Analysis of high-frequency activity in electroencephalogram of animal epilepsy model

High frequency oscillations (HFO) are believed to be a new specific biomarker of epileptogenic tissue. According to the reported findings, we hypothesized that HFOs occur more specific to the epileptogenic tissue. To test this hypothesis, we analyzed intracranial electroencephalograms (iEEG) of three subjects of animal epilepsy model. Overall 180 minutes of iEEG records were processed by the automatic high frequency activity detector and further analyzed. The dominant frequency of each segment was determined and categorized as ripples (80-200 Hz) or fast ripples (200-1000 Hz). Even though overall number of HFO detections predominate in the hemisphere where epileptic focus is located, in comparison of each brain structure separately our hypothesis cannot be fully confirmed.

Enzymatic and inhibiting activity in boar epididymal fluid

Sperm maturation, represents a key step in the reproduction process. Spermatozoa, particularly the plasma membrane, are exposed to epididymal fluid (EF) components representing the natural environment essential for their post-testicular maturation. Changes in the sperm membrane proteins are influenced by proteolytic and glycosidic enzymes present in the EF. Accordingly, the occurrence of inhibitors in this reproductive organ is very important for the regulation of sperm membrane protein processing. In present study, we monitored protease and glycosidase activities, and inhibitors of metallo- and serine proteinases in boar EF. Additionally, we studied acrosin inhibitor in fluid, spermatozoa and tissue along the epididymis. We chromatographically separated boar EF into several fractions. These fractions were subjected to SDS-electrophoresis and the separated proteins were either studied by zymographic methods or transferred to nitrocellulose membranes for detection of metallo- and serine proteinases and their inhibitors, and acrosin inhibitor by specific antibody, respectively. Acrosin inhibitor was monitored also in the sperm and tissue of the boar epididymis. In boar epididymal fluid, several metallo- and serine proteinases with different molecular masses, and inhibitors of metalloproteinase MMP-9 and acrosin were found. We measured strong activity of mannosidase in this fluid. Using specific antibody, we registered the increasing signal of acrosin inhibitor from caput to cauda epididymis in the spermatozoa, fluid and also tissue. Proteinases and their inhibitors in reproductive fluids may play a significant role in reproduction processes. Especially, acrosin inhibitor in the reproductive tract inactivates prematurely released sperm acrosin and protects spermatozoa and reproductive epithelium against proteolytic degradation. High mannosidase activity in boar EF suggests evident role of mannose structures in the sperm interaction during reproductive events.

This bachelor's thesis focuses on the process of evaluation and selection of proper ERP system for company, and summarizes the steps leading to its successful implementa-tion. The thesis tries to cover all the steps from selecting software distributor and sign-ing a contract, to pilot operation and evaluation of implementation results, using specif-ic metrics. In the end, I choose three best known local ERP systems and briefly refer about them.

Study of Electrophysiological Function of the Heart in Experimental Cardiology

Srdeční poruchy, jejichž příkladem je ischemie myokardu, infarkt, hypertrofie levé komory a myokarditida, jsou v experimentální kardiologii obvykle studovány na modelu izolovaného srdce. Kritéria pro detekci srdečních poruch však nejsou pro zvířecí modely standardizována, což komplikuje srovnání a interpretaci výsledků různých experimentálních studií. Obzvlášť složitá situace nastává při současném výskytu několika patologických jevů, jejichž vzájemná součinnost komplikuje rozpoznání jejich individuálních účinků. Korektní posouzení stavu srdce vyžaduje také zohlednění mnoha faktorů spojených s akvizicí dat. Tato práce je věnována kvantitativnímu hodnocení elektrofyziologických změn způsobených globální ischemií myokardu. Vliv ischemie byl hodnocen pro fyziologická srdce a srdce se zvětšenou levou komorou a dále pro srdce nabarvená napěťově-citlivým barvivem di-4-ANEPPS. Přestože jsou oba fenomény často zastoupeny v animálních studiích, nebyl dosud popsán jejich vliv na manifestaci ischemie v elektrogramech (EG), ani nebyl kvantifikován jejich vliv na přesnost detekčních algoritmů pro identifikaci ischemie. Práce shrnuje kvantitativní změny srdeční funkce vyvolané ischemií (v normálních podmínkách, při hypertrofii levé komory, a při administraci barviva) založené na hodnocení EG a VKG parametrů. Dále práce obsahuje rozbor důležitých aspektů akvizice záznamů, jako je umístění snímacích elektrod, způsob výpočtu deskriptorů z EG a VKG (s použitím výsledků manuálního rozměření záznamů, nebo bez něj) a identifikace okamžiku vývoje ischemie v preparátu. Nedílnou součást práce tvoří návrh, realizace a ověření metod pro automatickou detekci ischemie v experimentálních záznamech. Výsledky práce dokazují, že dosažení opakovatelných a věrohodných výsledků je podmíněno zohledněním všech výše uvedených faktorů souvisejících jak se stavem srdce, tak s metodikou záznamu a analýzy dat.

Arsenic speciation analysis by hydride generation – cryotrapping – atomic fluorescence spectrometry with flame-in-gas-shield atomizer

The main target of this work was further improvement of detection limits in the method of arsenic speciation analysis of tri- and pentavalent human metabolites in complex biological matrices. The method combines selective hydride generation with preconcentration and subsequent separation of substituted arsines in a cryogenic trap.

NP search problems and reductions among them Renáta Ševčíková In the thesis we study the class of Total NP search problems. More attention is devoted to study the subclasses of Total NP search problems and reductions among them. We combine some known methods: the search trees and their relation to re- ductions, the Nullstellensatz refutation and the degree lower bound based on design to show that two classes of relativized NP search problems based on Mod-p counting principle and Mod-q counting principle, where p and q are different primes, are not reducible to each other. This thesis is finished by a new separation result for p = 2 and q = 3.

System for Detection of APT Attacks

The thesis investigates APT attacks, which are professional targeted attacks that are characterised by long-term duration and use of advanced techniques. The thesis summarises current knowledge about APT attacks and suggests seven symptoms that can be used to check, whether an organization is under an APT attack. Thesis suggests a system for detection of APT attacks based on interaction of those symptoms. This system is elaborated further for detection of attacks in computer networks, where it uses user behaviour modelling for anomaly detection. The detector uses k-nearest neighbors (k-NN) method. The APT attack recognition ability in network environment is verified by implementing and testing this detector.

The Bachelor Thesis deals with the importance of communication for a manager's work. The thesis is divided into two parts. The first part introduces a manager surroundings, describes abilities, skills and characteristics of manager and focuses on communication of manager. Furthermore introduces the term coaching and staff growth. Based on detections of the research is elaborated a second part, which includes its own research. The practical part contains research in the form of structured interviews with managers at different levels of management. Subsequently, the detected information from managers is evaluated and compared.
