The diploma thesis deals with problems of depression in older people. Mainly the work is focused on identifying and analyzing the role of nurses in screening for depression in older people in primary care, acute care, long-term care and home care. This thesis was focused on theoretical direction and was used the method of design and demonstration. In this thesis was set one main goals with five research questions. The main goal was to identify and analyze the role of nurses in screening for depression in the elderly. RQ 1: What is the role of the nurse in screening for depression in the elderly? RQ 2: What is the role of the nurse in the primary care in screening for depression in the elderly? RQ 3: What is the role of the nurse in screening for depression in hospitalized patients in acute care? RQ 4: What is the role of the nurse in screening for depression in seniors in long-term and home care? RQ 5: What rating scales and methods are used in screening for depression in the elderly? The thesis introduce the concept of depression. The following are specified the causes of and the important factors that affect depression in the elderly. It also deals the differences in the clinical symptomatology of depression in old age. It explains possibilities and various barriers in the diagnosis of depression. Another chapter introduces complete geriatric examination, diagnostic classification systems, possible screening methods and scales for detection of depression in the elderly population. It also deals methods of pharmacological and non-pharmacological treatment and its possible complications associated with older age. By reason of increased suicide rate caused by depressive disorder the issue of suicidal behavior in the elderly is introduced. The next chapter deals with the nursing process, which is used by nurses in practice. It consists of the evaluation of the patient's health condition, making nursing diagnosis, creating nursing plan and subsequent implementation and evaluation. The nursing process is also needy for providing quality care. The nursing process in the stage of nursing diagnosis, introduces possible nursing diagnosis for a patient suffering from depression, which are based on the latest classification. Finally is described the role of nurses in screening for depression in the elderly in different health facilities and their contribution to the timely evaluation of depression in the elderly. This chapter introduces the role of nurses, nursing screening and collaboration with a physician. The role of nurses in screening for depression in different medical facilities is based on the first phase of the nursing process of assessment. On the basis of objective and subjective information, the nurse will assess the overall health and mental condition of the patient. Primarily, it was investigated what is the role of the nurse in screening for depression. On the basis of content analysis and synthesis it was necessary to used and processed domestic and foreign literature. A number of relevant sources are the results of various studies and Meta-analyzes, mostly from abroad, but also from the Czech Republic. The thesis can serve as a basis for nurses. STUDY OF POTENTIAL PHARMACOLOGICAL PROTECTION OF CARDIAC CELLS AGAINST OXIDATIVE STRESS AND ANTRACYCLINE ANTICANCER DRUGS
Jansová, Hana ; Šimůnek, Tomáš (advisor) ; Hrdina, Radomír (referee) ; Jun, Daniel (referee)
CHARLES UNIVERSITY IN PRAGUE FACULTY OF PHARMACY IN HRADEC KRÁLOVÉ DEPARTMENT OF BIOCHEMICAL SCIENCES Candidate: Mgr. Hana Jansová Supervisor: Doc. PharmDr. Tomáš Šimůnek, Ph.D. Title of Doctoral Thesis: STUDY OF POTENTIAL PHARMACOLOGICAL PROTECTION OF CARDIAC CELLS AGAINST OXIDATIVE STRESS AND ANTRACYCLINE ANTICANCER DRUGS Development of cardiovascular disorders is associated with various risk factors and oxidative stress plays an important role in many of them. Iron-catalysed production of highly toxic and reactive hydroxyl radicals may contribute to oxidative stress. Chelation of free iron seems to be a promising strategy to prevent the propagation of oxidative stress. However, the use of classic iron chelators in pathological conditions without iron overload is associated with the risk of toxicity due to the iron depletion. Hence, this study deals with cardioprotective properties of iron chelators as well as prochelators derived from them. We focused on prochelators with almost no affinity for iron ions until they are activated under disease-specific oxidative stress conditions. For a long time, it has been assumed that oxidative stress is also the main denominator in an anthracycline-induced cardiotoxicity. However, the previous studies suggested alternative mechanism(s). Therefore in the...

Revealing phosphoproteins playing role in tobacco pollen activated in vitro
Fíla, Jan ; Honys, David (advisor) ; Fischer, Lukáš (referee)
5 Abstract Tobacco mature pollen rehydrates in vivo on a stigma tissue, and develops into the rapidly-growing pollen tube. This rehydration process is accompanied by the de-repression of stored mRNA transcripts, resulting in the synthesis of novel proteins. Furthermore, such metabolic switch is also likely to be regulated on the level of post-translational modifications of the already-present proteins, namely via phosphorylation, since it was shown to play a significant regulatory role in numerous cellular processes. Since only a minor part of proteins is phosphorylated in a cell at a time, the employment of various enrichment techniques is usually of key importance. In this diploma project, metal oxide/hydroxide affinity chromatography (MOAC) with aluminium hydroxide matrix was applied in order to enrich phosphoproteins from the mature pollen and the 30-minute in vitro activated pollen crude protein extracts. The enriched fraction was separated by both 2D-GE and gel-free liquid chromatography (LC) approaches with subsequent mass spectrometric analyses. Collectively, 139 phosphoprotein candidates were identified. Additionally, to broaden the number of phosphorylation sites identified, titanium dioxide phosphopeptide enrichment of trypsin-digested mature pollen crude extract was performed. Thanks to the...

Patients with Ulcerous Stomach Disease and Duodena and their Observance of the Regime Precautions
HESOUNOVÁ, Ilona
The topic of the Bachelor Thesis is "Patients with Ulcerous Stomach Disease and Duodena and their Observance of the Regime Precautions". In the Czech Republic the ulcerous stomach disease occurs with 3 to 10% of the population and it afflicts patients particularly in their active age. The theoretical part describes the disease history, its classification, aetiopathogenesis, clinical picture, diagnostics, compilation, treatment, regime precautions, patient´s education and the role of the nurse in the nursing process. The practical part deals with three aims which were set at the beginning of the research. The first aim was to find out if the patients with the ulcerous stomach disease and duodena observe the regime precautions. The second aim was focused on the role of the nurse in the education of patients suffering from the ulcerous stomach disease and duodena. The last aim supposed to use the research results and elaborate information material concerning the regime precautions for patients suffering from the ulcerous stomach disease and duodena. Four hypotheses were made. The first hypothesis presumed that patients suffering from the ulcerous stomach disease and duodena know the regime precautions. The second hypothesis claimed that patients suffering from the ulcerous stomach disease and duodena follow the regime precautions. The third hypothesis said that patients suffering from the ulcerous stomach disease and duodena are aware of the importance of following the regime precautions. The fourth hypothesis supposed that nurses play an important role in the educational process of patients suffering from the ulcerous stomach disease and duodena. There was used the quantitative method of the research inquiry. The chosen technique for data collecting was the anonymous questionnaire. The research set formed patients with the ulcerous stomach disease and duodena from the Internal Department of the Hospital in Písek, from a General Practitioner Surgery in Písek, from the Internal and Surgery Department of the Hospital in Strakonice, from the Gastroenterology Department of the Hospital in České Budějovice and from the Internal Outpatients´ Department of the Clinic in Milevsko. On the basis of the evaluation of the anonymous questionnaire the first, the second and the third hypothesis was not proved. On the other hand the fourth hypothesis was proved. On the basis of the research inquiry there was elaborated information material concerning the regime precautions of the patients suffering from the ulcerous stomach disease and duodena. This material can be an inspiration for the educational activity of nurses too.

Roles of cytoskeleton in mouse polyomavirus trafficking
Klímová, Lucie ; Forstová, Jitka (advisor) ; Šmahel, Michal (referee)
6 Roles of cytoskeleton in mouse polyomavirus trafficking ABSTRACT: Mouse polyomavirus (mPyV) is small non-enveloped DNA virus. Its endocytic pathway is studied for a potential utilisation of polyomaviral virus-like particles in gene therapy and/or immunotherapy. mPyV enter cells by internalisation into smooth monopinocytic vesicles. During it's journey through the cell, it pass through early endosomes, and at the time 3 hours post infection, it is localised in endoplasmic reticulum and recycling endosomes. Many aspects of mPyV trafficking and nuclear entry are not clear yet. Time-lapse live imaging fluorescence confocal microscopy was used to describe the mouse polyomavirus intracellular movements. For these studies, we utilised mPyV fluorophore-labeled virions and cells expressing GFP-tagged g-actin or alpha-tubulin. Some virion-loaded vesicles were seen to move with actin organised into dynamic structures. Some of these structures resembled actin comets created by Listeria or vaccinia virus. At the same time post infection (40-60 min post infection), movement of the virion loaded vesicles along mirotubules was observed suggesting the simultaneous involvement of actin and tubulin during mPyV trafficking. Dynamitin, a dominant negative inhibitor of dynein-dynactin function reduced mPyV infection. Taken...
