The application of electrochemical impedance spectroscopy for investigation of enzyme kinetics
Pospíšilová, Michaela ; Sedláček, Jiří (referee) ; Fohlerová, Zdenka (advisor)
The application of electrochemical impedance spectroscopy (EIS) in theory and experiment for investigation of redox enzyme kinetics has been already described. The impedance studies are quite sensitive to the changes at the interface caused by adsorption of charged species and therefore can be used to study the kinetics of adsorption of macromolecules and also enables us to study enzyme kinetics since the action of the enzymes on their substrates involved modification and generation of charged species. In this work, we would explore the potential of electrochemical impedance technique to follow the kinetics of glucose oxidasesubstrate reactions on the immobilized surface. The enzyme will be allowed to interact with different concentrations of its substrate and the resulting reaction will be recorded in real time. Changes in the imaginary component of the impedance at various substrate concentrations will be expected to follow MichelisMenten kinetics. The experimental procedure should be simple and can be carried out directly in buffer solutions most suited for the required interaction to take place, without the requirement of any additional redox probes.

Gated communities: residetial separation and privatisation of public space in Czechia
Brabec, Tomáš ; Galčanová, Lucie (referee) ; Sýkora, Luděk (advisor)
The diploma thesis is focused on the phenomenon of gated communities which represent the extreme form of residential separation. Gated communities are becoming the new and specific sign in present-day globalizing cities. The number of objects of gated communities is growing quickly all over the world, and now this issue appeared in the Czech Republic. But common or expert debate concerning the topic is not developed here. The main aim of this piece of work is to contribute to this debate, and at the same time, to be the constitutive empirical research applied to the Czech scientific discourse. This study intends to help with the overall recognition of gated communities. The work is primarily aimed at evaluation of development of housing construction, features, territorial extension, causes of formation and consequences of gated communities in the capital Prague. It means in the place with the highest concentration of gated communities in the Czech Republic; 59 of these objects were found there. Extensive construction began four or five years ago; since then gated communities have been available not only for the richest class. Compared to other countries, gated communities are localized mainly in the inner city, and there is a difference in the cause of formation: in these states it is the fear of...

Injury to the distal tibiofibular junction in the growing skeleton
Pešl, Tomáš ; Bartoníček, Jan (advisor) ; Koudela, Karel (referee) ; Krbec, Martin (referee)
In adults the complex fractures of the ankle joint have been thoroughly discussed and there are several therapeutic schedules proposed. Weber distinguished three types of ankle joint fractures according to the type of fibular fractures. In agreement with this classification we are certain that fibula is an important element of the immature ankle joint. We concentrated on the anatomy of immature ankle joint, especially on the mutual position of the distal fibular physis and the plane of the tibiotalar joint. The results are summarized as follows: 9.1. AUTOPSY At first we performed autopsy of six ankle joints in stillborns in which there were no orthopaedic and/or neurological lesions. We paid attention to anatomy of the distal tibiofibular junction. Two adult ankles were anatomised for correlation the findings. We ascertained, that in all preparations there was articulation between tibia and fibula, as it is formerly described in the literature. In all six stillborn joints we found well formed ligaments of the distal tibiofibular junction: anterior, posterior and interosseal tibiofibular ligaments. The anterior and posterior tibiofibular ligaments begin at the anterior, respectively posterior surface of the distal tibial epiphysis and terminate at the anterior, respectively posterior surface of the distal...

Discrete Surface Fairing
Matýsková, Martina ; Kostková, Jana (advisor) ; Suk, Tomáš (referee)
This thesis proposes a new method for fairing of surfaces, which are represented by a triangulated mesh. Motivation for our task is a 3D reconstruction, which results in a triangulated mesh. Such mesh is typically corrupted by noise (due to data and processing inaccuracy), which is required to be faired out. We solve the problem on a discrete surface, where two criteria for optimization are defined: surface curvature and non-uniformity. The curvature deals with surface smoothness, while the non-uniformity with surface regularity. The problem of fairing is solved by a discrete diffusion. Since our aim is to fair surfaces with boundaries, first we describe the fairing of curves, then fairing of closed urfaces, and finally fairing of surfaces with boundary (which is in fact a combination of the former ones). We have tested the algorithm's performance on synthetic as well as real data with good results. The experiments showed that our proposed algorithm is robust, stable and does not shrink an object, while its main surface characteristics are preserved.

The concept of the system state tourism policy Czech Republic with a focus on cultural tourism
Tittelbachová, Šárka ; Patočka, Jiří (advisor) ; Cimler, Petr (referee) ; Drdácký, Miloš (referee)
The focus of the thesis is an issue of system approach to formation and implementation policy of tourism in the Czech Republic with an emphasis on intersectoral cooperation in the field of cultural tourism. The work is divided into eight chapters, the first two are theoretical. The first chapter is devoted to the formulation and definition problem of the scope of the work. The second chapter deals with theoretical foundations and approaches to tourism policy issues both in general and in terms of the status of tourism in the national economy and in relation to the EU tourism policy. Attention is given to the objectives of cohesion policy, culture and tourism in the area of cultural tourism. The following two chapters deal with basic features, analysis and evaluation of policy concepts of tourism in the Czech Republic in the period after 1989. The core of the thesis is concentrated into chapter six, seven and eight. The assumption for the design of complex, dynamic tourism policy for the formation of functional institutional structure of the tourism sector management is to assess the fulfillment of the objectives of the different concepts of state policy. To identification the most important and recurrent problems of tourism policy are used the results of comparison of objectives and measures and practical experience of the author from the creation and evaluation of the conception documents. There is discussed in particular an evaluation framework and in this context there are suggested recommendations for its determination. In the field of cultural tourism is proposed a model of interdepartmental cooperation state-funded organizations CzechTourism and the National Heritage Institute. The proposal model is based on its own research, which is focused on the analysis and evaluation of assumptions monuments administered by the National Heritage Institute to participate in the creation of cultural tourism products. There are utilized data from the author primary survey and as well as secondary data from the National Heritage Institute. The model of cooperation is based on exemplification of the causal relationships between strategic objectives and processes within the strategies of both organizations by the model BSC (Balanced Scorecard) and from a proposal for their implementation using model 7 "S".
