoking in the Czech Republic and in the world. The last chapter in the theoretical part is dedicated to a detailed overview with possible therapies. The bachelor thesis deals with evaluation of antifungal activity of selected peptide fractions isolated from potato tuber against Fusarium spp. fungi. Literature review contains information about isolated potato tuber proteins from Solanum tuberosum L., short description of genus Fusarium, species F. solani and F. oxysporum, information about plant defense mechanisms against fungi and at least information about antifungal proteins. The experimental part deals with evaluation of antifungal activity of selected peptide fractions from potato tuber of Solanum tuberosum L. against F.solani and F. oxysporum which were prepared by separation techniques.

Understanding sentences in Czech children at risk for dyslexia and SLI

The aim of this thesis was to evaluate the level of comprehension of czech sentences with a different syntactic complexity in children with specific language impairment (SLI) and children at family risk for dyslexia (experimental groups) compared to typically developing peers (control group). Statistically significant differences were proven both between the SLI group and the family risk group and between the SLI group and control group. The assumed descending tendency of achievement in the test blocks was not substantiated.

Waxes and Lipids Extraction from Miscanthus Giganteus Stems.

In the framework of the project BIORAF, supported by Technology Agency CR (project no. TE01020080), plantations for experimental production of Miscanthus × giganteus and Miscanthus sinensis have been founded by the project partner AGRA Co. Tall stems of this plant contain a broad spectrum of various substances (waxes, lipids, carotenoids, etc.) being potentially exploitable in cosmetics.Extraction processing of crushed dry stems using non-polar solvent is a promising way for separation of these substances into an extract, whereas the waste raffinate containing waste biomass has, after being pressed into the form of pellets, the potential for energetic utilization as green fuel.\n

Ionization detector for environmental scanning electron microscope

Presented work deals with environmental scanning electron microscopy. The construction of the device is described concisely in the beginning. Important part is devoted to interaction of electrons with specimen and signals emitted from the specimen. The work aims especially at detection of secondary electrons using the ionization detector. Experimentally is determined effect of working environment and size of electrode system of ionization detector on detected signal.
