Supervisor: Prof. PharmDr. Petr Pávek, Ph.D. Title of doctoral thesis: Novel approaches for development of in vitro liver cell models. The liver is a main metabolizing organ in the body. Therefore, the evaluation of hepatic metabolism is a crucial step during drug development. Moreover, a liver damage induced by drugs is another task to be assessed in drug development. In vitro liver cell models allow addressing some of these concerns. Up to date, primary human hepatocytes are considered as a "gold" standard of in vitro liver cell models. Additionally, other liver model systems are used such as liver tissue slices, subcellular fractions, liver cancer cell lines and hepatocytes derived from stem cells. Despite the significant progress towards right estimation of pharmacokinetic and toxicological parameters of drug candidates during drug development, current in vitro systems still suffer from various drawbacks. One of these limitations is their insufficient similarity with in vivo-like phenotype associated with low metabolic capacity of the models. Preparation of anti-cancer vaccine / / immunotherapy for ovarian cancer under conditions of good manufacturing practice

Generation of clinical-grade dendritic cell-based vaccine for immunotherapy of ovarian cancer Introduction: Dendritic cells (DCs) the most potent antigen presenting cells. Recent technological advances allow for generation of large numbers of DCs from peripheral blood monocytes. Administration of activated DCs loaded with tumor antigens is thus an attractive approach for immunotherapy of cancer. Prerequisite for the initiation of clinical trials in cancer immunotherapy is the development of protocols for DC-based vaccine generation according to Good Manufacturing Practice (GMP) conditions. Aim of the study: Development of protocol for the generation of clinical grade vaccine based on activated DCs loaded with killed tumor cells for use in the immunotherapy of ovarian cancer. Materials and methods: Immature DCs were generated from peripheral blood monocytes of healthy donors. We tested Cell Gro and RPMI+5% pooled human serum (5% PHS) as clinical grade culture media. Immature DCs were then activated by three distinct stimuli (Poly I:C, LPS and cocktail of proinflammatory cytokines (TNF, IL-1 and IL-6)). Výzkum vhodnosti lokalit v ČR z hlediska zásob větrné energie a zpracování metodiky pro posuzování‚ schvalovací a aplikační řízení pro zavádění obnovitelných zdrojů energie‚ zejména větrných elektráren

Cílem výzkumné zprávy je na základě vlastního výzkumu a zahraničních zkušeností, převážně z Německa, zformulovat postupy, které umožní optimalizovat numerické modelování proudění, vymezí rámec jednotného přístupu k formulování předběžného posouzení větrných poměrů stanoviště na základě výpočtů meteorologických charakteristik matematickými modely a k formulování posouzení větrných poměrů stanoviště na základě stožárového měření složek větru, případně dalších parametrů. Součástí zprávy jsou dvě studie: 1. Studie větrného potenciálu. Optimalizace větrného parku. Větrný posudek a analýza výnosů větrného parku pro objekt Albrechtice u Frýdlantu; 2. Možnosti umístění větrných elektráren v Krušných horách z pohledu krajinného rázu. Další přílohy tvoří tabulky a mapy průměrných rychlostí větru, umístění meteorologických stanic a studie, 3-D pohledy, metodiky.

Práce se snaží zanalyzovat finanční situaci společnosti Český Telecom a na jejím základě navrhnout vhodná opatření, která by zásadním způsobem zlepšila finanční zdraví podniku, zejména v kontextu prohlubujícího se konkurenčního prostředí, které lze sledovat v posledních letech v České Republice.
