Factors regulating the expression and activity of digestive enzymes in the tick \kur{Ixodes ricinus}

The intracellular proteolysis of ingested meal plays an essential role in tick development. The thesis focuses on the factors influencing the expressions and activities of digestive enzymes in Ixodes ricinus females during the feeding and post-feeding period. We have revealed the effect of fertilization on blood feeding and digestion. The females cannot reach the rapid engorgement phase without being fertilized. The rate of mated females in the nature proved the presumption that mating can occur even off the host. Implementation of in vitro feeding technique further extended our current knowledge about tick digestive apparatus. Adult females were fed on hemoglobin-rich and hemoglobin-poor diet and the mRNA expression levels of digestive proteases were determined. In line with obtained data, we assumed that albuminolysis is conducted by the same or similar pathway as hemoglobinolysis. The gene silencing method and protein immuno-detection were used to unequivocally identify the isoforms of 'early expressed' IrCL1 and 'late expressed' IrCL3 isoform of cathepsin L.
Cultivation of hops (Humulus lupulus L.) and its use

Hop differs from other agricultural plants in view of the fact that it is a perennial plant. It can grow at one site up to 20 years. There are three recognized hop production areas in the Czech Republic such as Žatecko, Úštecko and Tršice. The quality of the cones is high mainly due to the range of plant variety and also due to the favourable environmental conditions. Hop cultivation technology was already explored a long time ago, and therefore the new resources are rarely available and also widely scattered. The aim of this thesis is to describe the hops cultivation technology and its food and non-food utilization. This work includes botanical characteristics, farming techniques, fertilization, and protection against pests, harvest and post-harvest treatment. Furthermore there is description of the influence of agricultural engineering or fertilization on quality of hops. This bachelors thesis also depicts food and non-food use of hops.
The technology of cultivation of spring barley (Hordeum vulgare L.) and its use

The aim of my thesis was to introduce you to using research to the cultivation of spring barley (Hordeum vulgare L.). Cultivation of barley, I mean everything from the biological characteristics through soil preparation to planting and harvesting. For this search, I used books by Czech authors who write on this subject, as I used articles of foreign authors writing about issues influence of fertilization on spring barley, and I took advantage of practical experience with the cultivation of spring barley agronomists working in the company ZKS AGRO Zahořany. I tried to encompass the whole issue of growing spring barley and all carefully recorded.
Cultivation of Vital hop variety

Předložená metodika shrnuje zásady pěstování odrůdy Vital ve dvou progresivních pěstitelských systémech, v systému integrované produkce a v ekologickém zemědělství. Pěstitelský proces je veden snahou o prevenci a potlačení výskytu škodlivých organismů podporou užitečné entomofauny pomo-cí výsevu vhodných plodin v okolí či na okraji chmelnic. Navrhovaná opatření směřují ke zvýšení půdní úrodnosti,schopnosti půdy vytvořit optimální prostředí pro kořenovou soustavu rostlin chmele a během celé jejich vegetační doby jim zajistit dostatečné množství živin, vzduchu a vody pro jejich růst a vývoj, směřující k maximální produkci chmelových hlávek. Při vyhnojování chmelnic se preferují organická hnojiva, jako jsou komposty, statková hnojiva aj. Velká pozornost je věnována možnostem uplatnění zeleného hnojení a trvalého zatravnění, které je aktuální zejména na svažitých pozemcích. Ochrana chmele vychází z monitorování výskytu škodlivých organismů, prognóz signalizace vybraných chorob a škůdců a systémů varování. Ošetření pesticidy se provádí jen v nezbytně nutném rozsahu až po dosažení prahu hospodářské škodlivosti. Doporučované přípravky jsou selektivní s co nejmenším vedlejším účinkem na necílové organismy a životní prostředí. Zásady integrované produkce a ekologického pěstování chmele jsou aplikovatelné v klasických vysokých konstrukcích i systému pěstování chmele v nízkých konstrukcích.

The issue of transfer selected hybrids of the orchids genera, grown under aseptic conditions in vitro, into non-sterile conditions

In this paper experiments were made with three taxa of orchids. The first part was to find out whether the treatment of plants with potassium permanganate (KMnO4) has any impact on mortality during the transfer from in vitro culture to the unsterile environment of routine cultivation. All three taxa were divided into two parts and only one part was always treated while the other not. The deaths in individual groups were recorded during the experiment. In the second part the effect of fertilization on the biomass growth was investigated on two taxa. The method was again based on the division of the taxa into two parts and the fertilization of only one part. Necessary data were obtained by the weighing of plants at the beginning and end of the experiment. The results of the first experiment indicate that treatment of plants during the transfer has no impact on the subsequent mortality. In the second part, the dependence of fertilization on the biomass growth has been successfully proven in only one of the two taxa.
The influence of factors at produce starch of potato

The thesis is focused on the factors influencing the production of starch potatoes. These factors include the influence of variety, seed, soil, stress, fertilization, climatic conditions and weather. The work also provides basic information about potatoes, the history of the potato and its importance, the use of potatoes, the biological characteristics, morphology and chemical composition of potatoes. There is also an evaluation of the economics for potato starch processing.
Effect of eutrophication on primary production of a herbaceous wetland

This work is part of a study of wet meadows within the project GA CR 526/09/1545. The objective of the project is to determine the importance of newly assimilated carbon for the plat-soil interactions of plants with in wet grassland ecosystems in changing environmental conditions. As part of this project, a wet grassland ecosystem near Hamr situated in the Nežárka river floodplain was assessed in terms of aboveground production. This work includes data from 2010 and 2011. Each year the biomass was sampled two times. At each sampling, 24 samples were collected from plots differing in the intensity of fertilization. The treatments included high intensity of fertilization, low intensity of fertilization and no fertilization. In 2010, the annual production of aboveground biomass was 863.88 gm-2 on plots with a high intensity of fertilization, 788.46 gm-2 on plots with low intensity of fertilization and areas 839.69 gm-2 on unfertilized plots. In 2011 the annual production of aboveground biomass was 1149.71 gm-2 on plots with high fertilization, 953.73 gm-2 in plots with low fertilization, and 930.25 gm-2 on plots without fertilization.
