Assessment of cytostatics used in the treatment of retinoblastoma by means of high-efficiency fluid chromatography

The goal of this bachelor's thesis was to develop and validate a new method enabling detection of etoposide in plasma and the vitreous humour. Etoposide belongs to the roup of cytostatics used for retinoblastoma and other tumours treatment. It is possible to determine etoposide by high performance liquid chromatography, enzimoimmunoanalyticly, or by radioimmunoanalysis. Etoposide is tied in plasma up to 95% to plasmatic proteins, namely albumin, and that is why the drug dosage is low. Monitoring of the etoposide level enables more effective and safer possibilities of treatment. Etoposide is determined by high performance liquid chromatography with UV detection at 229 nm. During the evaluation of the method its accuracy, robustness, linearity and range and limit of detection was checked. The method was checked on 200 plasma and vitreous humour of rabbits. Calibrating curve is linear up to 80 μg/ml in plasma as well as in the vitreous humour. Variation coefficient regarding the repeatability, reproducibility and robustness was lower than 10%. It was found out that repeated de-freezing of the sample does not effect the etoposide determination. This method is suitable for determination of etoposide in vitreous humour and plasma. Determination of pesticides enantiomers in water

This Barchelor's project deals with chiral herbicide mecoprop and its determination by high-performance liquid chromatography method with teicoplanin based chiral stationary phase. The teoretical part focus on basic facts conserning chirality, chiral pesticides and problems related to their usage and effects to the enviroment. It also involves the ways of their determination by the above mentioned method and gives several examples of previous separations. The experimental part is based on method optimalization and determination of changes in mecoprop enantiomeric ratio in various water samples during different storage conditions.
Preparative diastereomeric separation of silybin and its sulfates by RP-HPLC

Silymarin is standardized extract isolated from fruits of Milk Thistle plant (Silybum marianum). The principal component of silymarin is silybin. This flavonolignan is mainly responsible for the medicinal effects of Milk Thistle fruits: antioxidant, hepatoprotective, anticancer and chemoprotective activities. Natural silybin exists as an equimolar mixture of diastereomers A and B whose preparative separation is very hard. It was shown that the biological activity of silybin A and B are different. Silybin in the blood conjugates mainly to sulfates. The structure or biological activity of the sulfates is not yet known. The aim of this work is to develop practically applicable method for preparative separation of diastereomers of silybin A and B, and sulfates, which are considered as one of the major metabolites of silybin. The preparative method for separation of silybin A and B in the mobile phase consisting of 50% MeOH on the chromatographic column Labio C18 25x250 mm, was developed. In addition, preparative method for separation of mixture of products accured from the sulphation of silybin-23-acetate in the mobile phase consisting of MeOH/H2O 60/40 (v/v) with addition of 10 ml/l HCOOH, was optimized.
Determination of enantiomers of selected drugs by HPLC

Drugs are often chiral compounds. In many cases only one of the enantiomers has required therapeutical effects. Citalopram belongs to a group of chiral drugs. This drug is used as an antidepresivum for treating anxiety, fear or suppressing of pain feeling. Its synthetical precursor is named citadiol and this compound is also chiral. HPLC method with chiral stationary phases based on macrocyclic antibiotics was applied for enantioseparation of above-mentioned compounds. Polar-organic and ionic-polar separation modes were found to be suitable for chiral separation of citalopram and citadiol enantiomers. The aim of this work was monitoring of enantioseparation "behaviour" of selected enantiomers and to find suitable separations conditions using HPLC method.
Determination of enantiomers of amino acids by HPLC

This Bachelor's thesis deals with determination of amino acids enantiomers, namely alanine in the experimental part, by high performance liquid chromatography using chiral stationary phases based on teicoplanin. The theoretical part is focused on summarizing the fundamental issue of chirality and chiral amino acids in foods, the characteristics of high-performance liquid chromatography, as one of the most appropriate methods to enantioseparation amino acids, and description of the methods of derivatization, which are used in determination of enantiomers of amino acids. The experimental part deals with the separation of alanine enantiomers, with the choice of the best separation system for the separation and determination of D-and L-alanine in fruit juice.
Bioethanol as a renewable energy source

Tato diplomová práce se zabývá sledováním fermentačních postupů zaměřených na produkci ethanolu. Ethanol je produkován kvasinkami anebo bakteriemi Zymomonas mobilis. Zymomonas mobilis je gramnegativní bakterie, která se vyznačuje vysokým teoretickým výtěžkem ethanolu během fermentace a toleruje vysokou koncentraci ethanolu. Přestože je Zymomonas mobilis výborným producentem ethanolu, není vhodná pro konverzi biomasy, protože zkvašuje pouze glukosu, fruktosu a sacharosu. Tato práce se zabývá fermentačními testy bakterie rodu Zymomonas mobilis na širokém spektru substrátů za rozdílných podmínek. Byl sledován proces výroby ethanolu pomocí Zymomonas mobilis, konkrétně kmeny z České sbírky mikroorganismů CCM 2770, CCM 2771, CCM 3881 a CCM 3883. Porovnávány byly kultivace na různých typech medií (sacharosovém, fruktosovém, glukosovém a obilné zápary) při různých koncentracích uhlíkatého zdroje a za různých podmínek (aerobní, anaerobní). Ke stanovení obsahu cukru a lihu bylo použito HPLC. Ukázalo se, že jsou vhodné především kmeny CCM 3881 a CCM 2770, které narostly během 48 hodin a bylo u nich dosaženo nejlepších výsledků výtěžnosti ethanolu ( až 99 % teoretického výtěžku). Kmeny CCM 3883 a CCM 2771 rostly velmi pomalu, produkce ethanolu u kmenu CCM 2771 byla zanedbatelná. Jako nejlepší substrát se jeví glukosa,...
