Inaktivace ICAM-1 genu metodou oligonukleotidů modifikovaných lysinem

Conjugation of ligands to antisense oligonucleotides is a promising approach for enhancing their effects on gene expression. In this study 2'-O-lysylaminohexyl group was linked to the uridine base, which replaces one, two or three thymine bases thus modifies the oligonucleotides. This exchange of bases was tested for improvement of silencing target protein expression. Effectivity of modifications in silencing target protein expression was examined with the alicaforsen sequence (DNA) and siRNA. Alicaforsen, currently in clinical trial 3, is a phosphorothioate targeting ICAM-1, which was the model used to evaluate the influence of modifications. The same target was chosen for siRNA to compare the efficiency of DNA and siRNA substances. For the first time, down-regulation of ICAM-1 was shown on the blood brain barrier cell line ECV304. Unmodified/modified antisense oligonucleotides and siRNA sequences were transfected into ECV304 cells with the help of a transfection agent lipofectamine 2000. After 24 hours of transfection cells were disrupted by a chemical lysis. Protein concentrations were determined by Bradford protein assay. ICAM-1 inhibition was assessed with western blot. Analytical evaluation of drugs by using chromatographic methods IV. Diploma Thesis Lenka Spáčilová Charles University in Prague, Faculty of Pharmacy in Hradec Králové Department of Pharmaceutical Chemistry and Drug Control Heyrovského 1203, Hradec Králové Rapid analytical methods are described to control quality of honeys, concerning residues of acaricides applied in hives to prevent the mite Varroa jacobsoni infestation. Tau - fluvalinate was analysed by the use of a SGX RP-18C column, 5 µm, a acetonitrile : water (40:60) mobile phase, flow rate of 1 ml/min and a UV detector set at 254 nm. An internal standard was used for quantitative determination. A calibration curve was constructed and verified, by means of which the concentration of tau - fluvalinate in honey can be determined. The elaborated method was validated by detection limit determination.

Calibration of indirect methods for maesurement of properties of alkali activated concretes

This work solves creation of calibration relations to determine cube compressive strength, dynamic and static elastic modulus of alkali-activated concrete by non-destructive methods. Alkali-activated concrete is spoken of as a new material used in civil engineering. It shows different properties than normal concrete based on Portland cement. That's why the modification of common calibration relation seems necessary. Fresh concrete was made in the concrete plan ŽPSV a.s., Uherský Ostroh in three mixtures and always in the number of 18 cubes and 3 prisms. The samples were tested by impact hammer Schmidt type L, type N, SilverSchmidt PC-N and by ultrasound in 6 time periods of three specimens. After that, the cube compressive strength was determined. Status of static elastic modulus was determined in a time period of 28 days. The results are calibration relations to determine the progress of compressive strength and modulus of elasticity for each method and their combination.

The Novels of Jaroslav Vrchlický

This thesis focuses on the prosaic writing of Jaroslav Vrchlický. In the first part it monitors an extension of the reception in these novels (it analyzes their editions with the help of the database Caslin). It also observes character of the period reception from the first publication of the corresponding titles up to the year 1918 (there is a support of the database Retrobi and Lexikon české literatury). The second part of the thesis analyzes the Vrchlický's novels according to methods of naratology and literary thematology. The third part of this writing focuses on a genre and a themed characteristic of the Vrchlický's novels in the context of that period.

Airtightness estimation of wood based residential houses using Blower-door test

This dissertation analyses airtightness of residential buildings which in many ways influences their energy performance and therefore also their costs of use. Properly conducted airtightening layer can significantly extend the lifespan of the construction. The main objective of the dissertation is to evaluate the construction of newly built buildings in the Czech Republic and to assess the most important parameters which affect airtightness. The Blower-door test was used to assess air leakage. This method belongs among the most widely used and conclusive methods of analysis. Its advantage is mainly the fact that it can detect construction defects of the airtightening layer in important stages of construction (and possibly enable these defects to be corrected before completion). The core of this method is to create a pressure difference of deltap 50 Pa between the exterior and the interior of the building and at the same time discover the faulty places which are different for positive and negative pressure. The air leakage was assessed in 345 residential buildings constructed between 2006 and 2016, especially in conventional wood frame buildings or with buildings made from wood panels such as cross-laminated timber. The development of values of air change rate through the building envelope was evaluated according to the Czech standard ČSN 73 0540-2 and the results were compared to previous outcomes of other authors. Analysis and photo documentation of the most frequently detected leakage points are also part of the dissertation. A significant decrease in values of the air change rate was discovered, on average by 40.49% in 2016 compared to 2006, while for passive houses the air change rate values were almost steady. For buildings with wood frame construction, it was the way of ventilation which was determined as the most significant parameter affecting air leakage. For wooden buildings with natural or combined ventilation, the average value of air change rate at the pressure difference of 50 Pa was 1,29 h-1, for buildings with mechanical ventilation and heat recovery it was 1,18 h-1 and for buildings with very low energy consumption for heating with mechanical ventilation and heat recovery it was 0,44 h-1. Other parameters affecting the values of air leakage include the construction company, the presence of a chimney, the place and method of construction of the wood frame construction.

New cryoprotectives in preservation of potentially probiotic bacteria of Bifidobacterium genus using freeze-drying procedure

The probiotic bacteria of the genus Bifidobacterium are especially used in the dairy and pharmaceutical industry. They are present in the form of viable cells in pro(syn)biotic fermented milk, dried and freeze-dried products for its positive effects on the health. Lyophilization represents one of the widely used methods for long-term preservation of bifidobacteria. The lyophilization process is challenging, but very needed, and for this reason it is still necessary to explore new cryo(lyo)protective substances, which protect the microbial cultures more effectively before the negative effects of cryopreservation and lyophilization. The aim of the thesis was to test the various mixtures of substances containing specific oligosaccharides, which have prebiotic properties, from the point of view of their potential cryo(lyo)protective effect of lyophilization process. For the purposes of the study, twelve bifidobacterial strains of human and animal origin were selected. As a potential cryo(lyo)protectives were used solutions on the basis of 5x concentrated TPY medium with addition of FOS, 10% fructooligosaccharides (FOS) and the solution on the basis of porcine gastric mucin. Number of viable cells was determined in lyophilisates enriched by the concrete solutions in the time intervals (30 days, 3 months and 6 months) after lyophilization and compared with the control lyophilized samples (fresh cultures without the additive solutions). The potential cryo(lyo)protective effectiveness of 5x concentrated TPY medium solution has been demonstrated only in B. boum, B. breve, B. longum subsp. infantis a Bombiscardovia coagulans, where decreased the number of viable cells about four orders of magnitude. The potential cryo(lyo)protective effectiveness in 10% FOS solution was found only in B. thermacidophilum subsp. porcinum and Bombiscardovia coagulans, where was found half the number of viable cells. The highest potential cryo(lyo)protective effectiveness was observed for a solution on the basis of porcine gastric mucin. The highest number of viable cells was demonstrated in B. animalis subsp. lactis, B. boum, B. longum subsp. infantis, B. longum subsp. longum, B. pseudolongum subsp. globosum and B. thermacidophilum subsp. porcinum lyophilisates using this potential cryo(lyo)protective solution.

Implementation and experimental comparison of selected methods for management of XML data

This document describes some methods for storing XML documents in relational dabase (RDBMS) based on encoding elements in XML tree. Selecting is done by translation XPath selects into SQL selects. Document also contains experimental comparison of selected methods with real XML data collections. Implemented methods are XISS/R, XpathAccelerator, PathStack.

Methods of measuring the power parameters of diesel engines

The thesis deals with the performance measurement of tractor engine through four used different methods. In the first theoretical part of thesis is analysis engine design with its main groups and the second part explains methods of measuring performance and measuring equipment. In the practical part of thesis is the explained methodology of applied methods of measurement, together with their evaluation. The first applied method is performance measurement on the dynamometer, the second method is measurement of these parameters during free acceleration, the third method is measurement of parameters during free acceleration with load and fourth method is measurement of parameters using GPS. In conclusion of this part is to compare the results of each used measurement methods between themselves and the conclusions drawn from it.

Support education to health form interventional kinetic programme for children preschool age.

The aim of my bachelor´s work is the formation of interventionist movement programme supporting the health education of children of pre- school age. As a partial goal I have determined the checking whether this programme is possible to be put into practice. Theoretical part of my work contains a lot of information about the importance of movement in the people´s life and about the psychomotor activity and its influence on health. In the practical part there is worked out a formed movement programme which is based on the psychomotor activity games and it is adapted to the age of children. Before and also after its aplication the other data were gained by the methods of watching and of guided interview. All collected data were processed and analysed. The results confirm that the movement programme positively contributes to creating of conditions for physical, psychical and social well-being of children and it is possible to be put into praktice.

The comparison of different methods for soil physical properties determination

This diploma thesis is focused on comparison of the methods which are used for soil physical properties measurement . A penetrometer and Kopecky rollers were used. To assess these methods, the soil properties were evaluated on the land of joint-stock company Sloveč near Městce Králové. The first part of this thesis consists of an introduction which describes the basic information related to this issue. The second part deals with literature review where the basic physical properties of soils are discussed, such as porosity and bulk density. The aim of this diploma thesis is mentioned in the third part. It is followed by the methodology in which the different methods of measurement are described. The next chapter focuses on the analysis of measured data and its evaluation. The next part deals with the technical-economic evaluation of measurement methods. The last part brings the conclusion which summarizes all the results of the thesis. The most interesting result of this diploma thesis that penetrometric measurement is affected by soil moisture content so significantly that it can show soil compaction entirely on other areas of the land than measurements using Kopecky rollers.
