This bachelor´s thesis describes problems of light microscopy for readers. Part of this bachelor´s thesis is dedicated to basic terms of the light microscopy. Next chapters are dedicated to observing techniques - brightfield illumination, darkfield illumination, oblique illumination, Rheinberg illumination, phase contrast, differential interference contrast, polarized light microscopy and fluorescence illumination.
Microscopic Traffic Simulation Model Calibration

This thesis main focus is microscopic traffic sumulation. Part of this work is the design and implementation of microsimulation model based on cellular automaton. Implemented model supports calibration with genetic algorithm. The results of calibration and simulations are included.
Image processing of data from the ultrasound system GE VingMed System Five

The master thesis is focused on the principle of ultrasonographs and their modes, and ways of denoising data obtained by ultrasonographs. This project will also concentrate on removing the speckle noise, because the speckle noise has the largest share of the noise in the measured data in ultrasonographs. The other main objective was to describe advanced methods of filtration, especially the modifications of the median filter, such as the modified hybrid median filter and modification of the anisotropic diffusion, namely generalizing Perona-Malik anisotropic diffusion using restricted dissimilarity functions. These two filters were tested on images with artificial noise and on data captured using the ultrasound system GE VingMed System FiVe. Also, the program for filtering and display filtered ultrasound data captured by GE VingMed System FiVe is described.
Software for Optimalization of Gear Speed

Rolling vehicle is being influenced both by driver and many other factors of physical nature. These factors affect the whole behavior of the car, its performance, fuel consumption etc. It is very reasonable to cope with these factors then. The main aim during vehicle evolution (both civil and racing ones) is to optimize the influence of all invisible enemies as much as possible. The aim of this work is a description of factors influencing the moving vehicle. Several practical examples are included. The theory of transmissions is fundamental to the programme for creating a saw-diagram and gear speed optimization. Finally a few analysis of the real vehicles and subsequent evaluation will be done. The output of the work is an overview of factors affecting the moving vehicle, the results of performed analysis and programme for optimization of gear speed.
RESIDUAL FATIGUE LIFETIME OF RAILWAY AXLES

This Ph.D. thesis deals with methodology for determination of residual fatigue lifetime of railway axles based on damage tolerance approach. This approach accepts an existence of potential defect, which could lead to fatigue failure of whole axle. The behavior of crack in railway axle is described by approaches of linear elastic fracture mechanics. There are plenty of factors, which more or less influence determined residual fatigue lifetime. The aim of this thesis is to quantify effects of these factors. The first part of Ph.D. thesis represents overview of studied problems relating to fatigue damage of railway axles. This part is focused on parameters, which influence fatigue crack growth in railway axle materials. The second part of thesis shows procedure for determination of residual fatigue lifetime, which was developed at the Institute of Physics of Materials of the Academy of Sciences of the Czech Republic. The main aim of this thesis was to improve current procedure for more precise estimation of residual fatigue lifetime. Significant part of this work is determination of significance of studied factors, which influencing calculated residual fatigue lifetime of railway axles (e.g. effect of threshold value, load spectrum, retardation effects, residual stress, axle geometry, material of axle etc.). The procedures described and results obtained can be also used for determination of residual fatigue lifetime of general mechanical component (not only railway axles). Therefore, results obtained in this Ph.D. thesis can be used e.g. for assessment of regular inspection intervals of cyclically loaded general mechanical parts.
Driven roller conveyor

The thesis describes design of driven roller conveyor for transport products on Euro pallets of a total maximum weight of 600kg. The design includes determination of the respective sizes, capacities and strength, as well as design of a support frame and final strength control in the most affected areas. The thesis also contains documentation with drawings of complete conveyor, section of the conveyor and last section of the conveyor.
Lifetime Prediction of the Welded Ribbed Generator Shaft

This master thesis deals primarily with the prediction of fatigue life of welded ribbed shaft. Recommendations taken from the International Institute of Welding (IIW) are used to express the stress values and the fatigue life. The stress values of examined place are expressed by approach of nominal stress, structural hot spot stress and effective notch stress. Results from stress-strain analysis are combined together with fatigue strength curves for prediction of the fatigue life. At the end of the analysis, the predicted fatigue life is compared with design life of welded ribbed shaft.
