The first part theoretically explains all the tools that are later used for project processing. The second part deals with the introduction and analysis of the current situation of the company. The first part, in theory, explains the concepts of knowledge, managerial decision-making and after that all the methods that are used during the processing of this work. The second part deals with the introduction of the company and the analysis of the current situation in the software market to support knowledge management. Experimental system for production of IL-15 on viral carriers

Interleukin 15 has great application potential such as in the biological treatment of cancer. It is involved in a variety of immunological processes, the most important of these involve influencing and induction of NK cells and T-lymphocytes proliferation. However, its therapeutic usages are limited by a low stability and short half-life. For this reason, there are various approaches of stabilization and expansion of its biological activity being explored. In this work, we analysed and developed a new approach, which uses viral nanostructures derived from major capsid VP1 protein of mouse polyomavirus as a carrier of IL-15. Moreover, VP1 proteins can be relatively easily modified and they are also capable to penetrate into the tumour cells. There were prepared two variants of IL-15 together with control nanostructures in the baculovirus expression system, one was composed of IL-15 and the other of the IL-15 fusion protein and truncated variant of VP1. Protein constructs were characterized by electron microscopy and biochemical methods. The total protein yield of VP1ΔC-IL15-HIS fusion variant was higher (up to 53 mg/L of complete medium) than IL-15 alone (8,5 mg/L). Analysis of posttranslational modifications of proteins by mass spectrometry

Viruses, etiological agents of many infectious diseases, are small noncellular particles that run their replication process solely inside host cells. It is generally assumed that the posttranslational modifications of viral capsid proteins are responsible for their infectivity (e.g. phosphorylation catalysed by kinases of host cells). The appropriate model for study of the viral phosphorylation "profile" relation with its infectivity is mouse polyomavirus of the Polyomaviridae family. By comparison of virions, as well as the major capsid protein VP1 of wild type mouse polyomavirus with viral mutant created by deletion of the part of the genome coding regulatory proteins of virus and produced in two different cell lines WOP and 3T6 was found by mass spectrometry a major phosphorylation in the three specific amino acids of VP1. These are considered important for proper morphogenesis of virions and their ability to infect host cells. The qualitative representation was not affected by the cell line selection. The role of JC virus in etiology and therapy of multiple sclerosis.

JCPyV is a human polyomavirus. Infections caused by this type of virus is often without any clinical symptoms and JCPyV mostly persists in kidneys of human for the rest of the life. The most serious disease caused by the JCPyV is progressive multifocal leukoencephalopathy, which causes severe demyelination of neurons in the brain. Multiple sclerosis, as well as progressive multifocal leukoencephalopathy, is characterised by the demyelinization of neurons. In the past few years, it was shown that the development of PML occurs in some pacients who suffered from multiple sclerosis and who were treated with the monoclonal antibody called natalizumab (Tysabri). This text describes the analysis of the conncetion between viral infection of JCPyV and the etiology of multiple sclerosis and progressive multifocal leukoencephalopathy. As the risk factors for the progression of progressive multifocal leukoencephalopathy in natalizumab treated multiple sclerosis patients the duration of the use, previous immunosuppressant therapy and the presence of the anti-JCPyV antibodies have been demonstrated.
