Effect of pharmaceuticals present in recipients of STP in fish.

Volume of consumed antibiotics in developed countries have increased eminently in past few decades. These substances are excreted from human body in various abundance and end up in the sewage. Sewage water flow to sewage treatment plants to be partly cleaned and finaly discharged to surface waters. Pharmaceutical residuals leaking to water environment of recipients affect local biota. Impact of pharmaceutical residuals present in biological pond Čežárka was assessed in this study. Čežárka biological pond (2,6 ha) is a recipient of discharged waters of sewage treatment plant in Vodnany, the town with 7 000 inhabitants. Biological ponds are good solution for final treatment of wastewater discharged from sewage treatment plant efluents. The nutrients from wastewater may enter the food chain of pond ecosystem. In total 62 pharmaceuticals were identified in water of sewage pond. Fish exposed in the pond for 30, 90, 180, 320 and 360 days were compared with fish of control group. Growth, condition of fish and biomarkers including level of vitellogenin in blood plasma, lipid peroxidation rate, antioxidative activity of catalase, glutathione peroxidase, glutathione reductase, glutathione S-transferase and superoxide dismutase were compared. The study proved the environmental impact of STP dischanrge containing pharmaceutical residuals in fish. Significant adaptation of fish to environment of biological pond was observed after 90 days of exposure.
Role of chromosomal aberrations to evaluate genetic risk of exposure to carcinogens.

(in English) Air pollution is a serious worldwide problem associated with the risk of cancer. The negative effect of carcinogenic polycyclic aromatic hydrocarbons (c-PAHs), including benzo[a]pyrene (B[a]P), on human health is analyzed using specific biomarkers. Among them biomarkers of early effect play an important role. This work summarizes the results of cytogenetic analyses performed by fluorescence in situ hybridization (FISH) (whole chromosome painting of chromosomes #1 and #4) and automated image analysis of micronuclei (MN). During the analyses a total set of 1304 samples was analyzed by the FISH method and 885 samples by the automated image analysis of MN. Studied groups including city policemen, garage men, bus drivers, administrative workers, mothers, newborns, healthy children and children with bronchial asthma and laboratory workers were from Prague, Ostrava and Ceske Budejovice. The locations significantly differed in levels of air pollutants and the type of air pollution. The exposure of participants of the study was assessed by personal and stationary monitoring. The impact of other factors including age, smoking or intake of vitamins was also evaluated in these studies. Use of biomarkers for assessing environmental pollution by platinum group metals

This diploma thesis in the theoretical part analyses the characteristic feature of individual platinum group elements, their toxicity, anthropogenic sources and the presence of platinum group elements in the environment of the Czech Republic. In addition, there are specified types of bioindicators focusing mainly on lichens, mosses and needles. There are described individual bioindication techniques and pre-treatment of samples for platinum metals determination in bioindicators. The subject of the experimental part is to verify the sorption efficiency and ability of platinum and palladium using the SPE method and to determine the actual platinum metal contamination in selected localities in the city of Brno. There was also the additionally subject to verify the sorption potential of selected bioindicators and determination of the environmental pollution by platinum metals using the bag technique. In the experimental part, the bioindicators of moss Pleurozium schreberi, lichen Xanthoria parietina, lichen Flavoparmelia caperata and needles Pinus nigra were used.
Voltammetric determination of homovanillic acid at boron doped diamond electrode for hollow fibre microextraction

Presented work deals with hollow-fibre assisted liquid/liquid/liquid microextraction (HF-LPME) and subsequent voltammetric detection of clinical biomarker homovanillic acid (HVA). Three main areas are discussed: hollow fibre assisted extraction of HVA, voltammetric detection of HVA under basic conditions, and sensing in very small volumes. Using HF-PLME, HVA was extracted with 1-octanol as a supported liquid membrane with enrichment factor of 55 after 60 minutes of extraction. A novel method of miniaturized voltammetric determination of HVA using cathodically pretreated boron doped diamond electrode was also developed. Using differential pulse voltammetry, limit of detection, limit of quantificati was 1.9 mu mol dm(-3) on 6.6 mu mol dm(-3).
Novel, versatile capillary electrophoresis instrument with laser induced fluorescence for analysis of various lipid peroxidation biomarkers

In this work we have developed a new sensitive method for quantitation of MDA in\nbiological samples by CE-LIF with LOD about 1.7 nM. First part of the work was\ndevoted to the construction of a new CE-LIF system. Then the derivatization process\nof MDA by TBA was optimized. The ability to use this method for the detection of\nMDA in a biological samples, such as EBC and plasma, was also demonstrated. For the\nfirst time it was possible to detect MDA in EBC samples by CE. Other biomarkers are\nnow being studied.
Using proteomics for screening of potential breast cancer biomarkers

Breast cancer is one of the most common malignant cancers and the prognosis is closely related to the stage of carcinogenesis process in which it was captured. Lately, attention was focused on the research of new compounds that would reduce the negative impacts of chemotherapy. It was found that retinoids are effective compounds in the fight against cancer. Retinoids have the ability to inhibit tumors of the mammary gland in an early stage of disease. This work deals with the proteomic studies of breast cancer cells and comparing the proteins contained in the cancer cells which were subjected to retinoid therapy. The differences in protein composition were investigated using gel electrophoresis, reversed phase and size exclusion liquid chromatography. The identification and characterization of selected proteins was carried out by MALDI-TOF MS. The use of two-dimensional gel electrophoresis led to the identification of molecular chaperons, which are considered to be important biomarkers not only of breast cancer.
Importance of biosensors and biomarkers for environment contamination assessment

This bachelor thesis is purely theoretical. It aims to explain the principle and nature of operation of biosensors and biomarkers, their properties, distribution and use in the assessment of environmental contamination. It describes their practical use. In the first part of the thesis are described biosensors. Biosensors combine biorecognition molecules and a suitable converter, this combination allows you to convert (bio)chemical information into electrical signals that can be easily measured and therefore are becoming more used for determination of different substances. The second section describes the early warning signs called biomarkers. Biomarkers provide information on changes in the ecosystem before it is irreversible damaged, and we can intervene early.
Search for new biomarkers for screening and early diagnosis of cancer

Thesis on the topic "Search of New biomakers suitable for screening and early diagnostics of cancer" is dedicated to the issue of cancer, a cancer of the breast, prostate, ovarian and after the new findings out as melanom determination of tumor markers and their implications for clinical practice. Their aim is skouted up biomaker suitable for screening and early diagnostics of carcinoma( cancer ) of the breast, ovarian, prostate and malignant melanoma with utilization of various kinds of immunoassay. The theoretical part gives a brief and comprehensive overview of the epidemiology of selected tumors and tumor markers used and their qualities. In the methodological part are given principles applied iminoanalytics methods a list of the statistical methods and a detailed description of the groups of patients. Results section contains tables with results of biomarkers diveded by sort of avalueted tumors. In the discussion are then analyzed facts that follow from the results. It discussed the process and actualy status of searching convenients biomekers and their combinations that can distinguish a population of benign tumors of the population with malignant tumors and markers which are able to distinguish healthy population from the population with malignant tumors of the breast, ovary, prostate and melanoma. This work may serve as a source of knowledge and results to continue in search of such marker that could be used for screening and early diagnosis of cancer and so to improve the prevention of these illneses in the population.
Application of geochemical analysis in the research of chemical fossils

Biomarkers (also termed chemical fossils) are specific organic compounds in geological samples which are derived from biochemical precursors by reductive or oxidative alteration processes. Chemical structures of biomarkers can be related back to the original compounds and utilized as indicators of biogenic, paleoenvironmental and geochemical processes. Examples of identified biomarkers in several studied coals, sediments, and wood fossils remnants are presented.
