Optimization of production and distribution in bakery

The subject of this bachelor thesis is optimization of production and distribution. The thesis aims to introduce the production problem in the case that the production limits are known and present its solution using a modified simplex method with lower or upper limits. The vehicle routing problem with one distribution centre and one vehicle will then be introduced. In the practical part of the thesis, these methods are applied in real-life data from the Martínek bakery business. The optimal production, which is targeting to maximize the profit under the given terms, and the optimal distribution from the bakery to shops, will be determined. At the end of the thesis, there is an evaluation of the given targets.

Voltammetric Determination of Trace Amounts of Genotoxic 2-Aminofluoren-9-one

of the Diploma Thesis Presented Diploma Thesis is focused on the electrochemical determination of genotoxic 2-aminofluoren-9-one (2-AFN), which belongs to derivatives of polycyclic aromatic hydrocarbons. Its possible carcinogenic and mutagenic effects may represent a risk to living and working environment. Optimal conditions for the voltammetric determination of 2-AFN were found in an appropriate ratio of buffered aqueous and methanolic phase. In the methanol - Britton- Robinson (BR) buffer (1:9) medium, voltammetric behavior of 2-AFN at a concentration of 1×10-4 mol/l was investigated in a dependence on the pH of the solution in the range from 2.0 to 13.0. For following measurements of concentration dependences, the optimal medium was chosen to be methanol - BR buffer of pH 4.0 (1:9). The achieved limits of quantification (LQ) were 2×10-7 mol/l for DC voltammetry (DCV) and 1×10-7 mol/l for differential pulse voltammetry (DPV) at a hanging mercury drop electrode (HMDE). In the medium of an acetate buffer of pH 4.0, the LQ attained using adsorptive stripping differential pulse voltammetry (AdSDPV) at the HMDE under optimal conditions for the accumulation of the analyte (Eacc = +100 mV and tacc = 120 s) was 4×10-9 mol/l of 2-AFN. Use of encapsulation techniques for production of food for infants

The Diploma thesis deals with use of selected probiotic strains Lactobacillus acidophilus and Bifidobacterium breve in different forms in food for infants. The theoretical part is focused on describing probiotics, encapsulation methods and intestinal gut microbiota of infants. Further, characterization of individual periods of infant feeding and food for infants were introduced. In experimental part the possibilities of encapsulation and lyophilisation of probiotic cells were observed. Probiotic cells were encapsulated into alginate particles. The encapsulator was used for preparation of particles and the most appropriate particles were prepared by encapsulation nozzle with size of 300 µm. Moreover, probiotics viability was monitored by Flow Cytometry, Fluorescence Microscopy and by cultivation (CFU method). Viability of probiotics was monitored during long-term storage in selected food for infants. The appropriate shelf life of non-lyophilized alginate particles in real food have been set at 1 to 2 months. Lyophilized alginate particles could be stored for more than 3 months. Finally, the stability of the particles and viability of encapsulated and non-encapsulated cells in the gastrointestinal tract conditions were also examined. The viabilities of lyophilized cells and cells encapsulated in lyophilized particles were also compared. From the results obtained, non-encapsulated probiotic bacteria cells are more susceptible to negative effects of digestive juices, the percentage of dead probiotic cells after digestion was approximately 80 %. On the other hand, alginate particles showed cell protection from digestive juices, after incomplete cell releasing from particles the percentage of dead probiotic cells did not exceed 20 %. After adequate rehydration, similar results were gained with lyophilized alginate particles. Lyophilized alginate particles have been determined to be the most suitable application form for infants’ food.

Vyhodnocení a analýza zaplevelení vybraných polních plodin v provozních podmínkách

This thesis is focused on weed infestation evaluation of winter oilseed rape and winter wheat growths on selected land of ZD Kelečsko Kelč, in years 2011 -- 2013, futhermore on assesments of weed infestation differences between crops and particular years. Weed infestation was detected by numerical counting method and evaluation were processed by DCA analysis. Leftover of previous crop appeared the most frequently in winter oilseed rape, with weeds from the family Brassicaeae - Thlaspi arvense, Viola arvensis and Capsella bursa -- pastoris followed. 22 weeds and weedy species were found in total. Galium aparine, Viola arvensis, Veronica spp. occured the most often in winter wheat. 21 weeds and weedy species were found here. The growth of winter oilseed rape were more weedy than wheat.

Identification and determination of metalloproteins and metalloproteinases in experimental models and in groups of patients

Introduction: Matrix metalloproteinases (MMPs) are zinc-containing metalloproteins which take part in many processes associated with extracellular matrix (ECM) remodeling. These enzymes participate in most processes degrading connective tissue during ontogenesis. Changes in MMP expression and activity can be observed in most inflammato ry, degenerative, and malign processes. Methalothionein also belongs to the group of zinc-dependent metalloproteins and it is involved in metal trafficing, detoxification or protection of cells against reactive oxygen species. Aim: The aim of th is study was to analyze MMP and metallothionein in various experimental models of caridac tissue remodeling (under hypoxia and methamphetamine administration) and in groups of patiens with dislipoproteinemia and traumatic brain injury. Methods: Laboratory rats were exposed to 1) hypoxia for 3 weeks and treated with MMP inhibitor 2) hypoxia and hypoxic hypercapnia for 4 days. 3) ln another experimental set methamphetamine was aplicated to rats for 9 weeks. 4) A group of patiens with dyslipoproteinemia was investigated before and after 1 month therapy by diet or hypolipidemics. 5) Blood samples were collected from patiens with traumatic brain injury during hospitalization. The aim of this diploma with the topic "Strategic analysis" is to elaborate the global analysis of the agricultural company ZEMPO-VOS, a.s. Strunkovice nad Blanicí and on the grounds of this analysis, i. e. analysis of influences of outer company environment, to elaborate the synthesis and the outline of strategic recommendations for the future development of the company. The stategic analysis is elaborate in conditions of branches of agriculture that have unequivocal specific characteristics. The diploma is divided into two parts, the theoretical part and the application part, where methods of strategic analysis are applied to the concrete agricultural company.

Daňový únik pomocou fiktívnych dokladov

Tato bakalářská práce se zabývá daňovou optimalizací. A to jak daňovou optimalizací ve smyslu snahy o legální daňové úspory na jedné straně, tak i ve smyslu snahy o obcházení zákonů spojené s realizací nelegálních daňových úniků. V práci jsou uvedeny možnosti legální daňové optimalizace v podmínkách slovenského podnikatelského prostředí. Na příkladě skutečných kauz z posledních let je popsán daňový únik na spotřební dani z minerálních olejů a daňový únik realizovaný pomocí bílých koní založený na principu nadměrného odpočtu daně z přidané hodnoty z fiktivních faktur. Dále se práce zabývá dvěma skutečnými firmami. Podnikání jedné z nich je v souladu se všemi zákony, druhá firma se dopouští daňových úniků. Na příkladu první z nich je vysvětlen postup stanovení daňové povinnosti podle slovenské legislativy. Druhá firma nezákonně optimalizuje svůj základ daně a tím i svou daňovou povinnost. Touto nelegální optimalizací se dopouští trestného činu úmyslného krácení daně. Dopad daňového úniku na státní rozpočet je v práci přesně vyčíslen. Práce nám také poskytne názory jednatelů obou společností a pak cenné názory odborníků z daňové praxe na tyto dvě firmy. Z uvedených názorů jsou v závěru práce formulována stručná obecná doporučení pro zlepšení boje s hospodářskou kriminalitou.
