Mapping, processing, and representation of a map for Unreal Tournament bots
Gemrot, Jakub ; Brom, Cyril (advisor) ; Yaghob, Jakub (referee)
In many computer games the navigation points are used for the movement of computer-controled players. With the growing power of desktop computers more sophisticated methods are used for the movement of players, namely the method of NavMesh. NavMesh is a set of convex polygons continuously covering the reachable places inside the game by characters. This approach was used in the recently produced games Half-Life 2 and F.E.A.R. Those commercial products can't be used by the academy, therefore the chance to experiment with NavMesh doesn't exist. But there is a simple way to control the characters in environment of older game Unreal Tournament. This work is about the advantages of using NavMesh against older approach of navigation points. This work then presents the way how to automatically generate the NavMesh for the maps of Unreal Tournaments. NavMesh is created using the algorithm from computer graphics for the conversion from CSG representation of the map into boundary representation out of which the NavMesh can be generated. The work also contains a demonstration of usage of NavMesh for the movement of players in Unreal Tournament's maps.

The role of the word of God in christian spiritual life
Olivová, Jana ; Kohut, Pavel Vojtěch (advisor) ; Ventura, Václav (referee)
RÉSUMÉ The problem of word of God in spiritual life of the Christian The topic of this work is Holy Scripture - Word of God - from the sight of spiritual life. What is the Bible for Christian? How it can be influent for our daily life? How the Church understood the Holy Scripture? Theoretic resource of this work is teaching of the Second Vatican council about word of God, especially constitution "Dei verbum". There were basic ideas of this teaching formed in Century 19th by biblical movement, which made an effort for more serious thinking in the faith through the come back to the source of the Holy Scripture. The principal points of the teaching of the Second Vatican council about word of God are followed: "The Scripture" is revelation of the God. The God reveals us in the Scripture his plan of the salvation which culminate by the coming the Son of God into the world. The Holy Scripture is inspired word of living God therefore the church has it in honest and love. The constitution "Dei verbum" speaks the table of the word of God in the same level as the eucharist. Through the Scripture we meet the God that is why it is for faithful "food" on the way to the salvation. The teaching of the Council had opened the way to the study of the Scripture based on the scientific level. It also encouraged lay people to...

One Side of the Hero- Interpretation of Cúchulainn's Story According to Campbell's Theory of Monomyth
Frühbauer, Jan ; Fárek, Martin (referee) ; Janák, Jiří (advisor)
The main intention of this work is an interpretation of a chosen mythical narration from the point of view of the theory of monomyth from american antropologist Joseph Campbell. In his well-known book The Hero of Thousand Faces he presents many myths and old legends from the whole wide world. In the way of comparison of the myths and the motives which they contain he demonstrates common spiritual grounds of the mankind. Campbelľ s approach is very familiar to the analytical psychology of C. G. Jung and he many times refers to him and his scholars. Currently he uses many of Jung' s terms, such as the idea of an archetype or Self (germ. das Selbst). Further, hismodel of monomyth of the hero carries resemblance to Jung' s image of the process ol individuation (germ. der lndividuationsprozess).

EU Budget as exemplar for solving problems of Czech public finances?
Maláník, Ondřej ; Štěpánek, Pavel (advisor) ; Chalupníček, Pavel (referee)
The main objective of this thesis is to point out to situation of Public finance in Czech Republic and analyse it from perspective of institutional framework of budgeting system to find weak its weak points. Simultaneously the similar analysis is compiled for institutional framework and budgeting procedures of EU budget in an effort to find such components, which would be usefull for solving recognized problems in Czech parallel. Theoretical background is provided throught description of particular budgeting systems based on so called Interinstitutional Treatment in case of EU Budget and by Budgeting rule law in case of Czech institutional framework. For better comprehension of Bugeting system in Czech Republic there is incorporated part dealing its evolution since 1918 with accent to recent decades, when the distinctive changes in budgeting rule law were ammended.

Islet transplantation represents an alternative approach in the treatment of patients with type-1 diabetes mellitus. It makes possible to give insulin-producing tissue to patients, which could not underwent surgery operative due to ill health. The second benefit of tissue transplantation is utilization of pancreases unsuitable for organ transplantation in term of surgical criteria. The dissertation includes two studies focused on study of some problems of islet transplantation. In the first study was tested a new method for induction of microchimerism by bone marrow transplantation and its effect on islet allograft survival. We reported, that short-term tacrolimus therapy significantly prolonged insulin independence in fully allogeneic rat model even if the immunosupresants were discontinued. Conditioning of recipients with bone marrow cells transplantation had not additional effect. In the second study there is presented an approach for imaging of intrahepatally-sited islets. The technique is based on labeling of islets by non-toxic superparamagnetic agent during two day culture. After transplantation islets are imaged as hypointense spots on T2 weighing magnetic resonance sections in both in vitro and in vivo experiments. Outstanding changes in MR images were demonstrated during acute rejection of...

The aim of the study was to create a new animal model of schizophrenia based on a neurodevelopmental hypothesis. The central idea of the project has been that a transient insult at early postnatal age will reveal itself in the form of delayed distinct behavioral changes that can be related to schizophrenia. The experimental design used intracerebroventricular (icv) infusion of N-acetyl-L-aspartyl-L-glutamate (NAAG) to rats at postnatal day 12, followed by combination of histology and quantitative morphology in brain tissue obtained within 24 and 96 hours of the NAAG administration and used to investigate possible neurodegeneration. Finally, a battery of behavioural tests was performed several weeks later (late adolescence/early adulthood). NAAG is the most abundant neuropeptide and interacts with the active site of metabotropic glutamate receptors (mGluR II), however, it is also an agonist at NMDA receptors. Neonatal icv infusion of NAAG resulted in detectable damaged neurones in gyrus dentatus. The damage appeared greater at 24 hours, as compared to 96 hours, after the infusion. The presence of damaged neurons was easily demonstrated by Nissl stain, Fluoro Jade В staining combined with Hoechst 33342 and by TUNEL technique. Neonatal administration of NAAG resulted in the appearance of specific, potentially...

Transdermal electroporation of cyclosporine I.
Pavlík, Bohumil ; Doležal, Pavel (advisor) ; Musilová, Marie (referee)
Rigorosal Thesis 2006, Bohumil Pavlík Abstract Effective topical application of cyclosporine is considered to be therapeutically important in the treatment of psoriasis, however, it is complicated by barrier properties of cornified skin layers. Potentially promising approach to solving this problem is to use electroporation. Thesis provides a basic overview of the current state of knowledge in the field of transdermal electroporation. The experiment focuses on penetration testing of cyclosporine in the two pulse protocols. The difference between the application of two or ten rectangular pulses of 10ms duration at time 0 and 6 hours, at a distance of 30 seconds and the voltage pulse 450 is reflected only in the period up to 2 hours from the start of permeation, long-term effect was demonstrated. When verifying the importance of arrangement of electrodes there was no difference between the type of needle-and needle arrangement with a flat surface electrode observed.

Anti-oxidative stress response in Drosophila melanogaster - The role of adipokinetic hormone and adenosine
ZEMANOVÁ, Milada
In this thesis, the phenomena of the oxidative stress (OS) and anti oxidative stress responses in insects are described in a comprehensive review, and the outcomes of the experimental work are presented. The focus of the work was on defence reactions and their putative control by the adipokinetic hormone (AKH) and adenosine in the Drosophila melanogaster model. For this purpose, we studied the effect of the paraquat (oxidative stressor) treatment on adult flies and larvae carrying mutations in Akh (Akh1) and adenosine receptor (AdoR1) genes, and in both these genes together (Akh1 AdoR1 double mutant). The initial mortality tests revealed the double mutant Akh1 AdoR1 was more sensitive to OS than either of the single mutants. The AKH synthesis under the OS condition seems to be out of the gene expression control, since the increase of an AKH amount in CNS was not linked with the stimulation of Akh gene expression after a paraquat treatment. Further, the gene expression of the antioxidant enzyme glutathione S transferase D1 (GstD1) increased rapidly with OS, though the enzyme activity increased negligibly regardless of both the OS and mutations. Interestingly, the relative expression of GstD1 gene was minimal in the double Akh1 AdoR1 mutant; thus, it was concluded that both AKH and adenosine are employed in the GstD1 gene expression control. Similarly, AKH and adenosine seem to act in tandem in glutathione (GSH) regeneration, since the GSH level was significantly lower in all untreated deficient flies with the maximal effect in the Akh1 AdoR1 double mutant; accordingly, the reduction in the GSH level was enhanced by paraquat treatment. Altogether, the important roles of both AKH and adenosine in the anti oxidative stress response in D. melanogaster were demonstrated.
