Reactions of Five- and Six-Year-Old Children to Work while Playing Games in a Nursery School

The work is focused on observing of child behaving when she or he has to choose from a lew possibilities offer and when the child is not given a notice about more options, when it depends on the child himself if it would take notice and utilize other variations. Based on four activities (which were compiled in the way that they could refer about the fact if the child would even take in account other suggestions) the response of twelve children was observed. Storytelling as a tool in prevention of socio-pathologic phenomena

This Bachelor thesis focuses on the possibility of using storytelling as a tool in prevention of socio-pathologic phenomena. In the theoretical part are illustrated symptoms of socio-pathologic phenomena in the pre-school education. Futher it puts forward the theory of effective work with a story, aiming to bring the child an instruction and enjoyment. To sum up, the positive impacts of storytelling on the development of children skills are mentioned. In the second practical part of the thesis is elaborated a story itself with focus on the prevention of some socio-pathologic phenomena. The efficacity of the storytelling is verified by the observational method, which elicits the behaviour of individuals as well as of a group and through its results confirms or disproves of the defined hypotheses.

Characterization and identification of wheat allergens from foodstuffs containing wheat

In some cases, ordinarily accepted food can evoke harmful clinical symptoms which are brought on by sensitivity on specific component of foodstuff. This harmful reaction on accepted food is called food allergy and it tends to go up in the world's population in last years. The food allergy affects especially children as well as adults. The important role in the food allergy plays wheat and foodstuff containing components of wheat. Wheat allergens from flour is already well-known, but man accepts wheat in food in the treated form (heat treated food) what can decrease or increase allergenicity. Moreover, enzymes presented in gastrointestinal tract of man can influence allergenicity of wheat allergens too. The goal of this thesis is characterization of allergens presented in heat treated wheat foodstuff and in wheat foodstuff digested by enzymes in gastrointestinal tract of man. Characterization of these altered and newly produced allergens can help find better diagnostic techniques, increase specificity of detection of IgE antibodies, enable pointed therapy of allergic patients or find and apply new and suitable diet.

Photon strength functions in 196Pt from two-step gamma cascade measurement

Photon strength functions (PSFs), using the statistical approach to a nuclear decay, seem to be a suitable tool for the description of the electromagnetic transitions in a nucleus. Since the early 50's, when PSFs were first introduced, a plenty of theoretical models were proposed however their validity is still a question. In this work the data from the measurement of so called Two step cascades in the nucleus 196Pt using the reaction 195Pt(n,gamma)196Pt are processed. The experiment was performed at the Nuclear Physics Institute ASCR in Řež near Prague. We assume that these experimental spectra contain important information on PSFs. Comparison of the processed data with a few Monte Carlo simulations is also a part of this thesis.

Alfons Breska, poet and translator

The work describes the personality and the work of the poet and translator A. Breska. It is the first monographic work about this author and it is based mainly on archive documents from Breska's personal collection in the literary archive Památník národního písemnictví in Prague. Further sources were Breska's manuscripts and typescripts, which are deposited in the Theatre Institute, Theatre Department of the National Museum in Prague and in the Archive of the National Theatre. Following these documents, the work recreates the biography of Alfons Breska and describes his own literary production as well as the translations in the literary context of the period. In a chronological way the work maps the publication of Breska's poetry books and translations, the publications in periodical journals and the reactions of the literary critics. Special attention is paid to the anthologies of German and Japanese poetry. The final part briefly outlines Breska's method of translation. An important part of the diploma thesis is so far the most complete bibliography of Breska's own and translated works, which includes his literary editions, journal entries, unpublished translations for theatres and responses to his work. Flow-chambers for microscopy of living cells

The perfusion chamber for long term observing of live cells by the means the Coherence-Controlled Holographic Microscope (CCHM) was designed. CCHM was built and designed at the Laboratory of the optical microscopy at the Institute of Physical Engineering, Brno University of Technology. CCHM can quantitatively evaluate dynamical changes inside live cells thanks to the quantitative information about phase shift in each pixel of the image. In order to demonstrate advantages of CCHM experimentally, it is important to keep the live cells in the good conditions. This is made by adding the fresh cultivation medium for studied cells directly in the microscope. In contrast to the stationary chamber the perfusion chamber allows both the cultivation medium exchange and the application of biological reagents without the necessity of removing the chamber from the microscope. Therefore we can study the vital signs of cells before and after the application of reagents. An original perfusion system with accessories compatible with CCHM was designed. The design is based on the previously published perfusion system solutions that are referred to in this thesis. The flow characteristics and medium exchange process was discussed and a modification of the internal geometry, based on numerical simulations, was introduced. The applicability of this perfusion chamber has been proven for the CCHM and even for different types of microscopes. The reactions of tumor and epithelial cells during the change of the environment from the cultivation medium to the physiologically solution were studied.

Optimization of separation and quantification of prospective products of big bang

The thesis is a contribution to research aimed at modeling the conditions of the Big Bang and monitoring possible reaction products. As the starting compounds were selected 2- amino-1-butanol, 3-methoxy-1-propanol, 2-methyl-1-butanol, formamide, some of which were subsequently bombarded by PALS at J. Heyrovsky Institute of Physics, Academy of Sciences. Gas chromatography hyphenated with mass spectrometry and liquid chromatography with UV detection were used for analysis. In GC-MS, calibration of 3- methoxy-1-propanol and 2-methyl-1-butanol was carried out and LOD and LOQ were calculated. Comparison of 3-methoxy-1-propanol and bombarded 3-methoxy-1-propanol did not show the emergence of new products since 3-methoxy-1-propanol was in excess. The found compounds can be more likely identified as impurities present in a solvent or in the individual standards. The optimized HPLC system has managed to separate the three standards. Formamide, which is unsuitable for analysis by gas chromatography, due to its high polarity, however, eluted in all tested systems with system peak (the dead time marker). Nevertheless, the bombarded sample was analyzed for comparison with the original standard. Comparison, however, showed no reaction products in the bombarded sample.
