Vlčková, Zuzana ; Flodrová, Dana (referee) ; Šalplachta, Jiří (advisor)
The aim of this Master Thesis is the optimization and verification of conditions of separation of proteins from biological materials. For the separation a developed technique called micropreparative isoelectric focusing is being used. The theoretical part describes analytical methods of biological materials with a focus on proteins which were used for the optimization of the developing system in general. This part of the Master Thesis also presents this method itself. Furthermore, basic characteristics of other techniques used to control the efficiency of the micropreparative isoelectric focusing are introduced. The experimental part depicts the individual steps of optimization of the micropreparative isoelectric focusing. The ideal separation procedure was firstly found out on the individual standard proteins, secondly on their mixture and lastly the optimised procedure was applied to a real biological sample. The efficiency of the micropreparative isoelectric focusing was verified by SDS-PAGE and MALDI-TOF mass spectrometry.

70 REGISTERING AND RECORDING LEGAL RELATIONS TO LANDS IN THE REAL ESTATE REGISTER Summary This MA thesis deals with the question of registering legal relations to real estates and with recording these legal relations in the real estate register. Since the principle of registering today is in a way connected to the historical context that not only influences the current state and legal form but also leads to various problems, there is a brief summary of the most important institutes and ideas of former ways of registering real estates and the legal relations to them at the beginning of this work. What follows is an explanation of the real estate register as a current instrument for registering real estates and the legal relations to them, its functions, subject matter and area, together with the main principles of its keeping. Attention is also paid to keeping the data recorded in the register in accordance with reality and to the resultant problems. The topic area of recording legal relations to real estates is then developed in more detail, beginning with the principles of their recording, continuing with their forms and their being carried out by real estate registers in practice, and ending with problems connected to the records. The tasks of real estate registers while keeping the real estate register's...
Reproductive isolation between diploid and tetraploid cytotype of Vicia cracca and possibilities of evolution of this aggregate
Vlčková, Zuzana ; Münzbergová, Zuzana (advisor) ; Mráz, Patrik (referee)
Master thesis investigates reproductive barriers in diploid-polyploid complex of Vicia cracca. Complex with basic chromosome number x=7 consists of diploid (2x=14), tetraploid (4x=28) and rare triploid (3x=21) cytotype. I studied prereproductive barriers between diploid and tetraploid cytotype: phenology of flowering, pollinators' behavior (preference of spieces of pollinators to cytotypes, sequence of visited cytotypes), variables, that could explain pollinators' behavior (amount of nectar as the main reward, size and amount of pollen grains as a potentional reward). To find out how strong the triploid block is I analyzed ploidy of seeds and seedlings from mixed-ploidy population. The habitat isolation showed up to be the strongest reproductive barrier. Pollinator's behavior meaningfully contributes to isolation, phenology of flowering contributes only minimally. Index expressing rate of prereproductive barriers is 0,956. Pollinator Bombus pascuorum visited on one locality preferably tetraploid plants and Andrena sp. preferred diploid plants. Even though tetraploid plants produce more nectar, no other analysis showed pollinators' preference to tetraploid plants. I prepared one squash of diploid V. cracca using method of in situ hybridization. This method needs to be optimilized for the studied taxon.
Virtual playground
Svoboda, Tomáš ; Vlčková, Zuzana (advisor) ; Pelikán, Josef (referee)
Present document describes algorithms used in program Virtual playground. This program was designed to simulate movement and interaction of perfectly rigid bodies in real environment with gravity. It had to be optimised to be able to simulate tens of bodies on contemporary PCs in real-time. It was in fact also to the prejudice of physical precision but I think that the result is physically plausible. There is also vehicle simulation added to the program. I will try to introduce difficulties I had during programming and I will do a comparsion with other algorithms that are used to solve similar kind of problems. In the end, I will describe program structure and provide user guide for the program.

Nowadays, video games are a big part of mobile phone software industry. Special place holds multiplayer video games, which can be divided to short-range and long-range. This work aims to short-range wireless network technologies as Bluetooth, WiFi and Infrared. The main goal of the work is to create a real-time multiplayer variation of Bomberman game based on universal network interface in that way, so it will by totally independent on particular wireless network technology. Universal communication interface, will be implemented by a separate module for each network technology. Therefore a deep understanding of short-range wireless communication is required, which the focus of this work is also targeted on.

Recording legal relations and other data to lands in the real estate register
Vlčková, Zuzana ; Drobník, Jaroslav (advisor) ; Franková, Martina (referee)
THE ABSTRACT This thesis deals with the question of filing legal relations and other data to real estates with recording these legal relations and other data in the real estate register, included some problems which are in the real estate register incident and incurred while keeping the real estate register's state of filing in accordance with reality. To better realization of a generic context, especially for comprehension of the real estate register principles and recent state of filed data in the real estate register, there is also the historical evolution of registering the real estate in this thesis mentioned. It is difficult to understand some specific problems without knowledge of history because these problems result from former methods of registering the real estate and legal relations to them. Great attention is paid to keeping the state of filing in the real estate register in accordance with reality that is why the illustrations of some particular cases and examples are shown. This was led by effort to connect theoretical principles with their practical use, the major model situations were assumed from the experience of Klatovy Real Estate Register's employees. In the conclusion, an evaluation of the current state of filing in the real estate register is provided and there are some prospects...
Configuration and routing mechanisms behaviour in ad-hoc
Vlčková, Zuzana
Technologies of local and personal wireless networks allows building ad-hoc networks which don't require prepared infrastructure of optical or metalic wiring. Besides mechanisms of channel sharing and mechanisms of routing, also mechanisms of topology control are very significant, in these networks. The goal of this diploma project is to evaluate behaviour of chosen mechanisms for topology configuration of wireless ad-hoc networks, optimizing suitable criterion - required energy or interference. Real node distribution of nodes in wireless networks doesn't agree with uniformly or Poisson distribution, published solutions are related to. Interesting would be to evaluate methods minimizing interference for more realistic node distribution (groups of nodes) and also in case of using frequency multiplexing.
ELECTROCHEMICAL STUDY OF CuSCN INORGANIC HOLE-TRANSPORT MATERIAL FOR SOLAR CELLS PREPARED BY ELECTRODEPOSITION FROM AQUEOUS SOLUTION

A comparative study is reported for electrodeposited copper(I) thiocyanate layers (ca. 500 nm) on two types of conductive/semiconductive substrates: i) carbon (boron-doped diamond_BDD, glass-like carbon_GC), and ii) carbon-free F-doped SnO2 conducting glass (FTO). SEM and Raman evidence that electrodeposition from aqueous solution results in homogenous CuSCN layers with dominant thiocyanate ion bounded to copper through its S-end (Cu−SCN bonding), as in spin-coated CuSCN layers. Electrochemical impedance spectroscopy (EIS) confirms the p-type semiconductivity of layers with a flatband potential from 0.1 to 0.18 V vs. Ag/AgCl depending on the substrate type, and the acceptor concentration (NA) of 5 x 1020cm-3 in all cases. The flatband potentials determined from Mott-Schottky plots (EIS) are in good agreement with the Kelvin probe measurements. The blocking quality of CuSCN layers was tested using Ru(NH3)63+/2+ redox probe. CuSCN deposited on BDD substrate exhibits better blocking properties compared to CuSCN deposited on FTO.
Usage of formylmethionin derivates in tumor immunotherapy (Model melanoma B16-F10)
VLČKOVÁ, Zuzana
Cancerous diseases and alternatives of their therapy belong to one of the most discussed issues of present day. Cancer is one of the most common lifestyle diseases. It is estimated that external factors cause the origin of 75 % of all cancerous diseases. The most influential external factors are hectic lifestyle, obesity, wrong eating habits, smoking, excess of alcohol intake, inappropriate sexual behaviour and negative approach to preventive check-ups and at last the permanently increasing lifespan of the population. The most serious problem is the occurrence of malignant tumors that are diagnosed so late. Therefore, the aggressive therapy is often the choice of treatment. But this kind of therapy often irretrievably damage the health of the patient. Modern medicine tries to find some new ways of cancer therapy. One of them is treatment of tumors by targeted immunotherapy using the naturally occurring matters. The main aim of this bachelor thesis is to describe and verify the possibility of treatment of malignant melanoma by formylmethionin derivates. The bachelor theses consists of two parts. Theoretical part is focused on describing the issue of cancerous diseases, tumor formation and classification. It focuses on tumors of skin, their diagnostics and possibilities of the therapy, especially of the immunotherapy by innate immunity activation. In experimental part of thesis is described the cultivation of melanoma cell line B16-F10 for in vitro experiments and the transplantation of cells into syngeneic mice strain C57BL/6 from Charles River Laboratories. Melanoma cells B16-F10 in the number of 400 000 cells were transplanted to 8 weeks old mice, which had a weight of 18 - 20 grams. The methods of measuring tumor size and comparing tumor temperature versus mouse body temperature is also described. The result of the first experiment concludes, that therapy of B16--F10 tumor by f-MLF-(K)12 motif bound by charge do not work, because it does not cause phagocytic attack. In second experiment it was found out that B16-F10 tumor therapy by 3mM f-MLF alone does not work, because it does not cause tumor reduction without the LPS in combination. In the third experiment f-MLF motif was used. It was anchored by lipophilic chain with stearic acid, it was used alone or in combination with LPS. In this experiment the complete temporary elimination of tumors occurred, but after therapy termination the tumor growth appeared almost immediately. The mouse treated by this compound survive for more than 52 days than control mice. Due to experiments performed in this bachelor thesis was found out, that anchoring of formylmethionin peptides to tumor cell have positive effect to decrease of tumor volume in successful immunotherapy.
