Gallery of Modern Art in Brno

The bachelor's thesis focuses on the design of a modern art gallery located in the city district of Brno-Center. The plot, with an elevation ranging from 205.200 to 207.100 meters above sea level, features an irregular layout. While the central location is advantageous, nearby roadways contribute to the noise in the local environment. The structure, consisting of five floors with two underground and three above-ground levels, is designed in accordance with the city's plan for mixed-use areas, taking into consideration the planned circular park. The main entrance is located on the first floor on the western side, featuring a large entrance hall, a café, a library, and office spaces. The second and third floors are designated for exhibition spaces, supplemented by rentable studio areas. The underground sections include double-decker garages and supply rooms. The design concept responds to the surroundings; the building's break follows the street line in the Kolište section, and the northern façade is parallel to the city hall. The building's façade is visually divided. The first above-ground floor is glazed with white plaster, while the other above-ground levels are covered with aluminum panels. The objective of the project is to integrate into the urban environment and provide a space that supports cultural and social activities in Brno-Center.
Shale gas extraction

This bachelor’s thesis on Shale gas extraction describes extraction and world shale gas resources. At the beginning it provides introduction to the terms of geology and overview of historical evolution of extraction and crucial technologies that are currently used in extraction. Shale gas exploration and extraction development is briefly mentioned. Horizontal drilling and hydraulic fracturing is then further described and also summary of the equipment is provided. Potential environmental risks associated with extraction or hydraulic fracturing itself are described. Subsequently world shale gas resources and potential extraction places are described. World technically recoverable resources are estimated at 7245 Tcf. Shale gas commercial production is currently underway in the United States, Canada and China.

SMS Application for JavaME Platform

The aim of this diploma thesis is to study the potential of Java ME platform for receiving and sending of SMS and to utilize obtained information subsequently in implementation of an application for this platform. The first part of the diploma thesis aims at explaining general terms of the Java language and its basic features. The next part deals with specific properties of the Java ME platform and properties of devices that support this platform. A design of the Java ME application based on theoretical knowledge is created then. This application has to be able to receive, process, store and send SMS and communicate with the server application based on the Java SE platform. The created Java ME application serves as an automated SMS voting server, which can be used in mobile devices that support the Java ME platform and can be remote-controlled via Internet from the Java SE platform application. The final chapter presents the results and conclusions of testing of the created application on real mobile devices.
Preparation of natural food supplements for children's nutrition

The presented diploma thesis is focused on complex characterization of chosen superfoods and subsequent composition of supplement suitable for children´s nutrition. Different kinds of superfoods were chosen for analyses, such as: barley grass (Hordeum vulgare), wheat grass (Triticum aestivum), moringa (Moringa oleifera), matcha tea (Camellia sinensis), microalgae chlorella (Chlorella pyrenoidosa), spirulina (Arthrospira platensis), flax seeds (Linum usitatissimum) and sesame seeds (Sesamum indicum). The theoretical part is focused on children´s nutrition issue and the most common food allergies and intolerances. Also, children food products legislation and characterization of chosen samples are included in theoretical part. Main macronutrients, such as saccharides, lipids and proteins, were analysed in the very beginning of experimental part. Fibre, fructan and -glucan, which have a high impact on intestinal microbiome and whole process of digestion, were analysed as well. However, barley grass was the best evaluated sample by containing 205,5 mg/g of insoluble fibre; 152,57 mg/g of fructans and 18,76 mg/g of -glucans. Finally, for complex nutritional profile, also samples pigments were analysed. Spectrophotometry, HPLC chromatography, gas chromatography and other analytical techniques were performed for experiments.

The subject of the thesis is to design and consider steel superstructure of a composite steel-concrete road bridge. It is a continuous beam with seven spans of 36,0 + 57,0 + 75,0 + 90,0 + 75,0 + 57,0 + 36,0 = 426 meters. The superstructure consists of welded I-beams, which pass into closed box girder. The calculations take into account the construction phase.
Enterobacter spp. resistant to beta-lactam antibiotics from urban and hospital wastewater

The increasing prevalence of antibiotic–resistant bacteria pose a huge risk for public health. Enterobacter spp. is a common in the environment but it is also documented as a frequent cause of human disease. Enterobacter spp. resistant to cephalosporins and carbapenems is increasingly detected globally both in and out of hospital setting. This thesis examines the occurrence of Enterobacter spp. strains resistant to –lactam antibiotics especially in wastewaters but also in treated water discharged into the environment. The aim of this study was to characterize a set of 205 strains of Enterobacter spp. selected on medium supplemented with cefotaxime and meropenem. The water samples were collected from three regional cities in the Czech Republic and originated from hospital wastewater, inflow and outflow of municipal wastewater treatment plant (WWTP), from a river above and below the WWTP. The isolates were screened for presence of eight genes encoding production of broad–spectrum –lactamases (ESBL; CTX-M, GES) and carbapenemases (GES, IMI/NMC-A, IMP, KPC, NDM, OXA-48, VIM) by the polymerase chain reaction (PCR) method. The phenotype of isolates carrying the gene of interest was tested for –lactamase production by the Mastdisc test and susceptibility to 24 different antibiotics by microdilution method. The phylogenetic relatedness was determined using restriction digestion in combination with pulsed-field gel electrophoresis (PFGE). From total of 205 strains, 36 isolates carried the gene of interest for –lactamase production. These isolates originated mostly from hospital wastewater (n=31) of all sampled sites (A, B and C). Also one isolate was detected in the WWTP influent in site B and four isolates came from the WWTP effluent in sites A and B. The following genes of interest were detected among the isolates: blaGES (n=16), blaCTX-M (n=24), blaOXA-48 (n=1) and blaNMC-A (n=1) a combination of two genes were detected in one isolate. All characterized isolates (n=36) showed a multidrug–resistant profile. High resistance to aminoglycosides was reported (76,9 %) with the exception of amikacin (19,4 %). In the case of carbapenems, resistance to ertapenem was observed (77,8 %), while a majority of strains were susceptible to meropenem, where resistance was detected only in 13,9 % of cases. Different resistance profile has been reported to cephalosporin antibiotics. All tested isolates showed resistance to antibiotics cefazolin and cefuroxime (100 %), lower rates of resistance were reported to antibiotics cefoperazone (80,6 %), cefotaxime (77,8 %) and cefepime (69,4 %). The lowest resistance in this group of antibiotics was found to cefoperazone/sulbactam (33,3 %). All selected isolates showed resistance to tetracycline antibiotics. Serine carbapenemase production (50 %) was the most frequently detected by phenotypic tests which is produced certain variants of –lactamase GES. Comparison of macrorestrictions profiles showed a high diversity among isolates represented by 20 different clusters, the related isolates originated from the same location. Wastewaters play a key role in the spread of antibiotics resistance. In this thesis, Enterobacter spp. strains with clinically significant resistance to cephalosporins and carbapenems were detected. These strains were detected mainly in hospital wastewaters but also in WWTP effluent. The current WWTP processes are unable to eliminate all bacteria that subsequently enter the environment.
The manipulator SMC MAP 205 control

The thesis deal with a model of electronically controlled assembly minicell MAP-205 based on pneumatic drives and its debugging. Assembly minicell simulates assembling and disassembling of simple four item assembly. Minicell is controlled by Phoenix Contact ILC 150ETH programmable logic controller. The minicell control program is composed of several subroutines/subprograms providing various functionalities to assembly minicell. There is integrated HMI created in Control Web application too. It allows to display actual status of each component and to control each drive in manual mode. Communication between HMI based on PC and PLC controller is supported by Bluetooth module and OPC server.

The diagnostic and prognostic ability of selected serum and urinary markers of prostate cancer Abstract Serum prostate specific antigen (PSA) is the only widely approved marker in prostate cancer (PC) diagnosis and follow up after treatment. Its role has remained controversial due to lack of specificity and the risk of overdiagnosis of insignificant PC. The aim of this work was to explore promising markers of PC and to improve current patient stratification to adjuvant treatment. Three main studies were performed using different media (urine and serum). The first study included the evaluation of Engrailed-2 (EN2) - a urinary marker of interest - in 90 patients with localized PC, 30 healthy controls, and 40 patients indicated for prostate biopsy. The second study evaluated 205 men with high-risk PC-features who underwent radical prostatectomy (RP) and were subject to a strict follow-up protocol of ultrasensitive PSA (UPSA) at close time intervals. The ability of particular measurements to predict biochemical recurrence (BCR) and thus the need for adjuvant therapy was assessed using the area under the curve (AUC) and a stratification model was created. Pevnost vybraných konstrukčních spojů s použitím modifikovaného dřeva buku

The thesis focuses on the influence of selected European beech wood modifications (Fagus sylvatica L.), i.e. acetylation and thermal modification at 200 °C and 220 °C, on the strength of structural joints used in the furniture industry. Based on the literature research, three types of structural joints were selected, which were subsequently tested. The strength of the glued joint was tested according to ČSN EN 205, withdrawal capacity of a screw according to ČSN 49 0135 and the corner screw joint. The results of the measurements were statistically evaluated using a one-way ANOVA and the measured groups were compared to each other. The results demonstrate that all types of modification lead to a decrease in mechanical strength. Wood acetylation lead to a statistically significant differences in the values for the glued joint. The thermal modification at 200 °C achieved statistically significant differences in the withdrawal capacity of a screw. The biggest drop in values came after a thermal modification at 220 °C, which were statistically significantly different when testing all selected joints. In the case of acetylation and heat modification up to 200 °C, it is not possible to unequivocally identify a method that showed significantly better results across all tests. Both metods of modyfying wood showed possibilities of use in furniture design. However, a decrease in the mechanical properties ofthe modifies material muse be taken into account when determining the dimensions of the structure.
