Methods for identification of PHA producing bacteria

This diploma thesis deals with testing, optimazing and comparing methods for the identification of bacteria producing polyhydroxyalkanoates. Work included cultivation and microscopy methods, wherein the bacterial cells were stained with lipophilic dyes Nile red and Sudan black. Further, we also used flow cytometry and spectroscopic methods - Raman spectroscopy and infrared spectroscopy with Fourier transformation, and molecular biological methods, which analyzed the presence of a gene encoding PHA synthase (phaC) by polymerase chain reaction (PCR). PCR assay consist of two reactions, the firt on eis based on amplification of phaC gene along with 16S rRNDA gene, which is common for all the bacteria (multiplex PCR). The second reaction is focused on specific amplification of PHA synthase catalyzing biosynthesis of mcl-PHA. In order to overcome false positive results typical for methods analyzing genotype and also to avoid false negative results occuring in fenotype analyzing methods, the best strategy is to combine both aproaches. According to our results, analysis of presence of phaC gene by PCR can be combined with methods capable of determining presence of PHA in bacterial cells. For this purpose, Raman microspectroscopy seems to be very promising tool, since it is able to detect low content of PHA in cells and PHA can not be confused with other lipid metabolites. The results provide an overview of test methods, their advantages and disadvantages and also to compare different criteria according to which it is possible to choose the method of identification in depending on the adjustable requirements.
Everyday life in Slaný around year 1600

Presented thesis deals with the everyday life of the 16th and 17th century in the royal town of Slaný in the central Bohemia. It recounts the eventful history of the city from its prehistoric beginnings to the present. Focus is the fourth and fifth chapter. The fourth chapter discusses the type sof criminal offenses and punishment for them in the early modern period grant. This includes offenses against life and healthy, against property, against morality and honor. This section we also learn about dishonest professions. The fifth chapter describes the everyday concerns of people at the time - the need to belong, cocern for the livelihood, concern for subsistence but also financial matters. Full text concludes humorous incident that happened in 1604.

This diploma thesis extends the theme of bachelor thesis, in which alternative site specific exhibition spaces, called "independent galleries", in the Czech Republic were observed and described in terms of motivation and the reasons for their establishment. The diploma thesis focuses on the study of specific mainly alternative exhibition possibilities of contemporary art and on a description of the contemporary phenomenon of exhibition space abroad regarding the Czech context. These findings are developing into its own site-specific art activity in public space, which is then transformed in educational structure and implemented with the students of a selected high school in Prague in public spaces around the school. The educational part of the thesis is complemented by a qualitative research, which helped to capture mediation specifics of the topic in the educational process.
Still life

The bachelor thesis deals with meaning of objects in our society. This thesis is divided into three parts - practical, didactic and pictorial part. The theoretical part examines an ordinary and necessary subject, subject as a bearer of meaning and a subject as a protective and ritual thing. I am simultaneously reffering to consumerism and sensitive understanding of the subject as a value in today's society. I compare the tools of the past with a mobile phone. The teoretical part is followed by the didactic part, which is composed of three artistic lectures I have given. Me and children discussed the following topics: Painted still life, Shop window and The box with my stuff. The pictorial part focuses on everyday objects. I have created the cycle of twelve still lifes. Each still life coresponds to one particular person, who is close to me, and symbolize their important things.
Contemporary textile art. Artwork of Sheila Hicks as Inspiration for Art Education

The first part of the thesis has the character of an exploratory probe, which is looking for the form and situation of textile art at present. It is based mainly interviews of experts in the field of textile art and experiences of the visited exhibitions. Part of this thesis is also personal creation, based on textile experiments. From these findings there was created a pivotal part in the form of a didactic project, inspired in particular by the creation of Sheila Hicks. Results and benefits of the work is to outline the current situation of textile art in our country and verified in a practical set of teaching practices focused mainly on the development of sensory sensitivity. The didactic methods are supplemented by reflections and extending or innovate variant sof lesson plans in Art Education.
Sensory experience in art education

The diploma thesis is called "Sensory experience in art education." The theoretical part explores sensory perception through the knowledge of philosophy, psychology and art theory. It contains examples of the art works of the selected artists. It also deals with the content of RVP in relation to sensory perception and didactics of art education, with a specific purpose of designing an art project in art education. The essence of the didactic part is to devise an art project with the focus on human senses. Each of the tasks is centred on one of the senses: sight, smell, hearing, touch and taste. This project is further tested in practice with the help of the second grade students of elementary school and high school. The results are then analysed and evaluated. The amended form of the project can be currently used by art teachers. The selected tasks are the basis for the creative, authorial process of art work, which reflects the effect of the tasks and develops the students' further abilities.
