This bachelor´s thesis describes problems of light microscopy for readers. Part of this bachelor´s thesis is dedicated to basic terms of the light microscopy. Next chapters are dedicated to observing techniques - brightfield illumination, darkfield illumination, oblique illumination, Rheinberg illumination, phase contrast, differential interference contrast, polarized light microscopy and fluorescence illumination.
Creating conditions for evaluation of high-strength concrete characteristics using non-destructive testing methods

High-strength concrete (HSC) belongs in the recent years to frequently used types of concrete. It allows realization of static challenging structures and also shows due to its dense structure greater durability especially against aggressive media. Currently HSC construction realization abroad is not exceptional. It’s using in the Czech Republic is still limited. When realized, then in a small scale in civil engineering works. The realization of high-strength concrete structures is closely related with the concrete construction quality verification. Good efficiency of the quality control methods can provide non-destructive testing methods (NDT), especially when investigating strength of concrete built in structure. A lack on relevant data for non-destructive testing of HSC in technical and normative rules is to be considered as a significant deficiency. Evident for HSC generally is the lack in literature on deeper analysis of the factors affecting their non-destructive testing, as well a meaningful methodology or practically usable calibration relationships. HSC differs from ordinary concrete not only by used components, but also by more compact structure with different strength – elastic characteristics. Considering these differences, HSC strength prediction can not be performed by using calibration relationships developed for ordinary concrete. Moreover, the question is to what extent the current knowledge of the NDT results influencing factors can be considered as valid. The paper presents findings on the effects of the key factors affecting the measurement results of Schmidt hardness method and ultrasonic pulse method, including recommendations for the practical application of these methods. The problematic of static vs. dynamic modulus of elasticity was also solved. Calibration equations for predicting the compressive strength of HSC from the non-destructive testing parameter were elaborated, showing high cohesion among variables and practically usability.
Study of influence of toxical and nutritional elements on cell metabolism using combination of Raman spectroscopy and Laser-Induced breakdown spectroscopy

In this work on-line discrimination of six bacteria strains by means of the Multivariate discrimination analysis (MVDA) is presented. Principal components analysis (PCA) was selected as most suitable technique. Two analytical methods – Laser-Induced breakdown spectroscopy (LIBS) and Raman spectroscopy were equipped for chemical analysis of bacteria strains. In order to obtain the best possible bacteria strains differentiation, the data from both methods was analyzed in two ways separately and together. The data from both measurements was normalized separately and afterwards combined in one data frame for simultaneous analysis. This data frame contained information from both analytical methods. Moreover an influence of cultivation period for each bacteria strain was studied. It was determined that Raman spectroscopy is able to discriminate two bacteria strains and LIBS even four bacteria strains. Using combination of both methods the complete discrimination was achieved. From information of first principal component it was determined that most valuable information in LIBS data is not included in different elemental composition but rather in influence of matrix effect. Solely the LIBS was utilized for studying the effect of cultivation period. It was possible to observe transformation of four bacteria strains within 24 hours. Based on results of this work it is possible to assume that combination of Raman spectroscopy and LIBS, because of the complementary information, is suitable for fast discrimination of different bacteria species and strains. Moreover it was determined that LIBS is able to observe the transformation caused by cultivation period.

The diploma thesis deals with analysis of the current situation of the company Domácí dobroty s.r.o., which produces and sells dried fruit. The aim of the thesis is to suggest a proposal of marketing strategy focusing on introduction of a new product and expansion of company promotion. The work also includes strategic analysis of an external and an internal environment of the company. The suggestion part of the thesis focuses on speci-fic proposals of new marketing strategy.
Two-axis manipulator with vacuum clamping of samples

This thesis focuses on design of a two-axis manipulator with vakuum attachment of samples. Linear guides, ball screws and stepper motores were chosen for the design. The design also includes calculation of the load on individual carts, the load capacity of ball screws and the load of stepper motors.
Development of abrasion-resistant concrete and testing methods

This diploma thesis deals with concrete abrasion resistance against abrasion. The constructions of transport and water management structures are most stressed by the action of abrasion by means of travel of vehicles and flowing water. Abrasion reduces the durability of the structures. The prerequisites for achieving the best possible abrasion resistance are examined. Furthermore, test methods that are currently used worldwide. The experimental part deals with the influence of individual types of aggregates amphibolite (Želešice), fecal gravel sand (Žabčice) and basalt (Bílčice) on abrasion resistance. Furthermore, the dependence between the strength of the concrete and the hardness of the surface for abrasion resistance. Abrasion resistance testing was determined by concrete abrasion, abrasion resistance by the Böhm method and by water jet.
Models of switching power supplies in MATLAB-Simulink

The Bachelor’s thesis is focused on the issue of models of switching power supplies. The first part describes the principle of switching power supplies. The second part is devoted to the design of the drive parameters and appropriate regulatory circuit. The last section is devoted to developing models of switching power supply in the MATLAB using the Simulink block Power system.

Automatic sample tilt compensation for chemical mapping by LIBS

This thesis deals with designing an algorithm for slope compensation of a sample examined using the LIBS (Laser-induced breakdown spectroscopy) method. It mainly fucuses on the purpose of elemental mapping. The sample slope noticeably affects the result quality and therefore a demand for a simple and inexpensive solution to this problem appeared that would also be applicable for commercial uses of LIBS.
Methodology of internet research

The origin of the Internet dates back to as early as 1969 when several computers were successfully connected without employing any central unit. However, it hadn't seen any substantial development until 1989 when the web pages came. Thanks to a dynamic development, it has reached an almost global expansion since then. The permanently growing number of interested users has gradually involved also social scientists. Originally, the Internet was perceived as interesting in itself, but in the course of time it has become also a methodological tool used in investigations into social issues. This paper deals, first of all, with the Internet as an option to use in data capture. It defines advantages and drawbacks of this tool in terms of social research. Its focus is on the elimination of risks and disadvantages related to this methodology. Further, it specifies some basic rules, which a researcher should follow when designing an internet research and creating a questionnaire. This work addresses principles related mainly to a quantitative research, however, in the last part there are also some possibilities to a qualitative or mixed research suggested.
