Gallium complexes for molecular imaging of bone tissue

Supervisor's Email address: kubicek@natur.cuni.cz ABSTRACT This thesis is focused on preparing new ligands for selective complexation of gallium, which might serve as potential radiopharmaceuticals for 68 Ga-PET bone imaging. Two new ligands were prepared, combining 1,4,7-triazacyclonone-1,4-diacetic acid macrocyclic skeleton and bis(phosphonate) pendant arm, bound to the remaining free nitrogen atom on the macrocycle. Macrocyclic skeleton is responsible for high kinetic and thermodynamic stability of the Ga3+ complex and the bis(phosphonate) pendant arm insures selective delivery of the complex to the bone tissue. Both new ligands were fully characterized by NMR and mass spectroscopy. Complexation of Ga3+ was studied by 31 P and 71 Ga NMR spectroscopy. Binding to bone tissue was simulated by adsorption of the complexes to hydroxoapatite. Targetting prostate tumor cells by polyomavirus virus-like particles

The aim of this thesis is to investigate the targeting potential of mouse polyomavirus (MPyV) based virus-like particles (VLPs) as vectors for directed cell delivery of therapeutic or diagnostic compounds. Major capsid protein VP1 of MPyV is able to selfassemble into the noninfectious VLPs. Our main goal is to retarget these VLPs from its native receptor to the prostatic cancer cells by changing the receptor binding site in the surface-exposed loop of VP1. We introduced a peptide ligand CTITSKRTC, which binds prostate-specific membrane antigen (PSMA), by insertion or substitution into BC loop of VP1. These modifications did not change the stability of the particles and genetic substitution prevented the native receptor binding. PSMA-specific binding of modified VLPs was tested by pull-down assay and surface plasmon resonance. In order to further utilize these VLPs, we tested several approaches for preparation of VLPs as vehicles for compounds delivery into eukaryotic cells. Thirty compounds (from which 29 are new) were prepared with moderate or good yields. All compounds were fully characterized by NMR and MS. Four new crystal structures were obtained by RTG monocrystal analysis. Derivative containing 4-nitrobenzyl group in the side chain was used for study of bis(phosphonates) adsorption parametres to hydroxyapatite (with quantification by UV-Vis spectroscopy). Several derivatives containing characteristic functi- onal groups (-NH2, -COOH, -NCS) were prepared. These compound are suitable for further derivatization, mainly for conjugation with more complex structures through amide bond or thiourea bridge. Conjugates of building blocks with fluorescent probe (fluorescein and dansyl) were prepared for potencial optical imaging. Marketing Strategy of a Newly Established Company

The main objective of the thesis is to work on a marketing strategy intended for a newly established private charter company. The general principles, marketing concept and strategy layout applied to the company in question are elaborated on. The primary objective is to conduct the analysis of both external and internal environment terms, to conduct a competitor analysis and to deduce the market potential for the particular market area. The next objective is then to identify the client and their requirements, to select the target of the client demand as well as to compare the client demands with the company potential supply. Based on the market survey, the marketing goals are specified and executable strategies are presented. The marketing strategy includes a future profit prediction based on the defined goals and a proposal for verification activities for a parallel plan-keeping control. In general, the aim of the present thesis is to create a marketing strategy that would be integrated in the company management and its business activities.
Customer satisfaction with Kia cars

The main subject of the thesis is to determine customer satisfaction with the Kia Motors Czech company cars. In the theoretical part of the thesis is explained the issues of the situational analysis, which is primary used to profit from information, needed for the correct application of the decision-making process of the company's senior management. Part of the situational analysis is market research, which includes the area of market segmentation and the definition of the concepts of targeting and positioning. The situational analysis is in the theoretical part of the work mainly focused on the analysis of the customers, where is defined loyalty and customer satisfaction, customer expectations, fidelity and designed ways of long-term care for the customers of the company. The practical part of the thesis is focused on the monitoring of customer satisfaction and customer loyalty in the selected company. Part of the work is to select the appropriate system for the measurement of results, which is applied to the selected company. By using a half structured questionnaire is collected data to identify customers, their preferences and requirements for the product, and the evaluation carried out the strengths and weaknesses of the product according to the opinion of the customers of the company. Using the comparison of internal company data and research is carried out a survey of current situation on the market. The conclusion is made a summary of the results achieved and making recommendations for the improvement of the marketing activities of the company and for the streamlining of the company's communication with the customers.
