Separation and determination of pharmaceutically important compounds using electrophoretic methods

This thesis deals with the use of electrophoretic methods in the analysis of pharmaceutically important compounds. In the theoretical part, the classification of capillary electrophoretic techniques is discussed. Capillary zone electrophoresis (CZE, capillary isotachophoresis (ITP), micellar elektrokinetic chromatography (MEKC), microemulsion elektrokinetic chromatography (MEEKC), isoelectric focusation (IEF), capillary electrochromatography (CEC) and capillary gel electrophoresis (CGE) are described. Further, on-line pre-concentration methods are described. Particular attention is given to the on-line combination of capillary isotachophoresis with capillary zone electrophoresis using column coupling arrangement. This combination enables the improvement of the selectivity of separation and the improvement of the limit of detection of analytes present in complex matrices of biological and environmental origin. Afterwards, the results of the experimental analyses are presented: 1. The first paper deals with the use of CZE in the analysis of ibuprofen and flurbiprofen in pharmaceuticals. Separation was carried out in a fused silica capillary (60 cm x 100 m I.D effective length 45 cm) at 30 kV with UV detection at 232 nm. Pressure loss valves in heating systems

The theoretical part of the thesis deals with the hydraulics of heating systems. The aim of the project is a design of the heating and hot water preparation system for an apartment building. The building has 6 above-ground floors and 2 underground floors. Two different options of heating systems were designed, comparison and results are presented in the thesis. In the first case the heat source during the winter season and for water heating, where chosen two gas condensing boilers, situated in separated room on the second underground floor. In the second case, each apartment has its own gas condensing boiler. The experimental part of the project deals with measurements of pressure losses in water armatures used in heating systems. The aim of this part is the determination of coefficients of added resistance for uncontrolled elements, which are normally presented as tabular value. In thesis are presented results for a ball valve, backflow preventer, filter, for slide valve and thermostatic valve. Measurements for thermostatic valve where done with different settings and results are compared with parameters listed in the datasheet.

Laboratory diagnostics of micrometastases in breast cancer patients

Introduction: The presence of circulating tumor cells (CTC) in the peripheral blood has been associated with worse prognosis and early relapse in breast cancer patients. CTC determination in the peripheral blood has been considered as a liquid biopsy. The aim of this project was to analyze the presence of CTC followed by their molecular characterization with the potential use not only as a new biomarker for real-time monitoring of therapy efficacy but also as a suitable tool for patient's stratification and individualization of treatment for breast cancer. Methods: A total of 54 patients with diagnosed early breast cancer were enrolled into a prospective study. Ten millilitres of peripheral blood were sequentially collected to test for the presence and characterization of CTC during the follow-up of patients. CTC isolation and detection was performed by AdnaTest BreastCancer™ (AdnaGen AG, Germany), which is based on the detection of EpCAM, HER2 and MUC1 specific transcripts in enriched CTC- lysates. cDNA from isolated CTC has been further used for newly optimized qPCR assays for breast tumor and therapy resistance associated genes: TOP1, TOP2A, CSTD, ST6GAL, KRT19 and reference gene actin. qPCR results have been analyzed by Genex software (MultiD Analysis). From machine to icon:car in popculture from the 60s

An automobile is one of the most important phenomenons of modern technology. Since its origins, it has profoundly shaped our physical, economic, politic, social, cultural and media habitat. The objective of this Master thesis is to analyze the evolution of the role of the car in the popculture from the 60s onwards and its dimensions, with a primary focus on Anglosaxon culture, with excursions to Asia and continental Europe, primarily Czechoslovakia/Czech Republic. The core foundation of the thesis lies in the analysis of the popcultural genres, selected films, TV series, computer games and comic books. The main purpose of the thesis is to reflect upon the identity and conceptual shifts of car, going from a mere transport machine through a vehicle of product placement towards the concept of the icon, a narrative hero of popculture. The hypothesis to verify is that the car has become an essential part of the popculture itself, reflected in many various dimensions: car as a toy, car as a friend, car as a status symbol, car as a narrative extension of the hero and finally, car as a hero and subject. Cross-border cooperation between Czech Rebulic and Slovakia during 2007-2013 programming period 2007-2013

As the time goes by, one former Czech-Slovak territory has separated, forming two independent republics and members of European union. Division of common territory by a border, however, cannot affect cultural and linguistic proximity, which can be clearly identified in cross-border regions. The purpose of this thesis is to evaluate the cooperation amongst six regions located near mutual border during 2007-2013 period. By the means of publicly available content, an analysis of allocation of financial resources from ERDF and national budgets is executed. The qualitative evaluation of cross-border cooperation projects is delivered by the proposal of matrix for territorial impact assessment. According to percentual fulfillment of given goals, impact on barrier effect reduction and selected projects analysis, the overall added value of cross-border cooperation between Czech and Slovak republic during 2007-2013 period is specified. Main findings are then a result of combination of qualitative analysis and quantitative indicators for the researched period.

Valuation of Limited Liability Company

The main goal of the thesis is to find the market value of the limited liability company as of the date 31. 12. 2014. Companys business activities are divided into four divisions : accumulators and batteries, measuring equipment, security systems and solar systems. Financial analysis is conducted for the enterprise as a whole, strategic analysis is due to the different sectors in which the individual divisions operate handled separately for each division. Based on the results of the financial and strategic analysis, the valuation method of the discounted cash flow variant entity was used to determinate the estimated market value. The prognosis of the value drivers and financial plan are also parts of this thesis.

Portable and Miniaturized Separation Techniques Applicable for Food and Biotechnology Analysis

Capillary electrophoresis was used for determination of 6 fractions of caseins. Those fractions were measured in 144 samples of cow’s milk originated from the feeding experiment focused on explanation the influence of the feeding onto casein productions. In this work were separated 6 fraction of caseins first time with total resolution of the peaks. Capillary electrophoresis was applied for determination of short-chain organic acids during fermentation of wine must. It was compared the fermentation of must fermented by different yeast. The difference of profile short-chain organic acids during fermentation were not statistically significant. The once difference was in the utilisation of the malic acid and production of the lactic acid. A portable miniaturized system for medium pressure liquid chromatography was developed. The components were tested and system was used for the isocratic and gradient elution of various analytes (food dyes, parabens). New line of electroluminescent diodes (LEDs) for deep-UV areas of wavelength based on a different materials substrate was characterised. The new line was compared with old line LEDs. The new line LEDs was incorporated in deep-UV absorbance detectors. Detectors were characterised and tested for a detection various analytes in modes flow injection analysis and chromatography separation. First time was characterised this new line of the LEDs and the origin of the parasitic emission band produced by deep-UV LEDs light sources was explained. This origin is given by disturbances of a materials substrates. This work is a contribution for an advance of low-cost and portable systems and detection devices in the field of analytical chemistry.
