GlcNAc8-dendrimer as a ligand of NKR-P1A receptor in effector-target cell interaction and G-protein signaling
Svoboda, Jan ; Kuldová, Markéta ; Huliková, Katarína ; Miller, R. G. ; Fišerová, Anna
GlcNAc8 glycodendrimer is a specific ligand of rat NKR-P1A receptor isoform. In effector cells triggers the Gq/11 protein-mediated signaling pathway, and induces subsequent changes in tumor microenvironment
Multivalentní glykokonjugáty ve studiu interakce lektinových receptorů NK/NKT buněk
Vannucci, Luca ; Pospíšil, Miloslav ; Rossmann, Pavel ; Chernyavskiy, Oleksandr ; Kubínová, Lucie ; Křen, Vladimír ; Luptovcová, Martina ; Svoboda, Jan ; Kuldová, Markéta ; Bezouška, Karel ; Fišerová, Anna
The events following the administration of GN8 in vitro, in vivo and ex vivo, lead to increased expression of CD69. The CD69-dependent IFNg production is modulated by co-activation of NKG2D and Ly49 receptors. Glycodendrimers can assist the dissection of activation network of NK/NKT cell lectin-like receptors in rat and murine models

Kuldová, Markéta ; Svoboda, Jan ; Kovářů, H. ; Kovářů, F. ; Šplíchal, Igor ; Fišerová, Anna
Adenosine receptor, NK cell, immunity systém, cytotoxicity, Salmonella typhimurium, colorectal carcinoma, murine model, newborn pigs model
Úloha A2A adenozínového receptoru v NK buněčné cytotoxicitě
Pompach, Petr ; Vančurová, Markéta ; Man, Petr ; Novák, Petr ; Bezouška, Karel
After studies with monoclonal antibodies, we would like to focus on the identification of proteins associated with membrane microdomains of human NK cells by mass spectrometry
Cell sorting in practise
Cimburek, Zdeněk ; Horváth, Ondřej ; Růžičková, Š. ; Fišerová, Anna
Cell sorting is used for prepare cell clones, functional studies, clonal analyzis, expression studies. We are using FACS sort for positive and negative sorting, for single cells cloning and in some cases is used immunomagnetic presort
Úloha sacharidových struktur v NKR-P1 zprostředkované interakci efektorových a cílových buněk

Masking oligosaccharide sequences can assist transformed cells to escape immune surveillance. N-acetyl-glucosamine coated dendrimers have binding affinity for NKR-P1 receptor and can also modify cancer cell glycosylation profile. Effects of treatment on immune cell subpopulations were analyzed in blood, lymphoid tissues and organs, and inside tumor in B16F10 melanoma bearing mice. Treatment enhanced tumor cell immunogenicity, and induced maturation, differentiation and homing of cytotoxic cells to tumor microenvironment.
