Crystallization, structural and modeling studies of photosynthetic membrane protein and water soluble haloalkane dehalogenase

This PhD thesis is devoted to study of a) water soluble protein haloalkane dehalogenase DbeA (wild type enzyme) and its mutant variant DbeA1 from bacteria Bradyrhizobium elkani USDA94 and also b) the inner membrane protein complex of Photosystem II (PSII) from Pisum sativum L. An active site architecture of the haloalkane dehalogenase enzymes DbeA, DbeA1, stabilization by loop insertion in a mutant form, and structure-function relationships of haloalkane dehalogenases have been studied by using experimental approaches protein crystallization and crystallography. The protein crystals have been grown and X-ray diffraction data on these crystals have been collected and 3D molecular structure has been solved for both dehalogenases DbeA and DbeA1 at 2.2 A? resolution, for DbeA crystals soaked with 1-fluoropentane as a substrate at 1.65 A? resolution, and with 1,3-dichloropropane substrate at 2.15 A? resolution. Crystallization and molecular modelling methods were applied on membrane protein PSII core complex from higher plants in order to find optimal conditions for the 3D crystals formation, to establish the crystallization protocol and to study light-induced processes in PSII reaction center.

Fiscal rules in the Czech republic after joining the EU

This thesis concerns fiscal rules which represent a possible solution of excessive deficits of public budgets. The chosen topic is very relevant as most developed countries are now facing the problem of fiscal imbalance. The aim of the thesis is to analyze existing rules in the Czech Republic and to formulate recommendations for its public budgets with regard to the specifics of the economic and political environment. The main hypothesis is that fiscal rules in the Czech Republic are not well designed and do not contribute to the stabilization of public finances. This hypothesis was partially proved true. Proposal to strengthen the fiscal framework by implementation of the reform of medium-term expenditure frameworks and by adopting debt rule at the constitutional level results from the realized findings. Further recommendation is to establish independent fiscal institution that would oversee the fiscal policy in the Czech Republic.

Localization and function of small protein subunits of the cyanobacterial Photosystem II complex

The Photosystem II complex (PSII) contains besides D1, D2, CP43, CP47 and several lumenal extrinsic proteins also a large number of small intrinsic subunits. These subunits are found in all kinds of oxygenic phototrophs and their sequences are usually highly conserved. It is assumed that the small subunits are involved in stabilization, assembly or dimerization of the PSII complex but their exact function remains largely unknown. Thus, we chose four small PSII proteins: cytochrome b-559, PsbH, PsbI and Psb28, and we studied their function and location in PSII of the cyanobacterium Synechocystis sp. PCC 6803. We attempted: (i) to better define the role of cyt b-559 in the assembly of PSII using various mutant strains lacking one or more PSII core protein subunits; (ii) to clarify the location of the PsbH subunit using NTA gold labelling methodology; (iii) to improve the available knowledge on function of the PsbI protein in the assembly and repair of the cyanobacterial PSII; and (iv) to confirm the presence of the Psb28 protein in PSII, to localize it and to establish its importance for the cyanobacterial PSII.

Determination of Parenthood Determination of parenthood is very actual topic due to the constant changes of the traditional understanding of the family and family relationships. It is primarily the development of modern medical science, biology or genetics that has brought new concepts into the issue of determination of parenthood. For example surrogacy, assisted reproduction or baby boxes. The legislation must responds to these changes in order to stabilized family relationships and to ensure their protection. The aim of this thesis is to describe and to analyze Czech legal regulation of determination of parenthood and to compare it with not only European but also global legislation. The analyse of valid legal regulation highlights questionable passages that may cause difficulties in its interpretation or practical application. The thesis is composed of nine chapters, each of them dealing with different aspects of determination of parenthood. Opening chapter introduces the historical development of relationships between parents and children in Roman and European law. It focuses on legal standards applicable in our country during the period of time from the establishment of the Czechoslovak Republic to the present times. Mate choice unter the influence of hormonal contraception: impact on partnership stability and quality

Previous studies have pointed out an interesting fact: the use of hormonal contraceptives modifies the partner preferences of women towards men more feminine and genetically less compatible. This master thesis first tried to experimentally verify whether these different preferences are reflected in real mate choice outside the context of the laboratory. It includes two studies comparing partner satisfaction and relationship dynamics in couples that have been formed under the influence of pills versus under the influence of the natural menstrual cycle. Study 1 includes 3116 mothers and was conducted through an online questionnaire. The second study collected representative data from 1605 Czech men and women aged 35 to 65 years representing the population of the CZ also through a hand-to- hand questionnaire. Study 1 confirmed the assumption in several ways: hormonal contraception users (during the mate choice) assessed their partners as less attractive and masculine compared with women who naturally cycled at that time. We also found their sexual satisfaction was reduced, however, they were more satisfied with non-sexual aspects of the relationship. - Institutional Arbitration Arbitration in its today's dimension is a phenomenon that represents an alternative system to a state organized litigation. Arbitration is way of settling disputes among subjects in various fields of economic activities. There is commercial arbitration which is the most spread and used in the world, sport arbitration, mixed arbitration and others. This thesis, also due to the fact that it has been assigned by The Department of Commercial law - International Private Law, is devoted to international commercial arbitration. Arbitration is a private system of litigation in which private neutrals are called by parties to resolve their dispute. Arbitration held by individual neutrals chosen particularly for determinate cause is called ad hoc arbitration. In response to a stabilization of arbitration procedure, the arbitration institutions were established to provide arbitrational services and other dispute resolution services. Arbitration administrated by such institutions is in theory called institutional arbitrational, which is the subject of present work. The basic mission o f the state is to create the conditions for the stability and to regulate the social climate, to absorb the social tension and by the development o f social activities to stabilise the situation in the society in order to enable the social subjects fully develop, affect and realise their targets by their own forces. These efforts intend to establish the conditions for the social defensibility of the social reform and/or minimize its impacts. In order to assure this target, the state disposes by the wide system of legal, financial and institutional instruments. The aim o f this thesis is the global evaluation of the actual legislation on the security of the people in socially indisposed situation; this people enters under the application of the Act No 117/1995 Coll., on social state aid, Act No 108/2006, on social services, Act. No 110/2006 Coll. about the existential minimum and Act No 111/2006 Coll. on aid in material necessity. Nursing care as provided to ante finem patients

The subject of death and dying is still considered a taboo in our society. It is awkward and anachronistic since our times require people full of life and vigor. The terminally ill are often snubbed by the society. Though being well aware of the unfavorable prognosis, many a doctor tries to treat the patients to the very last moment, and he or she perceives the death as a personal defeat. In 1967 Cicely Saunders, an English doctor familiar with the dismal conditions of dying in the then society, founded in London the St. Christopher's Hospice, the first modern facility of this kind. The philosophy of this effort has been perpetuated in the Hospice of the Agnes of Bohemia established in 1995 as the first hospice in the Czech Republic. The Bachelor Paper aimed to assess the possibilities of providing the ante finem care by health-care personnel at a hospital and a hospice. The Paper investigated these hypotheses: H1 - Health-care workers provide the ante finem patients with health care in conformity to the standards applicable to both types of the facilities; H2 - Approach of the personnel to the care is dependent on their emotional stability; H3 - Health-care personnel in hospitals are not aware that the art of accompanying is a part of the nursing process. The surveyed sample was compiled of nurses on the staff of the Internal Medicine Ward and the Post-Treatment Ward of hospitals in České Budějovice, Jihlava and Třebíč and the hospices in Prachatice, Červený Kostelec and Brno. The data were gathered through a questionnaire. Altogether 170 questionnaires were distributed and 136 out of them were processed in the final assessment. As follows from the assessment results, the care provided to the terminally ill patients in hospitals is affected primarily by time pressures and by the fact that the nursing staff are not fully aware of the possibilities that palliative care offers, while in hospices the conditions available for the palliative care have a quality basis. Results of the Paper will be given to one of the hospitals and two hospices specified above, where they can inspire some new ideas about possible improvement in providing the palliative care.

The Comparison of the Czech and Slovak Governments between 1992 and 2002

The Master thesis "The Comparison of the Czech and Slovak governments between 1992 and 2002" deals with the problem of the establishment of governments, the main coalition and oposition parties and the main situations of political life of Czech and Slovak governments in this period. The thesis is divided into three chapters. Chapter one is an introduction to the topic and focuses on the process of constitution development in both countries, which was taking place in the period of the Federation. The second chapter covers the period from 1992 to 1998 and the third chapter 1998 to 2002. The following section constitutes the comparison of these specific periods. The main comparison parameters are: the stability of the party systems, the consolidation of the society, and the foreign policy development. The Czech Republic appears to be better in this excercise. It's party system is far more stabilized and it's society is much more homogenic. As argued in the thesis, this result is mainly due to the lack of any anti-democratic and authoritative element on the political scene, as it was in the period of Meciar's government in Slovakia. The aim of this diploma thesis Transforming of Executive Coalitions in Prague in the Electoral Period 2010 - 2014 is to analyze influences, which have an important role in a process of establishing executive coalitions and also in their stability. The analysis is based on a classical theory of coalitions, discursive analysis, interviews with direct participants and on official documents. One of the aims is to describe the level of possibility in application of theoretical concepts of coalition theory in the area of Prague's political system. There are many points of view, from which we look at stability and forming of coalitions. For example, very important point of view is an influence of making and implementing politics or an influence of politicians from national level. A big importance is focused on negotiating between participants, their statements in media and their real steps and decisions. The intention of this thesis is not an invention of general model which should be applied on similar local system, but to explain and describe the uniqueness of Prague local system and the election period of 2010 - 2014.
