Time Lapse from Stationary Camera Image
Turek, Lukáš ; Bařina, David (referee) ; Herout, Adam (advisor)
The topic of this master's thesis is the time lapse from stationary camera images. Unwanted phenomena, which arise in time lapse, were analyzed and algorithms to overcome these limitations were designed. The algorithms were implemented and compared using the captured dataset. The resulting application creates time lapse from the video input and allows users to choose the processing technique including the setting of appropriate parameters.

Parameter Optimization of Multi-Level Diffraction Gratings
Matějka, Milan ; Kolařík, Vladimír ; Horáček, Miroslav ; Král, Stanislav
Originally, the e-beam lithography (EBL) is a technique for creating high-resolution black and white masks for the optical lithography. Multi-level relief structures can be also prepared using EBL patterning. Their preparation is based on the image patterning with a gradient of exposure doses. Large-area multi-level structures can be effectively prepared using the electron beam pattern generator with a variable shaped beam. We present several writing strategies. Basically, the main writing strategy uses one stamp (i.e. one elementary exposure of the shaped electron beam) per one elementary area with the same exposure dose. This simple approach is fast and flexible, however it does not guarantee optimal results. The main problem is an imperfection of the stamps (size, shape, and homogeneity). Advanced algorithms are based on multiple\nexposure of the same elementary area, the total local exposure dose is a sum of several different elementary exposures (stamps). Using these algorithms, a smoother surface of the structure can be achieved. On the other hand, the writing speed is considerably decreased. Tradeoff between the achieved parameters and the writing speed is discussed for selected set of writing strategy algorithms.

V případě sledovaného problému to znamenalo použití dat z dostatečně věrohodných zdrojů (skupiny Křehký lom ÚFM AVČR) a se známou metalurgickou historií.

Estimation of Genetic Parametrs in Selected Reproduction Traits of Sheep and Revision of Existing Evaluation of Animals
Schmidová, Jitka ; Vostrý, Luboš (advisor) ; Jan, Jan (referee)
The objective of this investigation was to select the proper model for genetic parameter estimation, to estimate genetic parameters, and to predict breeding values for litter size in most common sheep breeds in Czech Republic. A total of 143 896 lambing records from 1990 to 2012 were analysed. Variance components and genetic parameters for litter size were estimated separately for each breed using the BLUP animal model with repeatability. The basic model equation contained ewe age as a fixed effect and random effects of contemporary group, permanent environment and direct additive genetic effect of the animal. Modifications of the basic model were examined when various combinations of mating effects were included (contemporary group of ewes during mating (harem), additive genetic and permanent environmental effect of service ram). Estimates of phenotype variance increased across breeds (0.236 for Šumava to 0.779 for Romanov) with increasing breed average for number of lambs per litter. Variance component estimates for permanent environmental effect of the ewe were low (0.0001 to 0.0262). The variance of common environment of contemporary group (from 0.0223 to 0.1309) had bigger influence on the total variability of litter size in almost all studied breeds then additive genetic variance (from 0.0146 to 0.0587). The lowest heritability and repeatability estimates were for the Šumava (h2=0.0619; w2=0.0823) and Romney breeds (h2=0.0626; w2=0.0811); while the highest were for Merinolandschaf (h2=0.1091; w2 = 0.1129). The effect of service ram ranged from 0.01 to 0.02 of phenotypic variance in Šumava sheep, in Romanov sheep it was from 0.05 to 0.10, and in Suffolk sheep it was from 0.04 to 0.05. Including effects of mating (service sire, harem, and/or ram´s permanent environmental effect) in the model decreased deviance information criterion, what means that these models are more proper than the basic one. Results from present study demonstrate that genetic parameters did differ among the investigated breeds, which should be taken into account in breeding value estimation. The service rams have low but a clearly detectable influence on litter size of their mates. Genetic parameter estimates indicate that direct selection on the service ram effect could increase litter size and achieve genetic gain through ram selection.

Strnad, Štěpán ; Sýkora, D. ; Vrkoslav, Vladimír ; Cvačka, Josef ; Maletínská, Lenka ; Pirník, Zdenko
Mass spectrometry imaging is a powerful technique suitable for visualization of the distribution of a wide variety of compounds within tissue sections. The main aim of the study was the development and optimization of a sample preparation procedure allowing determination of the distribution of orally dosed metformin in mice kidney sections. Metformin is the first-line medication for the treatment of type 2 diabetes. The optimization of the sample preparation step before imaging experiments included the selection of a suitable matrix and the optimization of various parameters of MALDI analysis. 2,5-dihydroxybenzoic acid was identified as the best matrix providing highest sensitivity. A sublimation method was successfully used for the matrix deposition. The highest relative concentration of metformin was found in the inner zone of kidney 30 minutes after the drug administration.

Flow Parameters Simulation Technique in Annular Turbine Cascade INLET
Jelínek, T. ; Němec, M. ; Uruba, Václav
This article summarizes the concept of equipment which produces the required flow parameters at the inlet of annular turbine cascade. The investigation of inlet flow parameters - the boundary layer on hub and tip inlet channel end-walls and the inlet flow turbulence are part of a project intent on a flow in multistage steam turbine arrangement. This variable inlet can produce the laminar or turbulent boundary layer, respectively the laminar or fully turbulent velocity profile in the inlet channel. The change of inlet turbulence level is controlled by the grid. The detailed measurement of inlet flow: the turbulence and the total pressure distribution across the height of inlet channel were performed by HWA (hot wire anemometry) probe and by total pressure probe, respectively. The measurement results characterize the inlet flow for several inlet channel arrangements and are used for the experimental investigation of the flow field inside the stage and as the inlet parameters for CFD simulations.

On the recirculation zone suppression behind HUMP profile using the DBD actuator
Procházka, Pavel P. ; Uruba, Václav
Previously, the DBD (dielectric barrier discharge) plasma actuator was used in rectangular channel to modify the properties of the boundary layer in spanwise and in streamwise orientation. The actuator was redesigned for using on the surface of the Glauert-Goldschmied body in different position to influence the point of the separation and the reattachment point as well as the total extent of the separation bubble. The most intensive effect occurs when the actuator takes effect in the point of separation. Further downstream, the ionic wind produced by DBD causes complex coherent structures in the wake for spanwise orientation in both direction. The effect of streamwise orientation is studied also. Actuator is operated in steady regime to produces continuous ionic wind as well as in unsteady regime when the vortex street is generated. The properties of vortex street is given by modulation parameters (frequency and duty cycle). The effect of these parameters on the wake is evaluated. This experiment is realized in the perspex channel connected to the blow-down wind tunnel. The HUMP profile is flush-mounted to the bottom side. The wire electrode of the actuator is situated in x/L = 0.63, 0.66, 0.69 and 0.72 of the chord length. The time-resolved PIV (Particle Image Velocimetry) is used as a main measurement technique. The flow field behind the profile is captured in longitudinal plane as well as in cross-section planes using 3D PIV. The results based on statistical quantities will be presented in this paper. More, next part will be devoted to the decomposition analysis of the flow dynamics (BOD, OPD).
