The application off molecular biological methods in diagnosing the cythomegalovirus in the clinical matherials

Human cytomegalovirus (HCMV or CMV) is a human viral pathogen from a group of herpetic viruses. Isolation of DNA can be done manually or with the automatic machines - isolators. In my work I concentrated to compare these two different isolations. I did manual isolation with the help of impervious QIAmp DNA Mini Kitu made by QIAGEN and automatic isolation with isolator MagCore using MagCore Genomic whole Blood Kit (RBCBioscience). To compare the isolations blood and plasma were used as clinical materials and their origin was the same. Both methods were used afterwards to set up the qualitative and comparative CMV DNA. The result means that both isolations are comparable - both can be used with the same quantitative result. In this work there is also compared the qualitative determination DNA CMV by using two PCR methods. One of them was the "In house" method in which are two sections of viral DNA CMV ampificated by the couples of specific primers and later there are detected by the agarose gel. Visualisation of these products on agarose gel is going on with the help of intercalatial reagent - ethidium bromide. This method is very sensible but it takes a lot of time.

The concept of a brand identity in the Czech Republic

Strong brand is a significant element of the differentiation in the highly competitive environment. The first step in the systematic brand building is to define its identity, which ensures the consistency of a brand over time. The goal of the master's thesis is to find out how czech brand consultants define the brand identity. In the master's thesis there is a comparison of the most frequently used elements in the theoretical models and brand bibles of foreign companies with approaches of czech consultants. Theoretical models and brand bibles have been analyzed by using conceptual content analysis. Qualitative research was used to find out approaches to the brand identity of czech consultants. Results of the qualitative research and the synthesis of the theoretical and practical part revealed that czech consultants get inspired from abroad, where some of them use models and some of them approach to defining brand identity instinctively. The master's thesis brings the primary insight into this field within the Czech Republic and can be used as a base for mapping the whole situation in the Czech Republic.

The usage of selected logistic methods in the manufacturing concern

The main aim of this work was to rate the usage of selected logistic methods and introduction of the operating system Kanban to the production line in the project {\clqq}C214 Top Roll`` of the company Faurecia Automotive Czech Republic, s.r.o, Interior Systems. A minor aim then was to find out the knowledge level of the Kanban method. Methodics Hypotheses: 1.The logistic flows used in the Faurecia company already function sufficiently enough, the proof of which is the stabilised production. 2.Using the operating system Kanban for functioning production, logistics and the whole company Faurecia is rather advantageous. 3.Training all the employees is a vital part in the process of introducing the operating system Kamban. Methods: deducion, situation analysis - collection of information,interviews, survey, questioning brief, modeling Structure of the practical part is following: company profile, character of pruduction and setting of the main logistical flows, determination of zones and stock sectors, process implementation and using of Kanban in the production line "C214 Top Roll". The first hypothesis can be confirmed by the situational analysis which evaluates the processing and logistic method. The individual methods {\clqq}met`` or {\clqq}nearly met`` the demands and because all the methods together partake of the production and the production has provided very good results in the previous months, we can consider the production being stabilised. The second can also be affirmed. If the production on a particular assembly line is stabilised, it is possible to introduce the Kanban method. Why not to use it if a there is a great advantage of this method in simplicity and lower production supplies which result in decreasing the total costs? It is beneficial for all participating on the production and for the Faurecia company as a whole. Third hypothesis can be confirmed as well. There has been a survey based on short questionaires and the knowledge of working and using the Kanban has been found inadequate. The best form of providing all the important information will be a training as a part of the process of introducing and using the Kanban method. Both aims have been resulted in stating possible recommendations and improvement suggestions: - production is stabilized, Kanban is possible to apply on 1.9. 2008 - training is necessary, meeting of responsible persons, control, messing, measures, - training of current and new workers, - motivation, communication improve, - system of regural records about information and control of chief (material and production planning)

Remote services control systems

In the present work we study possibilities of network monitoring system creation. We will audit computers in networks and services running on them. System must allow adding new kinds of tests into it. So if some test is not yet implemented it should be possible to add it into the system. As a result there is a implementation of such a system (modular with coordinator) written in C languague for FreeBSD operating system. Concrete testing methods are implemented as standalone modules which comunicate with coordinator by network protocol. In the case of error location the coordinator will send information by given way to set persons. Concrete methods for sending information are also implemented as modules.

Placement on labour market: graduates of humanities from ZČU

Thesis is focused on job placement of graduates of study program Humanistika, Faculty of Arts, University of West Bohemia in Plzeň. In the theoretic part transition from college to work is analyzed, such as methods of job search, efficiency of social networks during job search and importance of human capital. In the situation, when in almost all the developed countries the number of tertiary graduates is markedly increasing, and the problem of overeducation arouses, there is a list of methods. Methods how forecast sectoral prospects of qualifications and occupations on Labor market. It monitors Czech and international surveys of higher education, which are based on graduates' evaluation. In empirical part there is an introduction into qualitative methodology of the specific sample. Then, there is an analysis and interpretation of collected data. It includes evaluation of study program, process and methods of job search, possible unemployment and identifying work values.

Corporate social responsibility and its influence on society

Bachelor thesis "Corporate social responsibility and its influence on society" deals with a form of responsibility in corporate behavior and its effect on surrounding world. A lot of new threats have come up these days because of still rising economic growth. That's why the traditional form of responsibility has to be changed. Corporations have started to implement concept of corporate social responsibility to their business strategies focused on wide spectrum of stakeholders. Thesis is based on qualitative research and presents corporate social responsibility itself and its role and position in society from different views of four groups of stakeholders. I chose these groups: corporations with strategic social responsibility, employees from corporations with and without the concept, nonprofit organizations and customers. Thesis deals with experience of the groups with corporate social responsibility, its influence on them and whether the concept is seen more like marketing strategy and PR or nonprofit helping and being responsible. Survey finally discovered educative influence of corporate social responsibility on society and that the groups are developed by the concept. Corporate social responsibility includes nonprofit helping and also marketing. In this case marketing could be a helpful support...

The idea of expectant parents about the screening of congenital defects in 1.trimester

The theoretical part of this bachelor thesis covers screening examination in pregnancy, screening of congenital developmental disorders in the first and second trimester and invasive methods for pre-natal diagnosis. Screening of congenial developmental disorders in the first trimester consists of use of biochemical markers and an ultrasound examination. Such a screening is called a combined test. The combined test is performed between the 11th and the 14th week of pregnancy. The screening in the first trimester is used to identify the risk of trisomy of chromosome 21 and trisomy of chromosome 18. The first goal has been to find out what the parents? expectations from the screening of congenital developmental disorders of a child in the first trimester are. The other goal of the thesis has been to find out whether parents are afraid of the results of the screening of congenital developmental disorders of a child in the first trimester. The research was conducted using a qualitative research by means of non-standardized interviews. The research set consisted of four couples being at the beginning of the first trimester and that did not have the screening of congenital developmental disorders in the first trimester yet. All responders have expected identification of a risk of congenital developmental disorders in the first trimester from the screening of congenital developmental disorders in the first trimester. All responders, both men and women, have found this screening very important; especially to make them sure the child is in good condition. The second research question has suggested that responders are afraid of the screening of congenital developmental disorders in the first trimester. One responder are not afraid of the results of the screening in the first trimester. This bachelor thesis can be used as a study material for students of a midwifery study programme and midwives at hospitals.

Comparison of pick-up balers Pottinger, Rollprofi 3200 LSC and Krone KR 10-16S during the harvested production.

This bachelor work is focused on evaluation of round balers. It is about comparsion of machines from major producers of harvest equipment Krone and Pöttinger. The goal was to find out qualitative and quantitative differences between round baler Pöttinger Rollprofi 3200 LSC with fixed compression chamber and Krone KR 10-16s with variable compression chamber. It was evaluated performance, forage quality, cutting quality, the measure of copressibility and economic indicators. The result was a finding, that in terms of quality of work, both machines are comparable. The differences are in purchase costs of machines, as a result of higher fixed costs of pick-up baler Krone KR 10-16s. Next differences appears, when different diameters of bales are needed. Then a pick-up baler with variable compression chamber must be used.

A Qualitative Study of Strategy Creation in a Sports Organization

This thesis discusses strategic management in sport. There are two main parts in the thesis. First part includes theory about strategic management, strategic management in sport and strategic planning process. In the second part there is a research which analyzes Czech hanball club HC Dukla Prague. The research is qualitive and done by personal interviews with managers, players and spectators of the club. The main purpose of the thesis is to find out how the club works with the strategy and what are the differences between theory of strategy and real actions of the club. In the end there are some recommendations for the club how to improve their strategic planning process.

Content optimization of ICT knowledge tests in terms of current requirements for students

This bachelor's thesis is focused on optimizing the content of information literacy tests regard of requirements on university students. The thesis includes analysis of internation requirements ECDL on basic level and qualitative investigatoin by interview with teacher at university. The collected data servers as a base for creating new or modifying existing tasks for student testings. Tasks are not assembled into fixed versions of test, but they are flexible for assembling into random versions of tests. Thesis also include testing two variants of test on a selected group of students, but not with IT specialization.
