Preparation of hydrogels with the addition of grape pomace extract

The scope of this bachelor thesis is the preparation of hydrogels with antioxidant properties. These properties are achieved by the addition of grape pomace extract. The theoretical part of the thesis briefly summarizes the basic characteristics of the gels and describes the basic quantities in rheology. Furthermore, the classification, preparation and use of hydrogels are described. Attention is paid in particular to gels falling into the category of edible hydrogels. Information on antioxidants and matolins is also given. In the experimental part, extracts from grape pomace are characterized in terms of carbohydrate content, phenolic compounds and antioxidant properties. The extraction itself was carried out in different concentrated aqueous ethanol solutions. The extract with the highest total antioxidant activity, as determined by the ABTS method, was used for the preparation of the hydrogels. Agarose is used for the preparation of hydrogels. The change in the rheological properties of the hydrogels due to the addition of the prepared extracts was observed. The specific structures in the hydrogels were determined by FTIR.
Microplastics in soil

This bachelor thesis deals with the appraisal of the suitability of infrared spectroscopy for the identification of plastics and microplastics in soils. Available techniques were DRIFT (diffuse reflectance) and ATR (attenuated total reflectance). Mixtures of model soil and pure PET were prepared for analysis in various concentrations. Furthermore, a total of 21 samples of soil and 3 samples of compost were evaluated. The obtained data were used for interpretation of infrared spectra, creation of calibrations of PET using ATR and DRIFT methods. Part of the work was the evaluation of validation models using spectra and basic soil parameters.
Energy dispersive X-ray spectroscopy of doped PVDF fibers

Tato diplomová práce zkoumá flexibilní materiál k produkci elektřiny založený na piezoelektrickém polymeru Polyvinylidenfluorid (PVDF). Inkorporací piezoaktivní keramiky lze vlastnosti piezoelektrického polymeru PVDF významně zlepšit a převést na užitečnou elektrickou energii. PVDF byl vytvořen elektrostatickým zvlákňováním do vláken o tloušťce 1,5-0,3 µm a poté studován různými analytickými metodami. Tato práce nabízí popis elektrostatického zvlákňování, přípravu vzorků a teoretický úvod do analytických metod, kterým byly vzorky podrobeny. Morfologie a distribuce nanostrukturované keramiky do polymerní matrice PVDF byla pozorována použitím skenovací elektronové mikroskopie (SEM) a energiově disperzní spektroskopie (EDX). Pro tvorbu fáze a podrobné fázové složení byly vzorky charakterizovány infračervenou spektroskopií s Fourierovou transformací (FTIR). Práce také obsahuje analýzu s použitím Ramanovy spektroskopie, metody používané k identifikaci a porovnání chemických sloučenin. Elektrické vlastnosti byly studovány dielektrickou spektroskopií a je poskytnuta korelace se složením. Jednotlivé komponenty dotovaných vláken jsou charakterizovány a vyhodnocovány v souvislosti s jejich budoucím využitím v senzorech.
Optical response of asymmetric plasmonic structures

This diploma thesis deals with study of resonance modes of plasmonic structures. First part provides an overview of theoretical models, which explain the resonanace modes in plasmonic structures. Next part describes technology of electron beam lithography. First section of experimental part deas with technological processes leading to an improvement of resulting structures made by electron beam lithography that is followed by lift-off process. Last part focuses on a study of reflectance spactra of plasmonic antenas and the identification of resonance modes.
Heterogeneous Radical Modification of Polypropylene

The diploma thesis deals with the heterogeneous technique of preparation of grafted polypropylene g maleic anhydride (PP-g-MAH). The knowledge of the course of the reaction and its impact on the PP grafting process are summarized in the theoretical part. The preparation of the grafted PP was carried out in a fluidized polymerization reactor at 115 °C, speed of mixing 210 rpm, reaction time 60 min and pressure 6 bar. The amount of grafted MAH was monitored by using a MAH concentration 3 and 5 wt% and an initiator concentration 0.25; 0.5; 1; and 1.5 wt%. Modification efficiency was compared by using 3 types of PP with different particle morphology and specific surface area. By creating the theoretical model, the maximum surface concentration of MAH was calculated and compared with experimental data. Characterization of PP materials was performed by Electron Scanning Microscopy (SEM), Differential Scanning Calorimetry (DSC) and particle surface analysis by BET. Quantitative analysis of grafted MAH was determined by FTIR spectroscopy and acid-base titration.
Developement and validation of method for ferulic acid isolation from wheat bran

This master thesis deals with an optimization of processes of isolation of ferulic acid from wheat bran. Process of isolation based on alkaline hydrolysis from the substance and its adsorption was tested. In the theoretical part were described wheat bran, adsorption proces and used methods. For adsorption were tested five types of adsorbents. Especially these adsorbents: activation carbon (2,53 ± 1,16 mg.l-1), Amberlyst A-21 (105,73 ± 11,87 mg.l-1, Amberlit XAD-16 (241,55 ± 10,42 mg.l-1), Amberlit IRA-900Cl (5,90 ± 0,68 mg.l-1) and Amberlit IRA-96 (189,16 ± 6,49 mg.l-1). As the most efficient adsorbent was determine Amberlit XAD-16. The whole process has underwent detailed optimization in order to maximize the purity of the final product. The isolate was characterized by HPLC and FTIR techniques for ferulic acid and related phenolic acids. Based on FTIR analysis was discovered that the isolate was contamined by proteins. A purification procedure based on deproteination of the raw material before the hydrolysis itself was proposed. As the ultimate solution that eliminated the presence of proteins in the isolate was determine ultrafiltration with capture of molecules with Mw > 10 KDa. Ferulic acid yield from one kilo of wheat bran was 1,00 ± 0,22 g.
The forensic analysis of soil by FTIR and NIR with multivariate analysis

This diploma thesis is about forensic multivariate soil analysis in the localities Ostrava and Třinec. A total of 52 samples were taken in the areas near metallurgical companies. These samples were measured for concentrations of chromium, manganese, copper, nickel, lead, mercury and zinc. Mercury concentration was analysed using AMA 254 device. The remaining metals were determined by flame atomic absorption spectrometry (F–AAS). Additionally, IR spectra were acquired by Fourier transform infrared spectroscopy (FTIR) using diffusion reflectance technique (DRIFT). In the end the results were used to create correlation models and statistical models of PCA (principal component analysis) methods with CA (correlation analysis).
Bioactive substances for face cream: Determination of stability of face cream incorporated with goat's milk

The aim of this bachelor thesis is a stability trial of a skin cream incorporated with goat milk lyophilizates prepared in usual laboratory conditions. Firstly, the goat milk was denatured, to be further analysed via FTIR, elemental analysis and thermogravimetry. To compare the denaturation effect, measurements with native goat milk sample were performed as well. Next, an emulsion was prepared. First stability tests were performed using analytical centrifuge and the cream preparation procedure and component ratio was optimized. Stability trials were carried out at specified time intervals. Rheological properties – especially yield stress values and structure strength, determined by linear viscoelastic region and complex modulus – thermal endurance measured via differential scanning calorimetry and visual validation via optical microscopy and sample observation were tested and performed. Skin cream stability of sample stored at laboratory temperature was demonstrated, showing moderate sings of structure destabilisation. Product suitability for dry skin was proved by sensory analysis. The skin feeling was pleasant, and the cream moisturized the surface area, that it was applied to, according to evaluators. Visually, the emulsion appeared to maintain its properties during the experiment time range, it did not undergo a complete degradation of separation into two original phases, nor it was degraded, for example by mold.
Sequential fractionation of organic matter of humic acid isolated from Leonardite

The aim of the diploma thesis was to optimize the sequential fractionation method of organic matter to be used for physico-chemical characterization of extracted fractions. Humic acid isolated from oxidized brown coal of Leonardite was used as a source matrix of organic matter. An eluotropic series was assembled and sequential fractionation was performed by extraction on a Soxhlet apparatus. The original humic acid and fractions were characterized by elemental analysis (EA) and thermogravimetric analysis (TGA), followed by Fourier transform infrared spectrometry (FTIR), molecular absorption spectrometry (UV/VIS), fluorescence spectrometry and potentiometric titration. Atomic ratios were determined from the results of the elemental analysis. From the measured UV/Vis and fluorescence excitation and emission spectra, the absorption coefficients, resp. fluorescence coefficients. Used fractionation method proved to be a suitable method for studying HA structure. A total of 62 wt. % of initial materiál was extracted, indiivdual fraction amounted from 0.36–30.92 wt. %. From the results of the structural analysis, it is clear that with increasing polarity of the organic solvent, fractions with long aliphatic chains were first isolated and their aromaticity graddualy increased. Non-polar organic solvents were suitable for the extraction of lipid-like coumpounds, while the most polar organic fractions were rich in polar groups and their structual parameters were close to the original humic acid. The fraction extracted with acetonitrile was the most unique fraction. This fraction was rich on nitrogen and amine groups and was similar to protein-like structures. In the last two fractions, extracted with alcohols, a significant bathochromic shift typical of fluorophore type V was observed. Among other things, they were also characterized by a higher content of plant carbohydrate residues.
Utilization of physico-chemical and spectroscopical methods in study on microbial extremofiles

The aim of this bachelor thesis is the determination od polyhydroxyalkanoates in the cells of extremophille bacterie, preformed by several methods. The chosen bacterium was Schlegelella thermodepolymerans DSM 15 344 Thermogravimetric analysis, infrared spectroscopy with Fourier transformation, and Raman spectroscopy were used in this thesis. The TGA method was used for the determination of extracellular and intracellular water in samples. The FTIR method was used in order to observe the amount od PHA, but aslo to observe the physical properties of the polymer in the sample, specifically to determine either amorphous or crystalline featuers of this polymer. Raman spectroscopy was used in oder to confirm the presence of PHA in the samples. All results e´were compared eith the results accomplished by gas chromatografy.
