This diploma thesis deals with the processing of project documentation in the documentation phase for construction of building object SO 01 – Detached house with facilities. The thesis is not worked out in full extent, as prescribed in Decree No. 62/2013 Coll. Amending Decree No. 499/2006 Coll., On Construction Documentation. It includes only part A (Data Accompanying Report), B (Summary Technical Report), C (Situational Drawings), D.1.1 (Architectural and Building Solutions) and D.1.3 (Fire Safety Solutions) of Appendix 6 of this Decree. In addition, a thermal-technical assessment of the building was elaborated. There are two specializations in small range as additional part of the documentation as well. The proposed two-storey house with small cellar is situated on flat land in the village of Lipová-lázně and layout of the object is divided into two parts, part of house with function of housing and part with facilities for cooking and catering for more persons.

The use of new technologies in teaching mathematics

Modern information technologies accompany all of us in our daily life. They not only use to serve to our work and for the entertainment, they even have got a great potential to be used in the education. The submitted thesis deals with the appropriate utilization of the special IT application in the pedagogic process, especially in teaching of maths, the topic of trigonometric functions. The aim of this thesis is to create structures with a high potential of the modern technology handling. To tackle this task the graphical program GeoGebra was chosen due to its qualities for creating dynamic structures and the other advantages presented further in the thesis. To the created constructions the detailed instructions how they can be constructed are added. The thesis is also accompanied by worksheets with solved tasks, focusing on the application of trigonometric functions in real situations.

Clinical biochemical indicators used in diagnostics of equine diseases

The thesis aims to compile overview of main biochemical indicators that are important in for diagnostics of equine diseases. Clinical biochemistry is a medical discipline that uses biochemistry and pathobiochemistry for the determination of diagnosis based on activity changes of analytes observed. Laboratory evaluation of analytes has three phases: pre-analytical (preparation of patient to sampling, sampling and sample storage and transport), analytic (analysis) and post-analytical (evaluation of results, veterinarian also contributes in this part). Pre-analytial phase is the most time consuming and also the most error prone. The errors mostly occur during preparation of the patient, during sampling or sample transportation. The most analyzed materials for biochemical analysis are blood, urine and liquor. Biochemical indicators are compound that are often influenced by the disease, therefore it is possible to use them for the diagnosis. Nitrogen metabolism indicators include proteins, urea, creatinine and ammonia. Enzymes (aminotransferase, gama-glutamyltransferase, glutamatedehydrogenase, lactatedehydrogenase, alkaline phosphatase, creatinkinase and sorbitoldehydrogenase are the main indicators of liver function (they contribute on the metabolism of another compounds) or of the fitness of horse. Glucose and lactate values indicate energetic metabolism, cholesterol and triglycerides indicate lipid metabolism. Water and electrolyte metabolism indicators are sodium, potassium and chlorides. The mineral profile is shown by calcium, magnesium and phosphorus. The result of determination of analytes is compared with reference values that are not only species-specific but can be also influenced by age or sex. These values represent the range of the compound concentration in body fluid under physiological conditions. It is important to take the clinical symptoms into the account. Not all indicators that are usually indicated in human medicine or different animals are suitable for diagnosis of equine diseases. Majority of indicators diagnose equine-rare diseases (i.e. diabetes mellitus or hepatitis). Determinations of muscle enzymes and lactate are important for the fitness analysis.

It's essential for a proper function of variety of crucial enzymes. On the other hand it's excess in the body is harmful and it is connected with progress of various pathological conditions. Hence iron chelation therapy is an effective tool to treat and protect living cells against injurious effect of iron. Iron chelators were originally used to treat iron overload disease. However, thanks to their interesting antioxidative and antiproliferative properties, their therapeutic potential has been significantly widened. Small sacral buildings in the area Macha region

Abstract The thesis maps and identifies a small religious monuments in the countryside and smaller settlements in the cadastral area municipalities Association Macha, which is located in the southwestern part of the Liberec region and its southern edge, which is Kokorin, extends into the Central Region. These small structures of sacred significance were set up at intersections of roads, trade routes, near Wells, but as expressions of gratitude and remembrance to joyful, significant and tragic event. Each of these small sacred building affects perception of a person's environment, enriching area villages and landscapes, along with accompanying greenery which acted as a refuge on the road, landmark or feature lightning form a common artistic and traditional value. His mission lost partially or completely inappropriate transfer. The main benefit and purpose of this work is to determine the current state of these monuments in the area, and create a detailed catalog, serving as a source of information for tourism, regional development, such as integration into existing and also into emerging hiking, biking and nature trails, but also as the basis for a partial analysis in the evaluation of the landscape, the creation of the zoning plan, the draft plan during land consolidation or the needs of a particular community. The catalog is compiled on the basis of professional and regional literature, detailed field survey, including cadastral territory of the municipalities that are part of the voluntary association of municipalities Macha maps and testimonies of some residents is compiled catalog of small sacred buildings. Buildings are photographically documented and described both in terms of the status quo, and in terms of landscape significance here is the location of the building indicating the GPS coordinates and plotting the current actual map of the selected area.

Leisure time of young generation reflecting its function of prevention in the Rychnovsko region

This dissertation focuses on free time as a new phenomenon in the industrially advanced society. The goal of this paper is firstly to map how the monitored group of children and youth from Rychnov nad Kněžnou and surrounding villages spend their free time and secondly how the above mentioned group comprehends the preventive function of free time. After the theoretical part which is focused on sociology of leisure, leisure education, and rural and urban area, there is an empirical part containing secondary and primary research (performed in form of questioning) with special focus on offering free time activities. The research found out that both groups of pupils (from urban as well as from the rural area) perceive the free time, possibilities of spending it, and their wishes for better spending the free time in similar way. Although the offer of free time activities in the rural area is significantly more limited compared to the free time activities offered in the urban area, the pupils living in the rural area are surprisingly satisfied with the offer. This limited offer is often accompanied with more common feeling of boredom which could grow into being dangerous; in view of preventive function of free time this fact is not insignificant.

Analysis of software tools to support team cooperation

This work is focused on the analysis of software tools that can support cooperation within small groups or large companies. The aim of this work is to characterize cooperation activities and describe technology, functions and principles that these tools uses. Another aims of this work are analysis of three software tools for cooperation and creating a decision tool for leaders of team projects and tasks. These objectives were achieved thanks to the knowledge of the studied materials and examination of individual tools and their functions. The benefit of this work is not only a global view of software tools for collaboration, but also creating of inspiration for future analyzes of this kind. Another benefit is creating of materials for business leadership or project manager to select and use appropriate tools and creating of material that initiate to the issues of modern technologies in the field of cooperation.

Intracellular distribution of hydrogen sulfide releasing enzymes in maturing porcine oocytes

The study of the molecular mechanisms regulating reproductive processes is crucial for increasing the efficiency of reproductive biotechnologies that are often used in animal breeding. A key part of many biotechnological methods is the cultivation of oocytes, which simulates natural conditions in the ovaries. However this simulation is still imperfect and presents a limiting factor for successful formation of mammalian oocytes.capable of fertilization. Meiotic maturation is influenced by many factors including for example gasotransmiter hydrogen sulfide (H2S). Until recently, hydrogen sulfide was known only as toxic gas polluting the environment. It was found that the hydrogen sulfide is at low concentrations important signaling molecule with a lot of physiological functions. Hydrogen sulfide is produced by enzymes CBS, CTH and 3-MPST in various tissues of the body, including the reproductive system. We hypothesised that enzymes responsible for the endogenous production of hydrogen sulfide, CBS, CTH and 3-MPST, are present in porcine oocytes and their presence varies during meiotic maturation in cellular organelles. The goal of this study was to prove this hypothesis by cultivating porcine oocytes, detection of mitochondria, endoplasmic reticulum and hydrogen sulfide-releasing enzymes using imunocytochemistry and colocalization analysis. Our results showed that enzymes releasing hydrogen sulfide, CBS, CTH and 3-MPST, are present in GV porcine oocytes and also in the oocytes matured to the MII stage, after 48 hours in vitro cultivation. We found that the distribution of these enzymes in cellular organelles in oocyte changed during meiotic maturation. There were significant changes in the distribution of enzymes 3-MPST and CBS in the mitochondria, levels of colocalization coefficients for enzyme 3-MPST increased, these coefficients for enzyme CBS decreased during meiotic maturation. In endoplasmic reticulum we found significant changes in the distribution of all three hydrogen sulfide-releasing enzymes. Levels of colocalitazion coefficients for CBS and 3-MPST increased, while levels of these coefficients for enzyme CTH decreased. Results of our experiments on porcine oocytes can help to clarify the role of gasotransmitter hydrogen sulfide during meiotic maturaion mammals of oocytes and are highly valueable because their possible application in breeding of farm animals and in human assisted reproduction.
