Applications of chiral and achiral chromatography in pharmacology and toxicology

Development and validation of methods for analysis of several drugs or their metabolites are decribed in this thesis. The document is presented as a commentary to the original papers, which were published in peer reviewed journals. Discussion on the optimization of each method is presented and covers also method development and influence of preanalytical aspects. Additionally, examples of the application of the developed methods in clinical pharmacology and toxicology are shown. This dissertation consists of three parts: enantiomeric determination of tramadol and its metabolite, determination of some antihypertensive drugs, and qualitative analysis of benzodiazepines. Development of a method for chiral analysis of tramadol and its desmethylated metabolite O- desmethyltramadol (ODT) in human urine and plasma is described in the first part of the thesis. Tramadol is a centrally acting analgetic drug, which is used as racemate in clinical practise. Each enantiomer displays different binding properties for various receptors: (+)-tramadol preferentially inhibits serotonin reuptake while (-)-tramadol mainly inhibits noradrenalin reuptake. The theoretical part of the Master´s thesis describes steroids, their features, and the methods for their determination in waters. In experimental part three steroid hormones, namely 1,4-androstadiene-3,17-dione (ADD), 1,4-androstadiene-3-one-17(beta)-ol (boldenone) and 17(beta)-estradiol were selected. The compounds were concentrated from water by LLE and SPE extraction and determined by gas chromatography coupled to mass spectrometry. Several experiments were conducted to assess the recovery of specific extraction for studied compound.
Biochemical methods as tool for study of reproductive proteins

Study of molecular mechanisms in reproduction is essential for the understanding of this outstanding process. Our lab studies proteins secreted by reproductive organs and sperm using various biochemical methods for a long time. We have expertise in protein extraction from spermatic cells using different approaches, and by kits for proteins from the sperm surface and distinct subcellular compartments. The proteins of reproductive organ fluids are separated by chromatographic methods, such as size exclusion chromatography, high-performance liquid chromatography with reverse phase (RP-HPLC) and affinity chromatography on matrices with various ligands. Proteins are subjected to SDS- or 2D-electrophoresis for their characterization and comparison of various extraction methods, different mammalian species, and sperm in different functional development. Electrophoretically separated proteins may be transferred onto nitrocellulose membrane (Western blot) for antibody detection or binding studies with lectin-labelled ligands (lectins, polysaccharides, zona pellucida glycoproteins). We use immunoprecipitation method with specific antibody for protein determination followed by the MALDI identification. Proteins are localized by immunofluorescent techniques on/in spermatic cells and tissue sections of reproductive organs. Isolation of proteins from reproductive tissues and fluids, and the antibody detection is crucial for the studying of reproductive protein origin.
Diagnostics Methods of Dissolved Gas in Transformer Oil Observation

This thesis is devoted to analysis of the diagnostic methods used in practice. It is divided into two parts: theoretical and practical. The theoretical part concerns with the general description of the transformers. Further it provils informatik about systems for sampling oil from transformers with insulating oil – paper system. Another important part is the description of gas chromatography and TRANSPORT X. Description of the most appropriate evaluation methods for assessment of the state of the transformer is included as well.
Analysis of selected nutritional parameters in the new varieties of gooseberries

Diploma thesis deals with the new varieties of gooseberries. The theoretical part characterizes gooseberry from a biological point of view, what is the content of healthful nutrients and states food technology aspects. Furthermore, the theoretical part deals with the main parameters examined nutrition: vitamin C, anthokyanům and antioxidant activity. In the experimental part was measured both weight solids content and soluble solids. Vitamin C was measured by HPLC, anthocyanins using a pH differential method and for antioxidant activity determination was selected method by Kanedy known also as DPPH.
Plasmid DNA vaccines

Plasmid DNA vaccines are the new generation of vaccines with a great potential in prevention of many diseases. Recent studies and clinical test are aimed at prevention against cancer, hepatitis, malaria, HIV, influenza and other diseases. Recent main challenges covering plasmid DNA vaccines are associated with optimalization of each step of production and mainly purification steps allowing production of pDNA at kilogram levels. Main purification techniques used are based upon chromatographic methods, but research and development also shows other potential methods, like two-phase aqueous systems or magnetic microparticles as carriers. In experimental part of this thesis isolation of pUC 19 plasmid from Escherichia coli JM 109 (pUC 19) cell culture was performed via method of alkaline lysis. Isolation was verified by agarose gel electrophoresis. Isolated samples were purified in four repetitions with lithium chloride and magnetic microparticle carriers and the extent of purification was verified spectrophotometrically. Purified samples were visualised via agarose gel electrophoresis and results were compared.
Substituted polyaromatic compounds in environmental components

This work is focused on substituted polycyclic aromatic hydrocarbons (PAHs) in environmental compartments. The thesis examined the development of these compounds and their entry into the ecosystem. Due to their proven negative characteristics to be addressed also their toxicology. The next section discusses the appropriate methods, including sampling, sample preparation and final determination. The conclusion of this work forms draft standard operating procedure for their determination.
Non-destructive diagnostics tests of transformers

Positive effects of conjugated linoleic acid on human health

This bachelor thesis deals with the characteristics of the fatty acids, their structure, nomenclature, properties, reactions and analysis. It is also dedicated to the history, sources, effects on human health and the correct ratio of omega-fatty acids in the body. The main issue of this work is conjugated linoleic acid, which is known to its countless positive effects, which can include weight loss and lower level of cholesterol, anticarcinogenic effects, prevention of osteoporosis and arteriosclerosis, etc. The work is focused on its characteristics, structure, occurrence in food, chemical production and a wide range of preparations available on the market. Last but not least, deals with the possibilities of its determinations, including gas chromatography, high performance liquid chromatography, gas chromatography-mass spectrometry, NMR spectroscopy, and combined methods.
Problems of phenols in water

This bachelor`s thesis deals with the issue of phenols in the waters. The theoretical part summarises the information about the characteristics of the phenols, its use, appearance in the environment, and its effects on human health. There are described methods of determination these substance in the waters. The last chapter disserts on possible methods of eliminating the phenols from the aqueous environment with special attention paid to the adsorption method. Part of this thesis is also dedicated to the chlorinated phenols which originate from chlorination of the phenolic waters and which cause the sensory interferences of the waters. The experimental part of this thesis is dedicated to removing the phenol from a model water through the adsorption into a powdery active carbon Norit SA SUPER.
