PhDr. Ondřej Císař, Ph.D. Abstract The thesis deals with collective behaviour, as a form of coordination between people for political or social purposes during a pandemic. I am particularly interested in this issue from the perspective of the transformation of political activism. For this work, I define collective behaviour as a meeting of at least three people, in order to express an interest or problem in relation to public institutions, both in person and online. The mentioned pandemic period means the term from March 1, 2020, when the first Czech patient was tested positively, until August 31 of last year. The aim of the theoretical part is to present the political activism itself, individual types of collective behaviour, to describe the strategic protest repertoire and a view of symbolic instruments. The empirical part provides an analytical and systematic capture of collective action in the Czech Republic during pandemic. For that, protest events analysis was chosen as a methodological tool. Field bryophytes of the Czech Republic

3 ABSTRACT More than 30 % of agricultural land in Europe also Czech Republic is covered by arable fields. There is no evidence about diversity of some arable fields bounded species groups. In general the diversity of arable land species is less than the wild land species. For increasing species diversity in agricultural land European commission comes up with agri environmental farming schemes in 90's. Till now real effectiveness of the agri environmental schemes is not clear for all species groups. In my thesis I have investigated whether the diversity and community composition of arable bryophytes species is influenced by agri environmental or conventional management on cereal fields. The influence of agri environmental management on arable bryophytes was never investigated in Europe unlike the other species groups like vascular plants, non vertebrates or vertebrates. The thesis stands on dataset provided by Ministry of agriculture for 3 Czech regions (kraj Vyso ina, Pardubický kraj, St edo eský kraj). My own research of arable bryophytes diversity held on 12 fields within 6 localities is the main part of the thesis supplemented by basic soil measurements and climatic characteristics extractions. Transcriptional activity of the genes characterizing developmentally competent cytoplasm in bovine oocytes.

4 Abstract The antral follicle provides a specialized microenvironment or niche, which is necessary for production of high quality oocyte. The developmental competence of bovine oocyte is influenced by the follicle size. Oocytes originated from medium or larger follicles (≥6 mm) have greater developmental competence (ability to develop to the blastocyst stage). The changes in cytoplasmic factors, for example mRNAs, could explain differences in oocyte developmental potential. Using Bovine Oligonucleotide microarrays the differences in gene expression profiles of oocytes at germinal vesicle and MII stages from medium (MF, 6-10 mm) or small (SF, 2-5 mm) follicles were characterized. The aim was to find differencies between oocytes diverse developmental competence. The expression fold change between the two experimental groups was in 61 genes. Subsets of 15 differentially expressed genes were validated by quantitative RT-PCR. Before maturation, significant differences were confirmed at the level of ATP5C1, MAP3K13, MTRF1L, TAF1A and UBL5. Subpopulations of oocytes were classified according to atresia of cumulus cells and follicle size. We determined the level of 12 individual transcripts after maturation. The Effect of EGF - like Peptides on Cumulus-Oocyte Complexes

In mammals, the ovarian cycle is controlled by follicle-stimulating hormone (FSH) and luteinizing hormone (LH), secreted by the anterior lobe of the pituitary gland. FSH is assumed to be essential for follicle growth. LH is required for the terminal phases of follicle growth, estrogen synthesis, ovulation, development of the corpus luteum and progesterone synthesis. Although cumulus cells and oocytes lack LH-receptors, LH causes cumulus expansion and meiosis resumption. The effect of LH is mediated via EGF-like peptides - amphiregulin (AREG), epiregulin (EREG) and betacellulin (BTC). The precursors of these peptides are present in the mural granulosa cells and EGF-like peptides are released by activation of metalloproteases in response to LH-stimulus. The aim of this thesis is to present EGF-like peptides in the wide context of the molecular mechanisms regulating the late phases of folliculogenesis and meiotic maturation of oocytes.
Signaling pathways and genes regulating gonadotropin-induced maturation of porcine oocytes and cumulus expansion

In vitro, meotic maturation of porcine oocytes and cumulus expansion are induced by FSH and EGF-like peptides AREG and EREG. FSH and EGF-like peptides induce expression of cumulus expansion-related genes (HAS2, PTGS2 and TNFAIP6). To define signaling pathways that control FSH- and AREG-induced cumulus expansion, porcine cumulus-oocyte complexes were treated with specific protein kinase inhibitors. Inhibitors of MAPK3/1, MAPK14 and ERBB1 significantly reduced both FSH- and AREG-induced expression of HAS2, PTGS2 and TNFAIP6. These inhibitors decreased FSH/LH-induced expression of AREG and EREG in mural granulosa cells. Surprisingly, inhibitor of PKA had no effect on AREG expression in cumulus-oocyte complexes but the inhibitor decreased expression of TNFAIP6 induced by AREG. Inhibitor of PI3K increased expression levels of AREG and PTGS2 but EREG, HAS2 and TNFAIP6 were reduced. Expression levels of the cumulus expansion-related genes were not affected by an analog of cGMP (8-CPT-cGMP). However, 8-CPT-cGMP blocked spontaneous in vitro meiotic maturation of porcine oocytes and its effect was abolished by FSH.
